The present study describes for the first time thermoluminescence (TL) characterization of turtle shell. A fossil shell was collected from the Dongargaon area in the Chandrapur district of Maharashtra, India. TL was recorded and a comparative study of TL for the above material was performed to understand the special TL characteristics of the shell. The shell was irradiated with 60Co γ‐radiation to study its TL properties. The sample displayed two good TL peaks at 135°C and 255°C. A linear dose–response curve for the irradiated sample was produced for the dose range 0.79–28.5 kGy; this sample of turtle shell (fossil) may be useful as a high dose dosimetry phosphor in this range. This geological sample was further characterized using X‐ray diffraction to confirm its phase structure and by scanning electron microscopy , Fourier transform infrared and wavelength dispersive X‐ray fluorescence spectroscopy to determine morphology, vibration, and elemental composition as ppm or percentage of the sample, respectively. Fungicide application is an effective management option to control late blight of potato (caused by Phytophthora infestans). Field experiments were conducted to evaluate the efficacy of recently introduced and previously used fungicides on late blight management and potato yields in the western plains of Nepal in 2015 and 2016 crop seasons. Fungicides and a non-treated control (NTC) were replicated three times in a randomized block design planted with late blight susceptible cultivar Cardinal. Chlorothalonil, copper oxychloride, dimethomorph, fenamidone + mancozeb, mancozeb and metalaxyl were sprayed in 2015. In 2016, carbendazim was used instead of chlorothalonil. The area under disease progress curve (AUDPC) was consistently reduced in years by dimethomorph (90% and 65% in 2015 and 2016, respectively), fenamidone + mancozeb (68% and 62%) and mancozeb (40% and 47%) compared with the NTC. Similarly, tuber yield was increased with the application of dimethomorph (266% and 146% in 2015 and 2016, respectively), fenamidone + mancozeb (211% and 155%) and mancozeb (136% and 116%) compared with the NTC. Chlorothalonil reduced AUDPC by 43% and increased tuber yield by 170% in 2015. Other fungicides either had inconsistent results or did not reduce late blight severity and consequent effects on potato yield. The overall benefit–cost ratio was highest for dimethomorph in both years. These results show efficacy of dimethomorph, fenamidone + mancozeb and mancozeb in reducing late blight severity and increasing potato tuber yield in the plains of Nepal. 相似文献

Voltage-dependent anion channels (VDACs) are pore forming proteins predominantly found in the outer mitochondrial membrane and are thought to transport Ca(2+). In this study, we have investigated the possible role of type 2 VDAC (VDAC2) in cardiac Ca(2+) signaling and Ca(2+) sparks using a lentiviral knock-down (KD) technique and two-dimensional confocal Ca(2+) imaging in immortalized autorhythmic adult atrial cells, HL-1. We confirmed high expression of VDAC2 protein in ventricular, atrial, and HL-1 cells using Western blot analysis. Infection of HL-1 cells with VDAC2-targeting lentivirus reduced the level of VDAC2 protein to ~10%. Comparisons of autorhythmic Ca(2+) transients between wild-type (WT) and VDAC2 KD cells showed no significant change in the magnitude, decay, and beating rate of the Ca(2+) transients. Caffeine (10mM)-induced Ca(2+) release, which indicates sarcoplasmic reticulum (SR) Ca(2+) content, was not altered by VDAC2 KD. Interestingly, however, the intensity, width, and duration of the individual Ca(2+) sparks were significantly increased by VDAC2 KD in resting conditions, with no change in the frequency of sparks. VDAC2 KD significantly delayed mitochondrial Ca(2+) uptake during artificial Ca(2+) pulses in permeabilized HL-1 cells. These results suggest that VDAC2 may facilitate mitochondrial Ca(2+) uptake and restrict Ca(2+) spark expansion without regulating activations of sparks under resting conditions, thereby providing evidence on the functional role of VDAC2 in cardiac local Ca(2+) signaling. 相似文献

