Biologists commonly assume that parasites are locally adapted since they have shorter generation times and higher fecundity than their hosts, and therefore evolve faster in the arms race against the host's defences. As a result, parasites should be better able to infect hosts within their local population than hosts from other allopatric populations. However, recent mathematical modelling has demonstrated that when hosts have higher migration rates than parasites, hosts may diversify their genes faster than parasites and thus parasites may become locally maladapted. This new model was tested on the Canarian endemic lizard and its blood parasite (haemogregarine genus). In this host–parasite system, hosts migrate more than parasites since lizard offspring typically disperse from their natal site soon after hatching and without any contact with their parents who are potential carriers of the intermediate vector of the blood parasite (a mite). Results of cross-infection among three lizard populations showed that parasites were better at infecting individuals from allopatric populations than individuals from their sympatric population. This suggests that, in this host–parasite system, the parasites are locally maladapted to their host.  相似文献

Recombinant DNA technology can be used to design and express collagen and gelatin-related proteins with predetermined composition and structure. Barley seed was chosen as a production host for a recombinant full-length collagen type I α1 (rCIa1) and a related 45-kDa rCIa1 fragment. The transgenic barley seeds were shown to accumulate both the rCIa1 and the 45-kDa rCIa1 fragment. Even when the amount of the rCIa1 was just above the detection threshold, this work using rCIa1 as a model demonstrated for the first time that barley seed can be used as a production system for collagen-related structural proteins. The 45-kDa rCI1a fragment expression, targeted to the endoplasmic reticulum, was controlled by three different promoters (a constitutive maize ubiquitin , seed endosperm-specific rice glutelin and germination-specific barley α - amylase fusion) to compare their effects on rCIa1 accumulation. Highest accumulation of the 45-kDa rCIa1 was obtained with the glutelin promoter (140 mg/kg seed), whereas the lowest accumulation was obtained with the α - amylase promoter. To induce homozygosity for stable 45-kDa rCIa1 production in the transgenic lines, doubled haploid (DH) progeny was generated through microspore culture. The 45-kDa rCIa1 expression levels achieved from the best DH lines were 13 mg/kg dry seeds under the ubiquitin promoter and 45 mg/kg dry seeds under the glutelin promoter. Mass spectroscopy and amino acid composition analysis of the purified 45-kDa rCIa1 fragment revealed that a small percent of prolines were hydroxylated with no additional detectable post-translational modifications.  相似文献

We have studied the effect of 3-nitrobenzothiazolo(3,2-a)quinolinium (NBQ) on the regeneration of the lens in adult newt Notophthalmus viridescens. NBQ has marked cytotoxic effects in tumor cells, intercalates DNA, and was found to enhance lens regeneration. Newt liver DNA was isolated, and the thermal denaturation temperature (Tm) determined to be 76.6% +/- 0.8%. The G-C content was determined to be 44.0% +/- 0.4% and 45.0% +/- 0.1%. Parameters of NBQ binding to newt DNA were determined by spectrophotometric methods and compared with those obtained for calf thymus and Micrococcus lysodeikticus. The association constant, K(o), was found to be 1.1 x 10(+5) M-1 with a site-size parameter, n, of 8.7 nucleotides. No explanation is apparent for the paradoxical stimulation of lens regeneration.  相似文献

Despite high taxonomic diversity elsewhere in North America during the Pleistocene, vertebrate faunas are exceedingly rare in the region of northern Mexico. Térapa, a unique fossil site located in the present-day desert of Northcentral Sonora, Mexico (29°41′N, 109°39′W, 605 m elevation), contributes to our understanding of the paleoecology and paleoclimate of the region during the Late Pleistocene, ca. 43,000-40,000 cal. yr BP. At least 60 vertebrate taxa, including amphibians, turtles, a crocodilian, snakes, birds and many mammals, have been recovered from an 11-m thick sequence of fossiliferous sediments. The diversity and tropical affinity of these taxa suggest a more-forested environment than the thornscrub desert habitat present in this region today.Isotopic analyses of tooth enamel carbonate from ancient mammalian herbivores suggest that the Sonoran desert has undergone considerable climate change since the Late Pleistocene. Bulk carbon (δ13C) and oxygen (δ18O) isotopes from nine mammalian fossils indicate a habitat mosaic with variations in diet that include browsers, mixed feeders and C4 hyper-grazers (δ13C range of − 10‰ to 2‰). Unique to this site are δ13C tooth enamel values of − 6.1 and − 5.6 ‰ for the deer Odocoileus, which suggest a more variable diet than strict browsing, including possibly feeding on CAM and/or C4 plants. Serial sampling of carbon (δ13C) and oxygen (δ18O) isotopes for ancient mammal teeth with hypsodont dentitions (fossil Equus and Bison,) as well as δ18O meteroric water estimates from well-supported climatic models suggest a cooler and more equable environment at Térapa during the Late Pleistocene. These results also support previous habitat reconstructions inferred from the macrobotanical and packrat midden records of northern Sonora (Mexico). High-resolution stable isotope geochemistry indicates that: 1) ancient Térapa was covered with forest and grassland habitats that extended northward into Mexico by about 350 km relative to their present-day northern limits during the Late Pleistocene; and 2) an Amount Effect (AE) is demonstrated in the fossil record at Térapa even though the climate was less seasonal compared to the modern desert habitat.  相似文献

Background

Adults with bipolar disorder (BD) have cognitive impairments that affect face processing and social cognition. However, it remains unknown whether these deficits in euthymic BD have impaired brain markers of emotional processing.

Methodology/Principal Findings

We recruited twenty six participants, 13 controls subjects with an equal number of euthymic BD participants. We used an event-related potential (ERP) assessment of a dual valence task (DVT), in which faces (angry and happy), words (pleasant and unpleasant), and face-word simultaneous combinations are presented to test the effects of the stimulus type (face vs word) and valence (positive vs. negative). All participants received clinical, neuropsychological and social cognition evaluations. ERP analysis revealed that both groups showed N170 modulation of stimulus type effects (face > word). BD patients exhibited reduced and enhanced N170 to facial and semantic valence, respectively. The neural source estimation of N170 was a posterior section of the fusiform gyrus (FG), including the face fusiform area (FFA). Neural generators of N170 for faces (FG and FFA) were reduced in BD. In these patients, N170 modulation was associated with social cognition (theory of mind).

Conclusions/Significance

This is the first report of euthymic BD exhibiting abnormal N170 emotional discrimination associated with theory of mind impairments.  相似文献

Promising approaches to produce higher alcohols, e.g., isobutanol, using Escherichia coli have been developed with successful results. Here, we translated the isobutanol process from shake flasks to a 1-L bioreactor in order to characterize three E. coli strains. With in situ isobutanol removal from the bioreactor using gas stripping, the engineered E. coli strain (JCL260) produced more than 50 g/L in 72 h. In addition, the isobutanol production by the parental strain (JCL16) and the high isobutanol-tolerant mutant (SA481) were compared with JCL260. Interestingly, we found that the isobutanol-tolerant strain in fact produced worse than either JCL16 or JCL260. This result suggests that in situ product removal can properly overcome isobutanol toxicity in E. coli cultures. The isobutanol productivity was approximately twofold and the titer was 9% higher than n-butanol produced by Clostridium in a similar integrated system.  相似文献
