Although ecological differences between native and introduced ranges have been considered to drive rapid expansion of invasive species, recent studies suggest that rapid evolutionary responses of invasive species to local environments may also be common. Such expansion across heterogeneous environments by adaptation to local habitats requires genetic variation. In this study, we investigated the source and role of standing variation in successful invasion of heterogeneous abiotic environments in a self-incompatible species, Lotus corniculatus. We compared phenotypic and genetic variation among cultivars, natives, and introduced genotypes, and found substantial genetic variation within both native and introduced populations. Introduced populations possessed genotypes derived from both cultivars and native populations, and had lower population differentiation, indicating multiple sources of introduction and population admixture among the sources in the introduced range. Both cultivars and introduced populations had similarly outperforming phenotypes on average, with increased biomass and earlier flowering compared with native populations, but those phenotypes were within the range of the variation in phenotypes of the native populations. In addition, clinal variation within introduced populations was detected along a climatic gradient. Multiple introductions from different sources, including cultivars, may have contributed to pre-adaptive standing variation in the current introduced populations. We conclude that both introduction of cultivar genotypes and natural selection in local environments contributed to current patterns of genetic and phenotypic variation observed in the introduced populations.  相似文献

The purpose of this study was to establish a method for eradicating a chytrid fungus (Batrachochytrium dendrobatidis; Bd) from the Japanese giant salamander Andrias japonicus. The emerging agent (Bd) has a high rate of detection in this endangered amphibian species, which is designated as a special natural monument in Japan. Four Japanese giant salamanders with Bd confirmed by PCR assay were bathed in 0.01% itraconazole for 5 min d-1 over 10 successive days. PCR assays were conducted prior to treatment, on Days 5 and 10 of treatment, and on Days 7 and 14 post-treatment. By treatment Day 5, all individuals tested negative for Bd and remained negative until the end of the experiment. No side effects associated with itraconazole were observed. The present method appears to be a safe and effective approach for Bd eradication and may contribute to reducing the threat and spread of Bd among endangered amphibians. Notably, this study represents the first reported Bd eradication experiment involving Japanese giant salamanders.  相似文献

Recent increases in international trade have increased the cost to control and eradicate exotic species. Although many species are under quarantine control for agriculture, forestry, and public health, most species invisible to the naked eye are ignored because of the lack of both specialized assessors and risk assessments. We developed a species risk assessment particularly adapted to fungi, nematodes, and mites, that might be unintentionally introduced with exotic forest products and become threats to terrestrial ecosystems. We developed our assessment with reference to existing risk assessments for exotic organisms, including their ecological features such as phoresy and parasitism. We then tested our assessment with well-known organisms and assessed the risks of organisms unintentionally introduced into Japan. The assessment demonstrated scientifically acceptable scores for each organism. We suggest quarantine control of risk pathways as a practical approach for controlling unintentionally introduced organisms that are invisible to the naked eye.  相似文献

Kin selection theory predicts a high coefficient of genetic relatedness among nestmates, explaining the frequent evolution of eusociality in a social hymenopteran colony. The bumble bee is a primitively eusocial hymenoptera whose colonies are founded by a single queen. In such a monogynous colony, mating frequency of the queen is the sole factor that affects genetic relatedness among nestmates. Although the queens of most bumble bee species are known to be monandrous, there are some species that are known to be polyandrous. Here, we estimated the effective paternity in the native colonies of Japanese bumble bees, Bombus ardens, B. diversus, and B. honshuensis, using genetic polymorphic data of microsatellites. We found no evidence for polyandry in any investigated colonies, which suggests that within-colony genetic relatedness is very high for all of these colonies.  相似文献

To develop a risk-assessment system for small organisms accidentally introduced with imported organisms, we investigated as a first case study parasitic canestriniid mites, which have been imported into Japan via pet lucanid beetles from Southeast Asia. We collected mites from pinned specimens of Japanese lucanids collected before 1999—when the Japanese government lifted a ban on the import of the beetles—and living mites from imported and native lucanid beetles collected after that. No foreign canestriniid was found on any of the native Japanese beetles. Because the mites collected from imported beetles were different from Japanese species, we conclude that the foreign mites have not yet established wild populations in Japan. However, because the Japanese mites migrate between hosts without host physical contact, introduced mites are assumed to be able to migrate from a foreign to Japanese host. In fact, possible contamination was observed in pet shops. We observed host switching in only one direction: Southeast Asian Canestrinia nr spectanda switched to Japanese Dorcus rectus, but Japanese Coleopterophagus berlesei never switched to Indonesian D. titanus. The foreign mites reproduced between 15°C and 25°C, suggesting that the mites could survive in mountainous sites in southern Japan and at low elevations in northern Japan. The ability of foreign parasitic canestriniids to infect and survive on Japanese hosts at temperatures characteristic of much of Japan leads us to conclude that these mites present a potential risk to Japanese endemic canestriniids as well as to native Japanese lucanids.  相似文献

