Several soft and white rot fungi were compared in their ability to degrade specifically 14C-labelled phenols and dehydropolymers of labelled coniferyl alcohol. Furthermore, plant material, which was expected to be specifically labelled in the lignin part was used in the degradation studies. The experiments showed that both groups of fungi were able to release CO2 from methoxyl and carboxyl groups of phenol-carboxylic acids, to degrade side chains of cinnamic acids and cinnamyl alcohols and even to decompose aromatic structures. With the dehydropolymers and the plant material a CO2 release from the methoxyl groups, the side chains and the aromatic carbons was observed. The time dependant course of the CO2 release from these different groups showed in the beginning a higher CO2 evolution from the side chain carbons than from the methoxyl groups, which were later on released to a higher extent. No laccase activity could be detected in the soft rot fungi and the peroxidase activity was lower than in the white rot fungi.  相似文献

Cold environments at high elevation and high latitude are often viewed as resistant to biological invasions. However, climate warming, land use change and associated increased connectivity all increase the risk of biological invasions in these environments. Here we present a summary of the key discussions of the workshop ‘Biosecurity in Mountains and Northern Ecosystems: Current Status and Future Challenges’ (Flen, Sweden, 1–3 June 2015). The aims of the workshop were to (1) increase awareness about the growing importance of species expansion—both non-native and native—at high elevation and high latitude with climate change, (2) review existing knowledge about invasion risks in these areas, and (3) encourage more research on how species will move and interact in cold environments, the consequences for biodiversity, and animal and human health and wellbeing. The diversity of potential and actual invaders reported at the workshop and the likely interactions between them create major challenges for managers of cold environments. However, since these cold environments have experienced fewer invasions when compared with many warmer, more populated environments, prevention has a real chance of success, especially if it is coupled with prioritisation schemes for targeting invaders likely to have greatest impact. Communication and co-operation between cold environment regions will facilitate rapid response, and maximise the use of limited research and management resources.  相似文献

Bio-composites of oil palm empty fruit bunch (OPEFB) fibres and polycaprolactones (PCL) with a thickness of 1 mm were prepared and characterized. The composites produced from these materials are low in density, inexpensive, environmentally friendly, and possess good dielectric characteristics. The magnitudes of the reflection and transmission coefficients of OPEFB fibre-reinforced PCL composites with different percentages of filler were measured using a rectangular waveguide in conjunction with a microwave vector network analyzer (VNA) in the X-band frequency range. In contrast to the effective medium theory, which states that polymer-based composites with a high dielectric constant can be obtained by doping a filler with a high dielectric constant into a host material with a low dielectric constant, this paper demonstrates that the use of a low filler percentage (12.2%OPEFB) and a high matrix percentage (87.8%PCL) provides excellent results for the dielectric constant and loss factor, whereas 63.8% filler material with 36.2% host material results in lower values for both the dielectric constant and loss factor. The open-ended probe technique (OEC), connected with the Agilent vector network analyzer (VNA), is used to determine the dielectric properties of the materials under investigation. The comparative approach indicates that the mean relative error of FEM is smaller than that of NRW in terms of the corresponding S21 magnitude. The present calculation of the matrix/filler percentages endorses the exact amounts of substrate utilized in various physics applications.  相似文献

Genetic recombination is considered to be a very frequent phenomenon among enteroviruses (Family Picornaviridae, Genus Enterovirus). However, the recombination patterns may differ between enterovirus species and between types within species. Enterovirus C (EV-C) species contains 21 types. In the capsid coding P1 region, the types of EV-C species cluster further into three sub-groups (designated here as A–C). In this study, the recombination pattern of EV-C species sub-group B that contains types CVA-21, CVA-24, EV-C95, EV-C96 and EV-C99 was determined using partial 5′UTR and VP1 sequences of enterovirus strains isolated during poliovirus surveillance and previously published complete genome sequences. Several inter-typic recombination events were detected. Furthermore, the analyses suggested that inter-typic recombination events have occurred mainly within the distinct sub-groups of EV-C species. Only sporadic recombination events between EV-C species sub-group B and other EV-C sub-groups were detected. In addition, strict recombination barriers were inferred for CVA-21 genotype C and CVA-24 variant strains. These results suggest that the frequency of inter-typic recombinations, even within species, may depend on the phylogenetic position of the given viruses.  相似文献

