Background
A large number of microsatellites have been extensively used to comprehend the genetic diversity of different global groups. This paper entails polymorphism at 15 STR in four predominant and endogamous populations representing Karnataka, located on the southwest coast of India. Determining which animal viruses may be capable of infecting humans is currently intractable at the time of their discovery, precluding prioritization of high-risk viruses for early investigation and outbreak preparedness. Given the increasing use of genomics in virus discovery and the otherwise sparse knowledge of the biology of newly discovered viruses, we developed machine learning models that identify candidate zoonoses solely using signatures of host range encoded in viral genomes. Within a dataset of 861 viral species with known zoonotic status, our approach outperformed models based on the phylogenetic relatedness of viruses to known human-infecting viruses (area under the receiver operating characteristic curve [AUC] = 0.773), distinguishing high-risk viruses within families that contain a minority of human-infecting species and identifying putatively undetected or so far unrealized zoonoses. Analyses of the underpinnings of model predictions suggested the existence of generalizable features of viral genomes that are independent of virus taxonomic relationships and that may preadapt viruses to infect humans. Our model reduced a second set of 645 animal-associated viruses that were excluded from training to 272 high and 41 very high-risk candidate zoonoses and showed significantly elevated predicted zoonotic risk in viruses from nonhuman primates, but not other mammalian or avian host groups. A second application showed that our models could have identified Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) as a relatively high-risk coronavirus strain and that this prediction required no prior knowledge of zoonotic Severe Acute Respiratory Syndrome (SARS)-related coronaviruses. Genome-based zoonotic risk assessment provides a rapid, low-cost approach to enable evidence-driven virus surveillance and increases the feasibility of downstream biological and ecological characterization of viruses.Surveillance of emerging viruses is one of the first steps to avoid the next pandemic. This study uses machine learning to identify many zoonotic viruses directly from their genomes. This allows rapid assessment of research priorities as soon as new viruses are discovered, focusing research and surveillance efforts on the viruses most likely to infect humans. 相似文献

The releasable factor adenosine blocks the formation of long-term potentiation (LTP). These experiments used this observation to uncover the synaptic processes that stabilize the potentiation effect. Brief adenosine infusion blocked stimulation-induced actin polymerization within dendritic spines along with LTP itself in control rat hippocampal slices but not in those pretreated with the actin filament stabilizer jasplakinolide. Adenosine also blocked activity-driven phosphorylation of synaptic cofilin but not of synaptic p21-activated kinase (PAK). A search for the upstream origins of these effects showed that adenosine suppressed RhoA activity but only modestly affected Rac and Cdc42. A RhoA kinase (ROCK) inhibitor reproduced adenosine''s effects on cofilin phosphorylation, spine actin polymerization, and LTP, whereas a Rac inhibitor did not. However, inhibitors of Rac or PAK did prolong LTP''s vulnerability to reversal by latrunculin, a toxin which blocks actin filament assembly. Thus, LTP induction initiates two synaptic signaling cascades: one (RhoA-ROCK-cofilin) leads to actin polymerization, whereas the other (Rac-PAK) stabilizes the newly formed filaments. 相似文献

Human lymphatic filariasis is a major tropical disease transmitted through mosquito vectors which take up microfilarial larvae from the blood of infected subjects. Microfilariae are produced by long-lived adult parasites, which also release a suite of excretory-secretory products that have recently been subject to in-depth proteomic analysis. Surprisingly, the most abundant secreted protein of adult Brugia malayi is triose phosphate isomerase (TPI), a glycolytic enzyme usually associated with the cytosol. We now show that while TPI is a prominent target of the antibody response to infection, there is little antibody-mediated inhibition of catalytic activity by polyclonal sera. We generated a panel of twenty-three anti-TPI monoclonal antibodies and found only two were able to block TPI enzymatic activity. Immunisation of jirds with B. malayi TPI, or mice with the homologous protein from the rodent filaria Litomosoides sigmodontis, failed to induce neutralising antibodies or protective immunity. In contrast, passive transfer of neutralising monoclonal antibody to mice prior to implantation with adult B. malayi resulted in 60–70% reductions in microfilarial levels in vivo and both oocyte and microfilarial production by individual adult females. The loss of fecundity was accompanied by reduced IFNγ expression by CD4+ T cells and a higher proportion of macrophages at the site of infection. Thus, enzymatically active TPI plays an important role in the transmission cycle of B. malayi filarial parasites and is identified as a potential target for immunological and pharmacological intervention against filarial infections. 相似文献

