The recent revival of old theories and setting them on modern scientific rails to a large extent are also relevant to mitochondrial science. Given the widespread belief that mitochondria are symbionts of ancient bacterial origin, the processes inherent to mitochondrial physiology can be revised based on their comparative analysis with possible involvement of bacteria. Such comparison combined with discussion of the role of microbiota in pathogenesis allows discussion of the role of “mitobiota” (we introduce this term) as the combination of different phenotypic manifestations of mitochondria in the organism reflecting pathological changes in the mitochondrial genome. When putting an equal sign between mitochondria and bacteria, we find similarity between the mitochondrial and bacterial theories of cancer. The presence of the term “bacterial infection” suggests “mitochondrial infection”, and mitochondrial (oxidative) theory of aging can in some way be transformed into a “bacterial theory of aging”. The possible existence of such processes and the data confirming their presence are discussed in this review. If such a comparison has the right to exist, the homeostasis of “mitobiota” is of not lesser physiological importance than homeostasis of microbiota, which has been so intensively discussed recently.  相似文献

The effects of exogenous phytohormones–gibberellic acid (GA3) and benzyl-aminopurine (BAP)–on the spore germination and morphogenesis of Polystichum aculeatum (L.) Roth gametophyte in vitro culture were studied. In the control, four stages of gametophyte morphogenesis were determined and their periods were established. Spore germination and protonema formation of P. aculeatum occurred according to the Vittaria-type and prothalium development according to the Aspidium-type. The spore germination percentage depended on the storage time duration. It was found that 80–95% of freshly collected spores germinated. Spore viability was within the range of 68–95% after 4–6-month storage under lab conditions and did not exceed 20% after 1.5 year of the storage period. High concentrations of exogenous GA3 (10–5 and 10–6 M) and BAP (10–5 M) significantly inhibited spore germination, whereas low concentrations (GA3 10–7–10–8 M) had an insignificant stimulating effect or did not affect germination at all (BAP 10–6, 10–7, 10–8 M). All concentrations of exogenous BAP were demonstrated to inhibit the development of P. aculeatum gametophyte at the protonema stage, which might be due to the removal of apical dominance. The inhibiting effect directly depended on BAP concentrations. The formation of abnormal thalli of the P. aculeatum gametophyte in the response to exogenous GA3 treatments occurred as a result of impairment of cell growth by elongation. A direct interrelationship between GA3 concentrations and level of morphological abnormalities and grade of thalli underdevelopment was demonstrated.  相似文献

The metabolism of nuclear phospholipid acyl components of liver in uneven-aged rats was studied in vitro under different dietary fat implications. The activity of phospholipases A1 and A2 in the nuclei was found to sharply increase in animals pretreated with excess of fat. The incorporation of labelled palmitic, oleic, linoleic and arachidonic acid into nuclear phospholipids is under control of age and diet manipulations. The observed changes in the level of fatty acids metabolism are more pronounced in cell nuclei of the young rat liver. The lipid composition of cell nuclei in the test 3-month old animals does not differ from that of the control animals. At the same time dietary implications induce deep changes in the composition of nuclear lipids in 24-months old animals.  相似文献

Ceramides are a novel class of biologically active molecules involved in the regulation of different signaling pathways. Ceramide is involved in regulation of the phospholipase D (PLD) activity and development of cell resistance to insulin. In this work, we have studied age-related features of insulin regulation of PLD activity and glucose metabolism in intact cells and modeled their resistance to insulin by exogenous ceramide and palmitic acid. Contents of ceramides and of free fatty acids (FFA) are found to increase with age, as well as on incubation of liver cells of young rats in the presence of the ceramide precursor palmitic acid. Under these conditions, the ability of insulin to activate PLD, the cell uptake of glucose, and glycogen synthesis sharply decreased. On incubation of hepatocytes of young animals in the presence of exogenous C2-ceramide, the contents of endogenous ceramides increased but not the contents of FFAs and of neutral lipids. These events were accompanied by suppression of the insulin-induced production of phosphatidylethanol (a result of ethanol transphosphatidylation by PLD), glucose uptake, and glycogen synthesis. Incubation of insulin-resistant liver cells of young rats and also of hepatocytes of old rats in the presence of myriocin (an inhibitor of the de novo synthesis of ceramide) was associated with a decrease in ceramide content in the cells and an increase in the cell sensitivity to insulin. The findings indicate an important role of ceramide in disturbance of insulin signaling due to inhibition of the PLD-dependent link in the liver cells of old animals.  相似文献