Population size is an important parameter to monitor for species conservation and management. This is especially important for rare and endangered species, as declines can give information about anthropogenic impacts and the need for new conservation measures. To estimate population size, various methods of analysis can be used, for which sample size is an important factor. Sample size is particularly important to consider when applying non-invasive sampling strategies such as sampling faeces or feathers/hairs as a source of DNA, as a means to limit disturbance and stress for the species of concern. We investigated a Black Grouse Lyrurus tetrix population in the eastern part of the Alps, in East Tyrol (Austria), and estimated population size using two approaches: capture–recapture and rarefaction. With a set of 12 polymorphic microsatellite markers, we identified genotypes from faeces and feathers (backed up with 23 tissue samples) and checked for population substructure and gene flow among sampling sites. We estimated population size using four different models from the two approaches (molecular capture–recapture: TIRM, TIRMpart; rarefaction: hyperbolic function – Kohn, exponential function – Eggert). To evaluate the impact of sample size on the estimations, we used the full dataset of 500 samples (‘complete’ dataset) and half the dataset of 250 samples (‘half’ dataset). We also estimated the population size for each sex separately using complete and half datasets to check for sex-specific differences in population size. We found similar results in three of four models (capture–recapture: capwire TIRM, capwire TIRMpart; rarefaction: rarefaction Kohn). Using just half of the data increased the uncertainties in the estimation of population size in all models used and deviations were particularly large in females, which indicated a sex bias. Only the complete dataset of males had an observation rate of more than two observations/individual, and this observation rate meets the recommendation for using the capwire models. This indicates that, for species with different sex-specific detectability, larger sample sizes do not generally imply higher observation rates. We conclude that calculating the observation rates and population-size estimations for each sex separately can improve overall population-size estimation, especially in species with biased sex ratios and those that exhibit sex-specific behaviour.  相似文献

The purpose of this study was to examine how a natural knee responds to the inputs of a total knee replacement testing standard developed by the International Organization for Standardization (ISO). This load control standard prescribes forces to be used for wear testing of knee replacements independent of implant size or design. A parallel ISO standard provides wear testing inputs that are displacement based instead of force based. Eight fresh frozen cadaveric knees were potted and tested in a 6 degree of freedom knee simulator using the load-control standard. The resulting displacements during load-control testing were compared to the prescribed displacements of the ISO displacement standard. At half the tibial torque prescribed by the load standard there was three times more average internal tibial rotation (20.3°) than is prescribed by the displacement standard (5.7°). The AP motion resulting from load testing was much different than is specified by the displacement standard. All eight knees had anterior tibial translation with respect to the femur during swing phase while the displacement standard specifies posterior tibial displacement. The variation in these motions among knees and their difference from the ISO displacement standard may be one factor that explains why wear results of total knee replacements based on ISO load or displacement testing frequently do not agree with each other or with clinical retrievals.  相似文献

Gynecological, endocrinological and histological tests on a 19-year-old female patient led to the diagnosis of Sertoli-Leydig cell tumor (arrhenoblastoma) of intermediate differentiation. For enzyme histochemical purposes the tumor tissue, removed from the right ovary by laparatomy, was frozen in liquid nitrogen. The following enzymes were demonstrated: nonspecific esterases, 3 beta-hydroxysteroid dehydrogenase (HSDH), 17 beta-HSDH, 11 beta-HSDH, and NADH tetrazolium reductase. Cryostat sections, prefixed with formaldehyde vapors, were used to localize testosterone production immunohistochemically with the PAP method. A large number of pseudotubules with Sertoli cells were observed; the Leydig cells in the interstitial space were often arranged in the form of islands. Strong nonspecific esterase activity weak 3 beta-HSDH activity, moderate 17 beta-HSDH activity, and strong 11 beta-HSDH activity were observed largely in the Leydig cells. Testosterone synthesis, demonstrated immunohistochemically, took place predominantly in the Leydig cells, but also to a small extent in the Sertoli cells.  相似文献

C-C Chemokine receptor 6 (CCR6), an important protein in inflammatory and immunological responses, has been previously reported to be associated with rheumatoid arthritis (RA). Therefore, in order to replicate these findings, a case-control study was conducted on 500 subjects (including 250 RA patients and 250 healthy controls) of Pakistani origin. The aim of this study was to determine the association of CCR6 rs3093024 variant with RA and identify its role in splicing events using bioinformatics tools. The clinical and demographic characteristics of the patients were collected using a well-designed questionnaire. The genotype frequencies of CCR6 rs3093024 variant were determined using tetra-primer ARMS-PCR (amplification of refractory mutation system-polymerase chain reaction) method. A significant difference was found between CCR6 rs3093024 genotype frequencies [P = 0.0016, χ2 = 12.915]. Similarly, a significant difference in the allele frequencies between RA patients and healthy controls was also observed [P = 0.0003 and OR (95% CI) = 0.63 (0.49–0.80)]. The stratification of patients showed that there was a significant increase in AA genotype against AG + GG in patients [P = 0.0014, OR (95% CI) = 2.0 (1.32–3.02)]. Furthermore, using bioinformatics analysis, it was found that CCR6 rs3093024 variant might create a potential splicing enhancer motif (SF2/ASF (IgM-BRCA1) with score of 77.92; Threshold 70.53), which might have important impact on the product of this gene. This study suggests that the A variant of CCR6 rs3093024 variant is significantly associated with RA-risk and its G variant is protective in Pakistani population but a multi-cohort large sized population study is needed to elucidate these results. Moreover, functional studies are needed to highlight the effects of this polymorphism on the function of CCR6 gene.  相似文献