We investigated natural populations of three Japanese native bumblebee species to determine the status of infestation by a tracheal mite, Locustacarus buchneri, which we had earlier detected in introduced commercial colonies of the European bumblebee, Bombus terrestris. We also investigated mite infestation in commercial colonies of a Japanese native species, B. ignitus, which are mass-produced in the Netherlands and reimported into Japan. We detected the mite in both natural and commercial colonies of the Japanese species. Comparison of 555 bp sequences of the mitochondrial DNA (mtDNA) cytochrome oxidase 1 (CO1) gene from the mite showed that there were seven haplotypes, on the basis of combinations of substitutions at eight sites in the gene. The haplotypes of the mites in the Japanese native bumblebees and the haplotypes of the mites in B. terrestris did not overlap; however, mtDNA of mites detected in the commercial colonies of B. ignitus possessed the same sequence as a European haplotype. These results indicate that transportation of bumblebee colonies will cause overseas migration of parasitic mites of different origins.  相似文献

Full-sib reconstruction from molecular marker data in the absence of parental information is an active research area. Such analyses can provide useful information for studies of mating systems and gene flow, and for estimating effective population size in the wild. Although various methods have been proposed, but their estimation accuracies for some applications are not known. Here we propose a modified version of Shared Loci Correspondence Analysis (mSLCA) to reconstruct full-sib families of haplodiploid species. We ran simulations to compare the accuracies of the original SLCA and mSLCA. mSLCA outperformed SLCA at various data settings. mSLCA produced accurate estimates of the number of full-sib families when the numbers of loci and alleles per locus were equal to or more than eight. We also used actual DNA data of commercial Bombus terrestris colonies to verify the validity of estimates with an increasing number of colonies. mSLCA outperformed SLCA at various colony sizes with slight underestimation.  相似文献

We used a recently introduced strain of medaka, the see-through medaka, whose internal organs can be seen through the skin, to develop an in situ toxicity assay of ethoxyresorufin-O-deethylase (EROD) activity that detected fluorescence from resorufin, a metabolite of ethoxyresorufin and thus an indicator of CYP1A activity. EROD activity in the liver and gills of 2-week post-hatch see-through medaka exposed simultaneously to various concentrations of 3-methylcholanthrene and 200 microg/L ethoxyresorufin for 24 h was proportional to the 3-methylcholanthrene dose. Activities in the liver and gills peaked at 40 microg/L of 3-methylcholanthrene and then decreased at higher doses, possibly because of 3-methylcholanthrene toxicity. At 1-week post-hatch stage, however, constant high EROD activity was observed in controls and at all 3-methylcholanthrene doses. Four-week post-hatch see-through medaka exhibited less EROD activity than 2-week post-hatch see-through medaka, and activity in the liver peaked at 100 microg/L of 3-methylcholanthrene. Adult see-through medaka were not suitable for fluorescence detection owing to their thick skin, muscle and/or tissue. In tests of oxidative activity response to ethoxyresorufin, 1-day and 1-week post-hatch see-through medaka exhibited high intrinsic EROD activity in the liver, gills, and other organs in the absence of 3-methylcholanthrene. This intrinsic activity declined with growth and explained the high constant EROD activity at 1-week post-hatch stage.  相似文献

For some wildlife commodities, rare species are especially sought after. The tendency for rare commodities to be of higher value can fuel their exploitation and as numbers dwindle, the demand can increase. Consequently, this can precipitate these rare species into an overexploitation vortex where they become increasingly rare, valued and exploited until eventual extinction. We focus here on the hobby of collecting stag beetles, to ascertain if the market value of these items is driven by rarity and if, consequently, these species are vulnerable to this overexploitation vortex. Stag beetle collections fuel a large and lucrative market in Japan, involving more than 700 species from all over the world, with over 15 million specimens imported a year. Some particularly valued species fetch more than US$5,000 a piece. We assessed the importance of species rarity as an acquisition criterion in this market using two methods: an Internet online questionnaire responded to by 509 participants and through examining the quantities imported in Japan and prices paid by collectors. We discovered that species rarity is one of the main choice criteria for acquisition by collectors: rare stag beetles are valued more than the common species and, consequently, stag beetles are vulnerable to the anthropogenic Allee effect in this market. Because of the sheer size of the market and the pervasive nature of this rarity paradox, the attraction to rarity equates to a potential extinction threat for many rare stag beetles species.  相似文献
