Conventional photomixotrophic micropropagation systems are inefficient due to the high rates of mortality upon the transfer
of plantlets from in vitro to ex vitro conditions. Exogenous medium sugar has been suggested to be the major cause of this
problem. The aim of this study was to investigate the role of sucrose supply on the metabolic profile of in vitro-grown potato
plantlets subjected to different tissue culture conditions consisting of Murashige and Skoog medium and without sucrose [photoautotrophic
(PAT) condition] or with 3% sucrose [photomixotrophic (PMT) condition]. Using gas chromatography–mass spectrometry, we identified
a set of 51 different metabolites in leaf tissues during the rooting phase. Most growth parameters, such as shoot length,
leaf fresh weight, leaf number, and leaf area/plant, were significantly lower under PMT than under PAT conditions. Moreover,
photosynthesis was inhibited due to partial stomatal closure under PMT conditions. The metabolomic profiles along with principal
component analysis and hierarchical cluster analysis revealed that the two treatments were characterized by distinct metabolic
signatures. PAT leaves were characterized by the accumulation of urea and erythritol. In comparison, PMT leaves were characterized
by the accumulation of metabolites belonging to the primary metabolism and catecholamines as well as compounds related to
abiotic stress conditions, such as proline, hydroxyproline, asparagine, γ-aminobutyric acid (GABA), soluble sugars, and myo-inositol. 相似文献

Breast cancer is the second most common cancer diagnosed worldwide. Human epidermal growth factor receptor 2 (HER2)-positive breast cancer represents about 20% to 30% of all breast cancers. Trastuzumab is used in the treatment of HER2-positive breast cancer. MicroRNA-21 (miR-21) is an oncomiR that acts by inhibiting many tumor-suppressor genes. We analyzed the relative expression levels of serum miR-21 in 20 HER2-positive metastatic breast cancer patients before and after 3 months of treatment with trastuzumab. miR-21 levels decreased with a high significant difference after trastuzumab therapy (P = 0.001). Although miR-21 expression levels were lower in responders than in nonresponders, the difference was not statistically significant ( P = 0.6). Our results demonstrated a significant negative correlation between its basal expression, expression levels after treatment, and time to progression ( P = 0.03 and 0.01, respectively). These results make miR-21 a potential prognostic factor for HER2-positive metastatic breast cancer patients. Additionally, it can be an interesting potential target in therapy using antisense oligonucleotides for miR-21. 相似文献

review details our current knowledge of malaria and control strategy in one part of
the Shield, French Guiana. Local epidemiology, test-treat-track strategy, the state
of parasite drug resistance and vector control measures are summarised. Current
issues in terms of mobile populations and legislative limitations are also

In this paper, a coupled bioluminescent assay, relying on the coupling of the enzymes acetylcholinesterase, S‐acetyl‐coenzyme A synthetase and firefly luciferase, for the detection and quantitation of organophosphorus pesticides, is presented. Using malathion as a model organophosphorus pesticide, the assay was optimized through statistical experimental design methodology, namely Plackett–Burman and central composite designs. The optimized method requires only 20 μL of sample. The linear range for the assay was 2.5–15 μM of malathion, with limits of detection and quantitation of 1.5 and 5.0 μM, respectively. Snake venoms are cocktails comprising combinations of different proteins, peptides, enzymes and toxins. Snake toxins have
diverse characteristics having different molecular configuration, structure and mode of action. Many toxins derived from snake
venom have distinct pharmacological activities. Venom from Bungarus fasciatus (commonly known as banded krait) is a species of
elapid snake found on the South East Asia and Indian sub-continent, mainly contains neurotoxins. Beta bungartotoxin is the major
fraction of Bungarus venom and particularly act pre-synaptically by obstructing neurotransmitter release. This toxin in other
snake species functionally forms a heterodimer containing two different subunits (A and B). Dimerization of these two chains is a
pre-requisite for the proper functionality of this protein. However, B. fasciatus bungartotoxin contains only B chain and their
structural orientation in yet to be resolved. Therefore, it is of interest to describe the predicted structure model of the toxin for
functional insights. In this work we analyzed the neurotoxic nature, their alignments, secondary and three dimensional structures,
functions, active sites and stability with the help of different bioinformatical tools. A comprehensive analysis of the predicted model

