We studied the genetic basis of resistance to two new acaricides, chlorfenapyr and etoxazole, which have different chemical structures and modes of action in the two-spotted spider mite, Tetranychus urticae Koch. The resistance ratios calculated from the LC50s of resistant and susceptible strains were 483 for chlorfenapyr and >100,000 for etoxazole. Mortality caused by the two acaricides in F1 progeny from reciprocal crosses between the resistant and susceptible strains indicated that the modes of inheritance of resistance to chlorfenapyr and etoxazole were completely dominant and completely recessive, respectively. Mortality in F2 progeny indicated that for both acaricides, the resistance was under monogenic control. Repeated backcross experiments indicated a linkage relationship among the two acaricide resistances and malate dehydrogenase, although phosphoglucoisomerase was not linked with them. The recombination ratio between the resistances was 14.8%. From this result, we suggest that heavy spraying of the two acaricides will lead to apparent cross-resistance as a consequence of crossing over; the two resistance genes are so close to each other that it would be difficult to segregate them once they came together on the same chromosome.  相似文献

In areas of endemic parasite transmission, protective immunity to Plasmodium falciparum malaria is acquired over several years with numerous disease episodes. Acquisition of Abs to parasite-encoded variant surface Ags (VSA) on the infected erythrocyte membrane is important in the development of immunity, as disease-causing parasites appear to be those not controlled by preexisting VSA-specific Abs. In this work we report that VSA expressed by parasites from young Ghanaian children with P. falciparum malaria were commonly and strongly recognized by plasma Abs from healthy children in the same area, whereas recognition of VSA expressed by parasites from older children was weaker and less frequent. Independent of this, parasites isolated from children with severe malaria (cerebral malaria and severe anemia) were better recognized by VSA-specific plasma Abs than parasites obtained from children with nonsevere disease. This was not due to a higher infection multiplicity in younger patients or in patients with severe disease. Our data suggest that acquisition of VSA-specific Ab responses gradually restricts the VSA repertoire that is compatible with parasite survival in the semi-immune host. This appears to limit the risk of severe disease by discriminating against the expression of VSA likely to cause life-threatening complications, such as cerebral malaria and severe anemia. Such VSA seem to be preferred by parasites infecting a nonimmune host, suggesting that VSA expression and switching are not random, and that the VSA expression pattern is modulated by immunity. This opens the possibility of developing morbidity-reducing vaccines targeting a limited subset of common and particularly virulent VSA.  相似文献

The genetics of resistance to three new types of acaricide tebufenpyrad, fenpyroximate and pyridaben was studied by crossing a resistant (R) with a susceptible (S) strain of Tetranychus kanzawai Kishida. The resistance ratios calculated from the LC50s of the R and S strains were 97, 1265 and 134 for tebufenpyrad, fenpyroximate and pyridaben, respectively. The responses to the three acaricides in F1 females from reciprocal crosses between the R and S strains showed that the modes of inheritance of resistance to tebufenpyrad, fenpyroximate and pyridaben were intermediate, incompletely dominant and completely recessive, respectively. A commercial colony of Bombus terrestris (L.) was introduced to Japan in 1992 for crop pollination in greenhouses. Since then wild colonies have developed and spread in some regions. In the present study, we measured the spatial distribution and temporal change in abundance of B. terrestris in the Chitose River Basin, Hokkaido, Japan to elucidate the relation of greenhouses to the bees distribution and to evaluate its potential effects on native bumblebees. Bumblebees were collected with window traps in windbreak forests roughly 1, 2, 4, and 6 km NNW and SSE of a large greenhouse. The peak catch of B. terrestris queens occurred in early June, suggesting that they had successfully hibernated in the field. The distributions of B. terrestris and the native B. ardens were mutually exclusive, while the native B. hypocrita appeared at all sites. Catches of B. terrestris were restricted to within 4 km of the nearest greenhouse, suggesting that the invasion was still in the initial phase in this area. The reduction in abundance of the native bumblebees in the sites of high B. terrestris abundance suggests the presence of interspecies competition between B. terrestris and the native bumblebees during the early part of the colony activity, although such reduction in B. ardens can be explained by habitat suitability.  相似文献

Background

Cerebral malaria (CM) is the most severe outcome of Plasmodium falciparum infection and a major cause of death in children from 2 to 4 years of age. A hospital based study in Ghana showed that P. falciparum induces eosinophilia and found a significantly higher serum level of eosinophil cationic protein (ECP) in CM patients than in uncomplicated malaria (UM) and severe malaria anemia (SA) patients. Single nucleotide polymorphisms (SNPs) have been described in the ECP encoding-gene (RNASE3) of which the c.371G>C polymorphism (rs2073342) results in an arginine to threonine amino acid substitution p.R124T in the polypeptide and abolishes the cytotoxicity of ECP. The present study aimed to investigate the potential association between polymorphisms in RNASE3 and CM.

Methodology/Principal Findings

The RNASE3 gene and flanking regions were sequenced in 206 Ghanaian children enrolled in a hospital based malaria study. An association study was carried out to assess the significance of five SNPs in CM (n = 45) and SA (n = 56) cases, respectively. The two severe case groups (CM and SA) were compared with the non-severe control group comprising children suffering from UM (n = 105). The 371G allele was significantly associated with CM (p = 0.00945, OR = 2.29, 95% CI = 1.22–4.32) but not with SA. Linkage disequilibrium analysis demonstrated significant linkage between three SNPs and the haplotype combination 371G/*16G/*94A was strongly associated with susceptibility to CM (p  = 0.000913, OR = 4.14, 95% CI = 1.79–9.56), thus, defining a risk haplotype. The RNASE3 371GG genotype was found to be under frequency-dependent selection.

