Sonchus oleraceus and Rapistrum rugosum are two rapidly emerging weeds of the northern grain region of Australia. To understand the ability of these weeds regarding their germination response to temperature and different soil moisture regimes, experiments were undertaken on the germination of these weeds at varying osmotic potential and temperature regimes. The experiment was conducted as a split-plot design with alternating day/night temperature regimes (15/5, 20/10, 25/15 and 30/20°C) as a main plot and osmotic potential regimes (0.0, −0.1, −0.2, −0.4, −0.6, −0.8 and −1 MPa) as a subplot. At different temperature regimes, there was 65–91% germination of S. oleraceus in water (0 MPa). There was 0–4% germination at −0.8 MPa and no germination at −1.0 MPa. Osmotic potential values that can cause 50% reduction in germination of S. oleraceus based on a sigmoid regression model ranged from −0.38 to −0.48 MPa. There was 33–81% germination of R. rugosum in distilled water (0 MPa), 1–3% germination at −0.8 MPa and no germination at −1.0 MPa. Osmotic potential values that can cause 50% reduction in germination of R. rugosum based on a sigmoid model ranged from −0.26 to −0.54 MPa. Results of the study were related to the emergence pattern of weeds during field survey and soil moisture profiles estimated by the Australian Landscape Water Balance Model and explain the emergence of these weeds outside the normal seasonal window of prevalence as a response to changes in weather. 相似文献

Several Nocardia and Pseudomonas spp., as well as some unidentified bacteria, isolated from lake water containing high loads of waste lignin, were tested for their capacity to release 14CO2 from specifically 14C-labelled dehydropolymer of coniferyl alcohol (DHP) or corn stalk lignins. The bacteria were selected according to their ability to degrade phenolic compounds. However, only some of them could release significant amounts of 14CO2 from the labelled lignin. The tested Nocardia spp. were more active than the Pseudomonas spp. and the unidentified bacteria. The most active strains belonged to N. autotrophica. These strains released CO2 significantly from the methoxyl group and transformed the other carbons from the phenylpropane skeleton of lignin also into CO2. Other less demethylating strains also released little CO2 from the other carbons of the lignin molecule. Survival of neuronal progenitors (NPCs) is a critical determinant of the regenerative capacity of brain following cellular
loss. Herein, we report for the first time, the increased spontaneous apoptosis of the first acute phase of Experimental Autoimmune
Encephalomyelitis (EAE) derived neurospheres in vitro. Neuronal as well as oligodendroglial loss occurs during experimental
autoimmune encephalomyelitis (EAE). This loss is replenished spontaneously by the concomitant increase in the NPC proliferation
evidenced by the presence of thin myelin sheaths in the remodeled lesions. However, remyelination depends upon the survival
of NPCs and their lineage specific differentiation. We observed significant increase (P < 0.001) in number of BrdU (+) cells in ependymal subventricular zone (SVZ) in EAE rats. EAE derived NPCs showed remarkable
increase in S-phase population which was indeed due to the decrease in G-phase progeny suggesting activation of neuronal progenitor
cells (NPCs) from quiescence. However, EAE derived neurospheres showed limited survival in vitro which was mediated by the
significantly (P < 0.01) depolarized mitochondria, elevated Caspase-3 (P < 0.001) and fragmentation of nuclear DNA evidenced by single cell gel electrophoresis. Our results suggest EAE induced spontaneous
apoptosis of NPCs in vitro which may increase the possibility of early stage cell death in the negative regulation of the
proliferative cell number and may explain the failure of regeneration in human multiple sclerosis. 相似文献

Solvent-accessible surface area calculations have been performed on two human telomeric quadruplex structures, the parallel crystal structure and an (3 + 1) anti-parallel structure determined by NMR methods. The differences in net ligand solvent-accessible surface area (ΔSASA) for four structurally distinct categories of small-molecule ligand have been computed, using docked structures of complexes with both types of quadruplex, as well as the relative contributions of polar and non-polar surface areas. It has been hypothesized that the surface area occupied by the ligand is a determinant of selectivity between different quadruplex topologies, where the ligand maximizes the accessible surface area by contacting all accessible atoms at one end of a quadruplex structure. This has enabled selectivity for a particular ligand to be assessed, for parallel compared an anti-parallel topology. The predictions for the ligands chosen, all of which have their quadruplex topological preferences experimentally determined and reported in the literature, are fully in accord with observation. It is suggested that this approach, which does not depend on energy functions, can be useful in the rational design of topology-specific ligands, especially in the case of polymorphic quadruplexes. 相似文献

The 3-methylcytidine (m3C) modification is widely found in eukaryotic species of tRNA(Ser), tRNA(Thr), and tRNA(Arg); at residue 32 in the anti-codon loop; and at residue e2 in the variable stem of tRNA(Ser). Little is known about the function of this modification or about the specificity of the corresponding methyltransferase, since the gene has not been identified. We have used a primer extension assay to screen a battery of methyltransferase candidate knockout strains in the yeast Saccharomyces cerevisiae, and find that tRNA(Thr(IGU)) from abp140-Δ strains lacks m3C. Curiously, Abp140p is composed of a poorly conserved N-terminal ORF fused by a programed +1 frameshift in budding yeasts to a C-terminal ORF containing an S-adenosylmethionine (SAM) domain that is highly conserved among eukaryotes. We show that ABP140 is required for m3C modification of substrate tRNAs, since primer extension is similarly affected for all tRNA species expected to have m3C and since quantitative analysis shows explicitly that tRNA(Thr(IGU)) from an abp140-Δ strain lacks m3C. We also show that Abp140p (now named Trm140p) purified after expression in yeast or Escherichia coli has m3C methyltransferase activity, which is specific for tRNA(Thr(IGU)) and not tRNA(Phe) and occurs specifically at C??. We suggest that the C-terminal ORF of Trm140p is necessary and sufficient for activity in vivo and in vitro, based on analysis of constructs deleted for most or all of the N-terminal ORF. We also suggest that m3C has a role in translation, since trm140-Δ trm1-Δ strains (also lacking m2,2G??) are sensitive to low concentrations of cycloheximide. 相似文献

The nature of excess body weight may be changing over time to one of greater central adiposity. The aim of this study is to determine whether BMI and waist circumference (WC) are increasing proportionately among population subgroups and the range of bodyweight, and to examine the public health implications of the findings. Our data are from two cross‐sectional surveys (the US National Health and Nutrition Examination Studies (NHANES) in 1988–1994 (NHANES III) and 2005–2006), from which we have used samples of 15,349 and 4,176 participants aged ≥20 years. Between 1988–1994 and 2005–2006 BMI increased by an average of 1.8 kg/m2 and WC by 4.7 cm (adjusted for sex, age, race‐ethnicity, and education). The increase in WC was more than could be attributed simply to increases in BMI. This independent increase in WC (of on average, 0.9 cm) was consistent across the different BMI categories, sexes, education levels, and race‐ethnicity groups. It occurred in younger but not older age groups. Overall in each BMI category, the prevalence of low‐risk WC decreased and the prevalence of increased‐risk or substantially increased‐risk WC increased. These results suggest that the adverse health consequences associated with obesity may be increasingly underestimated by trends in BMI alone. Since WC is closely linked to adverse cardiovascular outcomes, it is important to know the prevailing trends in both of these parameters. 相似文献