Immune surveillance system can detect more efficiently secretory tumor-specific antigens, which are superior as a target for cancer immunotherapy. On the contrary, immune tolerance can be induced in the thymus when a tumor antigen is massively secreted into circulation. Thus, the secretion of tumor-specific antigen may have contradictory roles in tumor immunity in a context-dependent manner. However, it remains elusive on the precise cellular mechanism of intrathymic immune tolerance against tumor antigens. We previously demonstrated that a minor thymic conventional dendritic cell (cDC) subset, CD8α(-)Sirpα(+) cDCs, but not the major subset, CD8α(+)Sirpα(-) cDCs can selectively capture blood-borne antigens and crucially contribute to the self-tolerance. In the present study, we further demonstrated that Sirpα(+) cDCs can capture a blood-borne antigen leaking inside the interlobular vascular-rich regions (IVRs). Blood-borne antigen selectively captured by Sirpα(+) cDCs can induce antigen-specific Treg generation or negative selection, depending on the immunogenicity of the presented antigen. Furthermore, CCR2 expression by thymic Sirpα(+) cDCs and abundant expression of its ligands, particularly, CCL2 by tumor-bearing mice prompted us to examine the function of thymic Sirpα(+) cDCs in tumor-bearing mice. Interestingly, tumor-bearing mice deposited CCL2 inside IVRs in the thymus. Moreover, tumor formation induced the accumulation of Sirpα(+) cDCs in IVRs under the control of CCR2-CCL2 axis and enhanced their capacity to take up antigens, resulting in the shift from Treg differentiation to negative selection. Finally, intrathymic negative selection similarly ensued in CCR2-competent mice once the tumor-specific antigen was secreted into bloodstream. Thus, we demonstrated that thymic Sirpα(+) cDCs crucially contribute to this novel process of intrathymic tumor immune tolerance. 相似文献

The analysis of morphological variation and RAPD polymorphism distinguished populations of A. herba alba from populations of A. judaica and A. monosperma. Higher morphological diversity is found in A. herba alba compared to the other two species, but molecular data derived from RAPD polymorphism also indicated that A. herba alba is more polymorphic than the other two species. However, RAPD fingerprinting also indicated sharp polymorphism among populations
of both A. judaica and A. monosperma. Geographic and local ecological variations related to elevation of the sites of the examined populations may be regarded
to have played a role in the genetic diversity of the examined populations of Artemisia species in the study area. The results are important for future plans for sustainable conservation of medicinal plants in

In the present study we evaluated the anti-tumor potential of samsum ant venom (SAV) from Pachycondyla sennaarensis on the human breast carcinoma cell line MCF-7. We found that SAV induced growth arrest of MCF-7 cells without affecting the viability of MCF-10 (non-tumorigenic normal breast epithelial cells) and normal PBMCs. We then analyzed its impact on IGF-1-mediated MCF-7 cell proliferation and its effect on the underlying IGF-1 signaling pathways. Using flow cytometry analysis, we showed that the percentage of apoptotic cells was fourfold higher in SAV-treated cells as compared to untreated cells. More importantly, treatment with SAV induced a marked reduction in actin polymerization and a subsequent marked reduction in IGF-1-mediated cell proliferation. In addition to growth-inhibitory and pro-apoptotic effects, significant reductions were also observed in the phosphorylation of AKT and ERK, but not p38MAPK, in SAV-treated cells as compared to untreated cells. The genetic diversity and ancestral relationships of a number of Trifolium species was revealed by using the amplified fragment length polymorphism (AFLP) and the random amplified polymorphic DNA
(RAPD) markers. Both markers produced few species-specific markers. Using distance and parsimony methods, in NTSYS-pc and PAUP software programs, we clearly differentiated the accessions of white clover from other closely related progenitors. The phylogenetic
trees, produced by PAUP, also reinforced the close affinity of T. nigrescens and the allopolyploid white clover in support of former views that this diploid species could have been the donor of one
of two genomes of the allotetraploid T. repens. In addition, the dendrograms, produced by NTSYS-pc, also indicated close affinity of T. nigrescens and T. occidentale to the accessions of T. repens. These data is congruent with karyological and phylogenetic affinities between the white clover and T. occidentale. The relationships between the examined accessions, in the T. repens gene pool, may be regarded to indicate the presence of shared alleles between T. repens, T. occidentale and T. uniflorum. Further, T. occidentale showed close phylogenetic relations to T. pallescens. 相似文献
