Endophyte-mediated adjustments in host morphology and physiology and effects on host fitness traits in grasses

Endophytic fungi have been shown to increase tolerance of hosts to biotic and abiotic stresses and in some cases alter growth and development of plants. In this article we evaluate some effects that clavicipitaceous endophytes have on development and physiology of plant tissues. We postulate that oxidative stress protection is the fundamental underlying benefit conferred by many endophytes, accounting for frequently observed enhanced disease resistance, drought tolerance, heavy metal tolerance and tolerance to numerous additional oxidative stresses. We hypothesize that endophyte-mediated oxidative stress protection of the host is the result of at least two processes, including: (1) secretion of reactive oxygen species (ROS) from endophytic mycelia into plant cells; and (2) secretion of auxin from endophytic mycelia into plant cells. Both processes result in an increase in ROS in plant tissues; and stimulate plant tissues to increase activities of antioxidant systems. Auxin is suggested to function in suppression of plant cell death and may be important in maintaining the endophyte–plant symbiosis.     

Metabolism of Glutamic Acid in Aspergillus ochraceus During the Biosynthesis of Ochratoxin A

The uptake and utilization of glutamic acid in the biosynthesis of ochratoxin A by Aspergillus ochraceus were studied. Uniformly labeled L[14C]glutamic acid was incorporated into both the phenylalanine and isocoumarin moieties of ochratoxin A. Penicillic acid was also labeled. During the early stages of development, the amino acid was used mainly for the synthesis of ribonucleic acid and protein. A portion of glutamic acid was oxidized and was recovered as metabolic 14CO-2. The initial uptake velocity of glutamic acid decreased with age and was pH and temperature dependent. No relationship was found between the initial uptake velocities and ochratoxin A biosynthesis.     

Three-dimensional structural analysis of the group B polysaccharide of Neisseria meningitidis 6275 by two-dimensional NMR: the polysaccharide is suggested to exist in helical conformations in solution

The solution conformations of the group B polysaccharide of Neisseria meningitidis were analyzed by DQF-COSY and pure absorption 2D NOE NMR with three mixing times. The pyranose ring of the sialic acid residue was found to be in the 2C5 conformation. The DQF-COSY analysis indicated that the orientations of H6 and H7 and of H7 and H8 are both gauche. In order to overcome the difficulties in analyzing the NOE data due to the two sets of proton overlaps, molecular modeling of alpha-2,8-linked sialic acid oligomers was carried out to investigate possible conformers, and theoretical NOE calculations were performed by using CORMA (complete relaxation matrix analysis). Our analysis suggests that the polysaccharide adopts helical structures for which the phi (defined by O6-C2-O8-C8) and psi (C2-O8-C8-C7) angles are in the following ranges: phi -60 to 0 degrees, psi 115-175 degrees or phi 90-120 degrees, psi 55-175 degrees. The weak affinity of anti-B antibodies for smaller alpha-2,8-linked oligosaccharides may be due to the fact that such oligomers are more flexible and may not form an ordered structure as the poly(sialic acid) does.     

Oligodeoxynucleoside phosphorothioate stability in subcellular extracts, culture media, sera and cerebrospinal fluid

Degradation of a synthetic oligodeoxynucleoside phosphorothioate was studied in six systems used for antisense inhibition experiments. Oligodeoxynucleoside phosphorothioates were degraded very slowly at 37 degrees C in all of the systems studied. Measured half-lives of pentadecamers were 12 +/- 1 h in rabbit reticulocyte lysate, 7 +/- 1 h in HeLa cell postmitochondrial extract, 14 +/- 2 h in RPMI 1640 with 10% fetal bovine serum, 8 +/- 1 h in undiluted fetal bovine serum, 9 +/- 1 h in adult human serum, and 19 +/- 7 h in rat cerebrospinal fluid.     

The influence of three days or three weeks of force feeding on the transport of plasma lipids in young female chickens (Gallus gallus L.).

1. Ten-week old broiler females were force-fed (FF) for 3 days or 3 weeks. 2. Control livers were lighter in weight and contained less total lipid, neutral lipid and phospholipid than either FF group, which did not differ. 3. Radioactivity incorporated into liver neutral lipid fractions from 1-[14C]acetate injection was greater in birds FF 3 weeks than controls. Those FF 3 days were intermediate. In all groups, the triglyceride fraction contained 90-94% of isolated radioactivity, the cholesterol fraction 4-8% and the cholesterol ester fraction 1-2%. 4. Plasma lipids were elevated in the birds FF for 3 weeks, but not in those FF 3 days. After injection of 1-[14C]acetate, plasma lipid specific radioactivities were not different between the 3 groups at 20 and 60 min post injection, but were greater in the controls at 120 min.     

Autoimmune Haemolytic Anaemia and Mefenamic Acid Therapy

Three patients developed autoimmune haemolytic anaemia while being treated with mefenamic acid. In each case the autoimmune haemolytic anaemia was of the warm antibody γG type, and the antibodies had some rhesus specificity. All three patients recovered when the drug was withdrawn.Attempts to inhibit or enhance the activity of the antibody in vitro were unsuccessful.Direct antihuman globulin tests were made in.the red cells of 36 patients receiving long-term mefenamic acid therapy, but only one was found to be transitorily positive.
