The amounts of DNA in interphase nuclei were compared with the amounts of DNA in metaphase and anaphase figures in Feulgen-stained tissue sections of 5 specimens of the human ovarian papillary serous adenocarcinoma. The relative amounts of DNA per cell were determined by cytophotometric measurements of interphase nuclei at a single wavelength and of mitotic figures by the two wavelength method. The 5 specimens conformed to the stem cell concept of cell proliferation since anaphase distributions of amounts of DNA were restricted to a narrow range of DNA values indicating the successful mitosis of a single cell type (stem cell) out of several cell types whose presence were suggested by the wide spread of interphase and metaphase values. In addition, the data indicated that, in some instances, only the amounts of DNA in anaphase figures can reliably identify the stem cell. Changes in the frequency of dividing cells having doubled amounts of DNA, and/or the presence of cells resulting from endoreduplication can distort the interphase distribution of amounts of DNA and thus give rise to a modal DNA interphase value which is not the same as the DNA value of the stem cell (anaphase figures). 相似文献

The stratified squamous epithelium of the oral gingiva and the hard palate is characterized by a tissue architecture and a cytoskeletal composition similar to, although not identical with, that of the epidermis and fundamentally different from that of the adjacent non-masticatory oral mucosa. Using immunocytochemistry with antibodies specific for individual cytokeratins, in situ hybridization and Northern blots of RNA with riboprobes specific for individual cytokeratin mRNAs, and gel electrophoresis of cytoskeletal proteins of microdissected biopsy tissue samples, we show changes in the pattern of expression of cytokeratins and their corresponding mRNAs in pathologically altered oral gingiva. Besides a frequently, although not consistently, observed increase in the number of cells producing cytokeratins 4 and 13 (which are normally found as abundant components in the sulcular epithelium and the alveolar mucosa but not in the oral gingiva) and a reduction in the number of cells producing cytokeratins 1, 10 and 11, the most extensive change was noted for cytokeratin 19, a frequent cytokeratin in diverse one-layered and complex epithelia. While in normal oral gingiva cytokeratin 19 is restricted to certain, sparsely scattered cells of --or near--the basal cell layer, probably neuroendocrine (Merkel) cells, in altered tissue of inflamed samples it can appear in larger regions of the basal cell layer(s) and, in apparently more advanced stages, also in a variable number of suprabasal cells. Specifically, our in situ hybridization experiments show that this altered suprabasal cytokeratin 19 expression is more extended at the mRNA than at the protein level, indicating that cytokeratin 19 mRNA synthesis may be a relatively early event during the alteration. These changes in cytokeratin expression under an external pathological influence are discussed in relation to other factors known to contribute to the expression of certain cytokeratins and with respect to changes occurring during dysplasia and malignant transformation of oral epithelia. 相似文献

The effects of 17 beta-estradiol (estradiol), synthetic progestin R5020 and their antagonists, tamoxifen (Tam) and synthetic RU38486 on lactate dehydrogenase (LDH) activity in MCF-7 human breast cancer cells during the growth period were studied. A specially developed quantitative cytochemical assay was used; LDH activity is expressed per cell, and is thus independent of the positive and negative growth effects of the hormones and antagonists. Estradiol and R5020 stimulated LDH activity after similar exposures (6-48 h) and the stimuli were concentration dependent over the range 10(-7) M to 10(-10) M. As for the antagonists, RU38486 stimulated LDH activity in much the same way as estradiol and R5020; Tam alone, on the other hand, does not stimulate LDH, but when added to estradiol, Tam inhibits estradiol mediated LDH activation. When present at half-stimulant concentration, estradiol + R5020 and estradiol + RU38486 exhibit additive effects on LDH activity. Thus LDH appears to be an interesting tool for the study of hormone and antagonist effects in MCF-7 breast cancer cells. 相似文献

Numerous genital papillae are described for the first time in the freshwater miteHydrovoliza placophora. In contrast to other actinotrichid mites, these organs are located on the epimeral plates, thus well removed from the genital opening. TGR(mREN2)27 is a transgenic rat harboring the murine Ren-2 gene and exhibit fulminant hypertension and marked heart hypertrophy. In order to study the role of angiotensin II in the increase of cardiac mass, these animals were treated with anti-hypertensive and non-antihypertensive doses of the angiotensin II receptor AT1 antagonist Telmisartan for 9 weeks. All doses led to significant reductions of heart hypertrophy detected by the evaluation of the diameter of cardiac muscle bundles. We conclude from this study that cardiac hypertrophy in TGR(mREN2)27 is characterized by an increased volume of cardiomyocytes and an unchanged amount of fibrous tissue and that angiotensin II plays an important role in the mechanisms leading to this phenotype. 相似文献

In the United States, the Native American Graves Protection and Repatriation Act (NAGPRA) provides a specific framework for the disposition of Native American Ancestral remains within its purview. However, samples such as a bone fragment, tooth, or other biological tissue taken from the remains of these Ancestors have been treated by institutions and researchers as independent of the individual from whom they were removed and used in destructive research such as paleogenomic and other archaeometric analyses without consultation, consent, and collaboration from Native American communities; are not cared for in keeping with the current best practices for Indigenous Ancestors; and are not likely to be repatriated to their communities. Here, we demonstrate that any biological samples removed from Ancestors who are covered under NAGPRA must also be handled according to the stipulations defined for “human remains” within the legislation. As such, we are not proposing a change to existing legislation, but rather best practices, specific to the context of the United States and NAGPRA, relating to the use of and care for biological samples taken from Native American Ancestors. 相似文献

Human gastric mucosal biopsies incorporate in vitro radioactive proline and fucose into macromolecular glycoproteins (mucin). Differences were found between the incorporation pattern of antral and fundic mucosae according to their pathology, confirmed by histology. Antral mucosae with abnormal histology showed a significantly higher incorporation of proline than normal samples. Fucose incorporation was also increased. Similar results were found with fundic biopsies. The ratio of total fucose to total proline incorporated into mucin secreted during incubation also increased significantly in these samples. Biochemical analyses on the other hand showed no significant change. The results suggest a breakdown in the processing, storage and secretory processes of mucin-type glycoproteins in pathological mucosae, but not in their biosynthesis. The mucosal samples could be classified as A or B types according to their proline and fucose incorporation: A mucosae have a lower proline incorporation than B mucosae (greater than 380 cpm/micrograms DNA for the antrum and greater than 200 cpm/micrograms DNA for the fundus). These results confirm the possibility of studying abnormal mucus secretion using gastric biopsy samples. 相似文献