BackgroundWorldwide, it is estimated that snakes bite 4.5–5.4 million people annually, 2.7 million of which are envenomed, and 81,000–138,000 die. The World Health Organization reported these estimates and recognized the scarcity of large-scale, community-based, epidemiological data. In this context, we developed the “Snake-Byte” project that aims at (i) quantifying and mapping the impact of snakebite on human and animal health, and on livelihoods, (ii) developing predictive models for medical, ecological and economic indicators, and (iii) analyzing geographic accessibility to healthcare. This paper exclusively describes the methodology we developed to collect large-scale primary data on snakebite in humans and animals in two hyper-endemic countries, Cameroon and Nepal.Methodology/Principal findingsWe compared available methods on snakebite epidemiology and on multi-cluster survey development. Then, in line with those findings, we developed an original study methodology based on a multi-cluster random survey, enhanced by geospatial, One Health, and health economics components. Using a minimum hypothesized snakebite national incidence of 100/100,000/year and optimizing design effect, confidence level, and non-response margin, we calculated a sample of 61,000 people per country. This represented 11,700 households in Cameroon and 13,800 in Nepal. The random selection with probability proportional to size generated 250 clusters from all Cameroonian regions and all Nepalese Terai districts. Our household selection methodology combined spatial randomization and selection via high-resolution satellite images. After ethical approval in Switerland (CCER), Nepal (BPKIHS), and Cameroon (CNERSH), and informed written consent, our e-questionnaires included geolocated baseline demographic and socio-economic characteristics, snakebite clinical features and outcomes, healthcare expenditure, animal ownership, animal outcomes, snake identification, and service accessibility.Conclusions/SignificanceThis novel transdisciplinary survey methodology was subsequently used to collect countrywide snakebite envenoming data in Nepal and Cameroon. District-level incidence data should help health authorities to channel antivenom and healthcare allocation. We analyse the affective-cultural assemblage of “Indian rape culture” across historical time and social space. We examine news coverage of a highly visible 2012 rape in Delhi in two newspapers, New York Times and Times of India, and the longevity of these articulations through an analysis of online discourses three years later. We further trace colonial-era materializations of Indian rape culture which emerged in the context of the “Indian Mutiny” in contrast to local perspectives, and which were rearticulated by development and human rights organizations. We show that at each moment, Global north voices and institutions dominate. The processes of articulation emanating from Global north institutions serve to realize a racialized transnational assemblage of Indian rape culture. Our framework points to the “soft” power and fluidity of racializing processes, the heterogeneity, and the multiple logics that appear disconnected, but which nonetheless flow and come together to sustain racialized power structures. 相似文献

The kidney is an important organ for arterial blood pressure (BP) maintenance. Reduced NO generation in the kidney is associated with hypertension in insulin resistance. NO is a critical regulator of vascular tone; however, whether insulin regulates NO production in the renal inner medullary collecting duct (IMCD), the segment with the greatest enzymatic activity for NO production in kidney, is not clear. Using an NO-sensitive 4-amino-5-methylamino-2′,7′-difluorofluorescein (DAF-FM) fluorescent dye, we found that insulin increased NO production in mouse IMCD cells (mIMCD) in a time- and dose-dependent manner. A concomitant dose-dependent increase in the NO metabolite (NOx) was also observed in the medium from insulin-stimulated cells. NO production peaked in mIMCD cells at a dose of 100 nm insulin with simultaneously increased NOx levels in the medium. At this dose, insulin significantly increased p-eNOSSer1177 levels in mIMCD cells. Pretreatment of cells with a PI 3-kinase inhibitor or insulin receptor silencing with RNA interference abolished these effects of insulin, whereas insulin-like growth factor-1 receptor (IGF-1R) silencing had no effect. We also showed that chronic insulin infusion to normal C57BL/6J mice resulted in increased endothelial NOS (eNOS) protein levels and NO production in the inner medulla. However, insulin-infused IRKO mice, with targeted deletion of insulin receptor from tubule epithelial cells of the kidney, had ∼50% reduced eNOS protein levels in their inner medulla along with a significant rise in BP relative to WT littermates. We have previously reported increased baseline BP and reduced urine NOx in IRKO mice. Thus, reduced insulin receptor signaling in IMCD could contribute to hypertension in the insulin-resistant state. 相似文献
