Abstract: Arylsulphatase C and estrone sulphatase activities of sheep hypothalamus-preoptic area-midbrain were examined for their susceptibility to phospholipase action. Russel's viper phospholipase A could completely inactivate estrone sulphatase without affecting arylsulphatase C. The latter was partially inactivated by S. aureus phospholipase C but not by C. welchi phospholipase C. Both arylsulphatase C and estrone sulphatase were inactivated to different extents by sodium deoxycholate, which is known to activate the intrinsic phospholipases of brain. Hydrophobic interaction chromatography on phenyl-Sepharose resulted in the differential elution of arylsulphatase C and estrone sulphatase. The results suggest that one enzyme is not responsible for arylsulphatase C and estrone sulphatase activities.  相似文献

The pathogenic effect of cotton blight disease is influenced by Xanthomonas malvacearum toxin. The toxin has been highly purified and the interaction between the toxin and the energy-generating system of mitochondria has been characterized. The significance of integrated nutrient management practices is well established in improving the productivity of chickpea (Cicer arietinum); however, the effects of the inoculation of cyanobacterial inoculants on nodule metabolism, microbiome and associated genes are less explored. In the present investigation, cyanobacterium Anabaena laxa (A. laxa) and biofilm developed using Anabaena torulosa, with Mesorhizobium ciceri as a partner (An-M. ciceri), were evaluated along with Mesorhizobium ciceri (M. ciceri) alone, in three chickpea cultivars. Microbial inoculation led to 40–70% enhancement in nitrogen fixation, leghaemoglobin and ureide content, and two- to threefold increment in nitrate reductase and phosphoenolpyruvate carboxylase activity of the nodules. An enhancement of 30–50% in the soil available macro- and micronutrients and plant growth attributes was also observed. A significant correlation between the soil microbiological and plant parameters was recorded, particularly in relation to the nitrogen dynamics. Increases in the leghaemoglobin content in nodules due to An-M. ciceri, A. laxa and M. ciceri ranged from 18 to 40%, particularly in chickpea cv. BG372 in which 60–80% enhancement was recorded. Whereas the nifH gene copies in the nodule tissues ranged from 5.00 × 106 to 3.35 × 107 g−1, the application of A. laxa led to higher abundances of nifH gene copies in desi chickpea cv. BG372 and kabuli BG1053 cultivars. An-M. ciceri, followed closely by A. laxa, was the top-ranking treatment, and chickpea cv. BG372 was the best performing cultivar; An-M. ciceri—chickpea cv. BG372 proved to be the superior combination for higher plant growth and soil nutrient-related traits.

The synthesis of poly(ε-L -lysine) is described. This is a poly(ε-amino acid) in which the ε-amino group of lysine is condensed with the α-carboxyl group to produce a chain backbone that is a variant of the usual one seen in proteins and the side chain is the α-amino group. Conformational studies of poly(ε-L -lysine) and its t-butyloxycarbonyl derivative suggest the likelihood of a chain order that is formally similar to the antiparallel pleated-sheet conformation of proteins.  相似文献

To easily edit the genome of naïve human embryonic stem cells (hESC), we introduced a dual cassette encoding an inducible Cas9 into the AAVS1 site of naïve hESC (iCas9). The iCas9 line retained karyotypic stability, expression of pluripotency markers, differentiation potential, and stability in 5iLA and EPS pluripotency conditions. The iCas9 line induced efficient homology–directed repair (HDR) and non-homologous end joining (NHEJ) based mutations through CRISPR-Cas9 system. We utilized the iCas9 line to study the epigenetic regulator, PRC2 in early human pluripotency. The PRC2 requirement distinguishes between early pluripotency stages, however, what regulates PRC2 activity in these stages is not understood. We show reduced H3K27me3 and pluripotency markers in JARID2 2iL-I-F hESC mutants, indicating JARID2 requirement in maintenance of hESC 2iL-I-F state. These data suggest that JARID2 regulates PRC2 in 2iL-I-F state and the lack of PRC2 function in 5iLA state may be due to lack of sufficient JARID2 protein.  相似文献
