The Chlamydomonas reinhardtii truncated light-harvesting antenna 4 (tla4) DNA transposon mutant has a pale green phenotype, a lower chlorophyll (Chl) per cell and a higher Chl a/b ratio in comparison with the wild type. It required a higher light intensity for the saturation of photosynthesis and displayed a greater per chlorophyll light-saturated rate of oxygen evolution than the wild type. The Chl antenna size of the photosystems in the tla4 mutant was only about 65% of that measured in the wild type. Molecular genetic analysis revealed that a single plasmid DNA insertion disrupted two genes on chromosome 11 of the mutant. A complementation study identified the “chloroplast signal recognition particle 54” gene (CpSRP54), as the lesion causing the tla4 phenotype. Disruption of this gene resulted in partial failure to assemble and, therefore, lower levels of light-harvesting Chl-binding proteins in the C. reinhardtii thylakoids. A comparative in silico 3-D structure-modeling analysis revealed that the M-domain of the CpSRP54 of C. reinhardtii possesses a more extended finger loop structure, due to different amino acid composition, as compared to that of the Arabidopsis CpSRP54. The work demonstrated that CpSRP54 deletion in microalgae can serve to generate tla mutants with a markedly smaller photosystem Chl antenna size, improved solar energy conversion efficiency, and photosynthetic productivity in high-density cultures under bright sunlight conditions.  相似文献

Ubiquitination and deubiquitination of post-translational modification play counter roles in determining the fate of protein function in eukaryotic system for maintaining the cellular homeostasis. Even though novel family members of growth-regulating deubiquitinating enzymes (DUB-1 and DUB-2) have been identified, their target proteins and functions are poorly understood. Dub genes encoding DUB-1 and DUB-2 are immediate-early genes and are induced in response to cytokine stimuli rapidly and transiently. In order to explore the possible proteins regulated by DUB-1, we performed the matrix assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF-MS) analysis followed by immunoprecipitation. We confirmed that DUB-1 interacts with dynein heavy chain, which is known to regulate the movement of organelles and microtubule binding ability. In addition, structural and immunoprecipitation analyses revealed that DUB-1 contains a putative PEST motif and is polyubiquitinated, indicating that DUB-1 is also regulated by the ubiquitin-proteasome pathway.  相似文献

Monocyte chemoattractant protein-1 (MCP-1) influences monocyte migration into sites of inflammation. This study highlights the importance of cytosolic phospholipase A2 (cPLA2)-mediated reactive oxygen species (ROS) signaling processes in the regulation of MCP-1 release as a result of toll-like receptor (TLR) activation. In macrophages, activation of TLR9 induced MCP-1 and cPLA2-phosphorylated arachidonic acid (AA) release. Inhibition of cPLA2 blocked CpG-induced MCP-1 and AA release. Although CpG stimulates phosphorylation of ERK, p38 and JNK, only inhibition of the JNK signaling pathways attenuated MCP-1 release, suggesting that the TLR9-mediated MCP-1 release was dependent upon the JNK pathway. TLR9 activation also stimulated ROS generation, while inhibition of NADPH oxidases (Noxs) blocked CpG-induced MCP-1 release. The CpG treatment increased macrophage Nox1 mRNA level, however it had no effect on macrophage Nox2 mRNA level. Overall, these results suggest that CpG enhances ROS generation through cPLA2-dependent pathways, which results in MCP-1 release.  相似文献

Diruthenium compounds supported by carboxylate or mixed carboxylate/carbonate bridging ligands were found to be active catalysts for aerobic oxygenation of organic sulfides. Ru2(OAc)3(CO3) (A), Ru2(O2CCF3)3(CO3) (B) and Ru2(OAc)4Cl (C) promote the conversion of organic sulfide to sulfoxide, and subsequently sulfone in an oxygen atmosphere at ca. 90 °C. The order of catalytic activity is A > B ? C. Catalytic reactions are operative in a number of 1:1 co-solvent-H2O combinations, and the highest reactivity was found in aqueous media.  相似文献

Recently, the population dynamic systems with impulsive controls have been researched by many authors. However, most of them are reluctant to study the seasonal effects on prey. Thus, in this paper, an impulsively controlled two-prey one-predator system with the Beddington–DeAngelis type functional response and seasonal effects is investigated. By using the Floquet theory, the sufficient conditions for the existence of a globally asymptotically stable two-prey-free periodic solution are established. Further, it is proven that this system is permanent under some conditions via a comparison method involving multiple Lyapunov functions and meanwhile the conditions for extinction of one of the two prey and permanence of the remaining two species are given.  相似文献
