Summary
Lambda repressor was purified from an E. coli strain which produces 150 times more lambda repressor than a single lysogen. The sequence of the fifty N-terminal residues was determined by automated Edman degradation. It contains 43% of all arginine and lysine residues of the chain and constitutes according to the genetic data of Oppenheim et al. G protein-coupled receptor 3 (GPR3) is a constitutively active receptor that maintains high 3′-5′-cyclic adenosine monophosphate (cAMP) levels required for meiotic arrest in oocytes and CNS function. Ligand-activated G protein-coupled receptors (GPCRs) signal at the cell surface and are silenced by phosphorylation and β-arrestin recruitment upon endocytosis. Some GPCRs can also signal from endosomes following internalization. Little is known about the localization, signaling, and regulation of constitutively active GPCRs. We demonstrate herein that exogenously-expressed GPR3 localizes to the cell membrane and undergoes internalization in HEK293 cells. Inhibition of endocytosis increased cell surface-localized GPR3 and cAMP levels while overexpression of GPCR-Kinase 2 (GRK2) and β-arrestin-2 decreased cell surface-localized GPR3 and cAMP levels. GRK2 by itself is sufficient to decrease cAMP production but both GRK2 and β-arrestin-2 are required to decrease cell surface GPR3. GRK2 regulates GPR3 independently of its kinase activity since a kinase inactive GRK2-K220R mutant significantly decreased cAMP levels. However, GRK2-K220R and β-arrestin-2 do not diminish cell surface GPR3, suggesting that phosphorylation is required to induce GPR3 internalization. To understand which residues are targeted for desensitization, we mutated potential phosphorylation sites in the third intracellular loop and C-terminus and examined the effect on cAMP and receptor surface localization. Mutation of residues in the third intracellular loop dramatically increased cAMP levels whereas mutation of residues in the C-terminus produced cAMP levels comparable to GPR3 wild type. Interestingly, both mutations significantly reduced cell surface expression of GPR3. These results demonstrate that GPR3 signals at the plasma membrane and can be silenced by GRK2/β-arrestin overexpression. These results also strongly implicate the serine and/or threonine residues in the third intracellular loop in the regulation of GPR3 activity. 相似文献

The study aimed to elucidate previously observed associations between morningness–eveningness and seasonality by analysing their distinct aspects separately: morning affect (MA) and time-of-day preference, different seasonal types and patterns (winter, summer, etc.), the degree of seasonality and perceived negative impact of seasonality. Students from Warsaw (N = 522) completed the Seasonal Pattern Assessment Questionnaire and the Composite Scale of Morningness. Winter seasonality was related to lower MA, but unrelated to time-of-day preference. Global seasonality score was negatively associated with MA in winter seasonality, but not in other seasonality patterns, and unrelated to time-of-day preference. These associations remained significant after controlling for sex, age and season of assessment. It is concluded that winter seasonality is related to low MA, but not to time-of-day preference. The above results indicate that MA can be considered as an all year round indicator of proneness to winter seasonality and eventually to seasonal affective disorder. The results also suggest that MA and time-of-day preference should be analysed separately in future research on morningness–eveningness. 相似文献

ABSTRACT There is growing interest in personality profiles associated with morningness–eveningness to foster an understanding of the behaviors and mental states of chronotypes. This study aimed to analyze the domains of emotionality, activity and sociability (EAS) temperament in relation to morningness–eveningness in adolescents. A sample of 539 school pupils aged 13–19 years completed the EAS Temperament Survey, the Composite Scale of Morningness and the Munich Chronotype Questionnaire. Amongst the five EAS domains (emotionality-distress, emotionality-fearfulness, emotionality-anger, activity and sociability), greater emotionality-anger was related to eveningness, while greater emotionality-distress was related to lower social jet lag. The results suggest that evening chronotypes can be temperamentally inclined to anger. The possible mechanisms of this association are discussed. 相似文献

Background
Genetic factors and a dysregulated immune response towards commensal bacteria contribute to the pathogenesis of Inflammatory Bowel Disease (IBD). Animal models demonstrated that the normal intestinal flora is crucial for the development of intestinal inflammation. However, due to the complexity of the intestinal flora, it has been difficult to design experiments for detection of proinflammatory bacterial antigen(s) involved in the pathogenesis of the disease. Several studies indicated a potential association of E. coli with IBD. In addition, T cell clones of IBD patients were shown to cross react towards antigens from different enteric bacterial species and thus likely responded to conserved bacterial antigens. We therefore chose highly conserved E. coli proteins as candidate antigens for abnormal T cell responses in IBD and used high-throughput techniques for cloning, expression and purification under native conditions of a set of 271 conserved E. coli proteins for downstream immunologic studies. 相似文献996.

Neurons in the sensory system exhibit changes in excitability that unfold over many time scales. These fluctuations produce noise and could potentially lead to perceptual errors. However, to prevent such errors, postsynaptic neurons and synapses can adapt and counteract changes in the excitability of presynaptic neurons. Here we ask how neurons could optimally adapt to minimize the influence of changing presynaptic neural properties on their outputs. The resulting model, based on Bayesian inference, explains a range of physiological results from experiments which have measured the overall properties and detailed time-course of sensory tuning curve adaptation in the early visual cortex. We show how several experimentally measured short term plasticity phenomena can be understood as near-optimal solutions to this adaptation problem. This framework provides a link between high level computational problems, the properties of cortical neurons, and synaptic physiology. 相似文献
