Mitogen-activated protein kinases (MAPKs) play pivotal roles in development and environmental interactions in eukaryotes. Here, we studied the function of a MAPK, NaMPK4, in the wild tobacco species Nicotiana attenuata. The NaMPK4-silenced N. attenuata (irNaMPK4) attained somewhat smaller stature, delayed senescence, and greatly enhanced stomatal conductance and photosynthetic rate, especially during late developmental stages. All these changes were associated with highly increased seed production. Using leaf epidermal peels, we demonstrate that guard cell closure in irNaMPK4 was strongly impaired in response to abscisic acid and hydrogen peroxide, and consistently, irNaMPK4 plants transpired more water and wilted sooner than did wild-type plants when they were deprived of water. We show that NaMPK4 plays an important role in the guard cell-mediated defense against a surface-deposited bacterial pathogen, Pseudomonas syringae pv tomato (Pst) DC3000; in contrast, when bacteria directly entered leaves by pressure infiltration, NaMPK4 was found to be less important in the resistance to apoplast-located Pst DC3000. Moreover, we show that salicylic acid was not involved in the defense against PstDC3000 in wild-type and irNaMPK4 plants once it had entered leaf tissue. Finally, we provide evidence that NaMPK4 functions differently from AtMPK4 and AtMPK11 in Arabidopsis (Arabidopsis thaliana), despite their sequence similarities, suggesting a complex functional divergence of MAPKs in different plant lineages. Eighty-three per cent of 200 different freshly isolated cultures of Staphylococcus epidermidis produced beta lactamase. Growth in the presence of acridine orange or ethidium bromide or growth at 44 C resulted in a high frequency of loss of the beta lactamase genes in some strains of S. epidermidis. The relationship between beta-lactamase production and resistance to mercuric, cadmium and arsenate ions differed from that observed in Staphylococcus aureus. It is postulated that the genes for beta lactamase in certain strains of S. epidermidis are on a plasmid. 相似文献

The wild tobacco species Nicotiana attenuata has been intensively used as a model plant to study its interaction with insect herbivores and pollinators in nature, however very little is known about its native pathogen community. We describe a fungal disease outbreak in a native N. attenuata population comprising 873 plants growing in an area of about 1500 m2. The population was divided into 14 subpopulations and disease symptom development in the subpopulations was monitored for 16 days, revealing a waxing and waning of visible disease symptoms with some diseased plants recovering fully. Native fungal N. attenuata pathogens were isolated from diseased plants, characterized genetically, chemotaxonomically and morphologically, revealing several isolates of the ascomycete genera Fusarium and Alternaria, that differed in the type and strength of the disease symptoms they caused in bioassays on either detached leaves or intact soil-grown plants. These isolates and the bioassays will empower the study of N. attenuata-pathogen interactions in a realistic ecological context. 相似文献

The grassland community of Lac du Bois Provincial Park in the interior of British Columbia has become increasingly invaded by Centaurea stoebe (=Centaurea maculosa; spotted knapweed). Allelopathy, through the production of the phytotoxin (±)-catechin by C. stoebe roots is believed to be partly responsible for knapweed’s invasive success. We used field sampling and greenhouse experiments to examine two questions: (1) Is increasing abundance of C. stoebe negatively associated with the abundance of specific native grassland species? (2) Do species that exhibit a negative correlation with C. stoebe abundance in the field demonstrate increased levels of susceptibility to application of (±)-catechin during germination? Thirty-eight plots were sampled in the grassland, encompassing areas of low—high knapweed abundance. Seeds from eight native species, exhibiting positive, neutral or negative correlation with knapweed abundance, were treated with three concentrations (0, 0.5, 2.0 mg/mL) of (±)-catechin. Root growth and percent germination were measured over a 6-week period. The results indicate that C. stoebe abundance is negatively correlated with native plant species abundance and may alter plant community composition. Moreover root radical growth was significantly negatively affected by treatment with (±)-catechin in all four native plant species that exhibited a negative correlation with knapweed abundance in the field. Past studies have failed to conclusively link greenhouse results with plant community patterns. Manipulations of the interactions between plants and their floral visitors remain the most successful path to an understanding of floral traits, which may have been shaped by both herbivores and pollinators. By using genetic tools in combination with old-fashioned field work the dual protective/advertisement functions of floral traits are being realized. The distinction between wanted and unwanted floral visitors is blurring, and plants with specialized pollination systems are being found capable of using alternative pollinators if the specialized pollinators fail to perform. 相似文献