Conclusions/Significance

The 371G allele of RNASE3 is associated with susceptibility to CM and forms part of a risk associated haplotype GGA defined by the markers: rs2073342 (G-allele), rs2233860 (G-allele) and rs8019343 (A-allele) respectively. Collectively, these results suggest a hitherto unrecognized role for eosinophils in CM pathogenesis.  相似文献

Although biological invasion has a devastating impact on biodiversity, it also provides a valuable opportunity for natural experiments on evolutionary responses. Alien populations are often subject to strong natural selection when they are exposed to new abiotic and biotic conditions. Native populations can also undergo strong selection when interacting with introduced enemies and competitors. This special feature aims to highlight how evolutionary studies take advantage of biological invasion and, at the same time, emphasizes how studying evolutionary processes deepens our understanding of biological invasions. We hope this special feature stimulates more invasion studies taking evolutionary processes into account. Those studies should provide fundamental information essential for formulating effective measures in conserving native biodiversity, as well as valuable empirical tests for evolutionary theories.  相似文献

ABSTRACT: BACKGROUND: Severe malarial anaemia (SMA) is a major life-threatening complication of paediatric malaria. Protracted production of pro-inflammatory cytokines promoting erythrophagocytosis and depressing erythropoiesis is thought to play an important role in SMA, which is characterized by a high TNF/IL-10 ratio. Whether this TNF/IL-10 imbalance results from an intrinsic incapacity of SMA patients to produce IL-10 or from an IL-10 unresponsiveness to infection is unknown. Monocytes and T cells are recognized as the main sources of TNF and IL-10 in vivo, but little is known about the activation status of those cells in SMA patients. METHODS: The IL-10 and TNF production capacity and the activation phenotype of monocytes and T cells were compared in samples collected from 332 Ghanaian children with non-overlapping SMA (n = 108), cerebral malaria (CM) (n = 144) or uncomplicated malaria (UM) (n = 80) syndromes. Activation status of monocytes and T cells was ascertained by measuring HLADR + and/or CD69+ surface expression by flow cytometry. The TNF and IL-10 production was assessed in a whole-blood assay after or not stimulation with lipopolysaccharide (LPS) or phytohaemaglutinin (PHA) used as surrogate of unspecific monocyte and T cell stimulant. The number of circulating pigmented monocytes was also determined. RESULTS: Monocytes and T cells from SMA and CM patients showed similar activation profiles with a comparable decreased HLA-DR expression on monocytes and increased frequency of CD69+ and HLA-DR + T cells. In contrast, the acute-phase IL-10 production was markedly decreased in SMA compared to CM (P = .003) and UM (P = .004). Although in SMA the IL- 10 response to LPS-stimulation was larger in amplitude than in CM (P = .0082), the absolute levels of IL-10 reached were lower (P = .013). Both the amplitude and levels of TNF produced in response to LPS-stimulation were larger in SMA than CM (P = .019). In response to PHA-stimulation, absolute levels of IL-10 produced in SMA were lower than in CM (P = .005) contrasting with TNF levels, which were higher (P = .01). CONCLUSIONS: These data reveal that SMA patients have the potential to mount efficient IL-10 responses and that the TNF/IL-10 imbalance may reflect a specific monocyte and T cell programming/polarization pattern in response to infection.  相似文献

Spatial and temporal characteristics of host plants can influence the population biology of the herbivores feeding on them. In this study, I examined the effect of variation in host plant characteristics on the development of acaricide resistance in the two-spotted spider mite Tetranychus urticae, a widely distributed agricultural pest. This investigation examined the geographic variation in the degree of resistance to two new types of acaricide, pyridaben and fenpyroximate. From mortality tests at field-level concentrations of the acaricides, many populations collected from fruit trees and roses had a high frequency of resistant individuals for acaricides while almost all populations collected from herbaceous crops had low frequencies of resistant individuals. These results, combined with those from a previous allozyme study, indicate that patch size and persistence of host plants regulate the population structure of the mites including gene flow between populations and, by extension, the development of acaricide resistance.  相似文献

Summary. Facultative parthenogenesis has great adaptive significance, especially with regard to low pairing efficiency. In the termite Reticulitermes speratus, females that fail to mate with males reproduce parthenogenetically and found colonies cooperatively with partner females or even alone. Comparison of colony foundation success at 400 days between colonies founded by single females (F), female-female pairs (FF), and male-female pairs (FM) showed that female-female cooperation promoted colony survivorship over monogamous foundation. We report here for the first time the mode of parthenogenesis in Isoptera. Combining chromosome observations and genetic analysis using microsatellites, we show that the mode of parthenogenesis is diploid thelytoky and that the restoration of ploidy is most likely accomplished by terminal fusion. Parthenogens show a higher mortality and a longer egg-development time than sexually produced offspring, probably due to reduced heterozygosity. In addition Wolbachia bacteria were detected in R. speratus. However, since Wolbachia was also detected in non-parthenogenetic R. flavipes, it is unlikely that Wolbachia is the cause of parthenogenesis in R. speratus.Received 30 October 2003; revised 4 March 2004; accepted 17 March 2004.  相似文献