of CUBN in evolution using phylogenetic testing. A total of 588 BLAST hits were found for the cubilin query sequence and these
hits showed putative conserved domain, CUB superfamily (as on 27th Nov 2012). A first-pass phylogenetic tree was constructed to
identify the taxa which most often contained the CUBN sequences. Following this, we narrowed down the search by manually
deleting sequences which were not CUBN. A repeat phylogenetic analysis of 25 taxa was performed using PhyML, RAxML and
TreeDyn softwares to confirm that CUBN is a conserved protein emphasizing its importance as an extracellular domain and being
present in proteins mostly known to be involved in development in many chordate taxa but not found in prokaryotes, plants and

The fungitoxic effect of various medicinal plants belonging to different families was evaluated in vitro on Rhizoctonia solani, the rice sheath blight pathogen. Of the various plant extracts, the leaf extract of zimmu (Allium cepa × Allium sativum) showed the maximum antifungal activity against R. solani and recorded an inhibition zone of 12?mm. The leaf extract of zimmu was also effective in inhibiting the growth of other agronomically important fungal and bacterial pathogens viz., Aspergillus flavus, Curvularia lunata, Alternaria solani, Xanthomonas oryzae pv. oryzae, Xanthomonas campestris pv. malvacearum and Xanthomonas axonopodis pv. citri. The antimicrobial compound was dissoluble in methanol and the methanolic extract showed the absorption maxima at 210?nm and 230?nm. Phenolic compounds were present in greater amounts in methanol extract of zimmu. TLC analysis showed the appearance of two blue spots at R f ?=?0.65 and R f ?=?0.90. The compounds eluted at R f ?=?0.65 and R f ?=?0.90 by preparative TLC exhibited strong antifungal activity against R. solani. 相似文献

This paper attempts to pinpoint the most original morphological anatomical features of the biology of filariae per se and those which are or could be important for triggering regulatory processes in the arthropod vector and uncontrolled pathogenic processes in the vertebrate hosts. The following stages are considered: the motile egg or newly-hatched larva, the microfilaria, in the lymphatic or blood vessels of its vertebrate host; the larva, its migrations and its intrasyncitial development in the hematophagous arthropod subverted as vector; its transfer to the vertebrate host, migratory properties through the lymphatic system, maturation, mating and, finally, egg laying in the tissues they reach. This synthesis is based on parasite morphological features and their functional interpretation, histological features in the different niches the filariae reach, and on quantitative analyses of filarial development at its different phases, as well as on the rare and valuable observations of living parasites in situ. Data have been drawn from various species of Onchocercidae from amphibians, reptiles, birds and mammals. These comparative analyses have revealed the major constraints to which the filariae, including those parasitizing humans, have been subjected during their evolution from their ancestors, the oviparous and heteroxenic spirurids. Emphasis is placed on mechanical events: resistance of the microfilariae to the currents in the blood or lymph vessels, regulatory processes induced in the vector mesenteron by the movements of the ingested microfilariae, transient disruption by the microfilarial cephalic hook of the vectors' tissues and cell membranes during microfilarial translocation, attachment of males to females during mating by means of 'non-slip' systems, etc. Like other nematodes, filariae are equipped with sensory organs and a locomotor system, composed of the muscles and of the original osmoregulatory-excretory cell. Any change in one of these elements will result in the destruction of the filaria, at some stage of its development. In the vertebrate host, the intravascular stages will no longer be able to resist being carried passively towards the organs of destruction such as the lymph nodes or the lungs. 相似文献
