Diabetes mellitus is a chronic metabolic disorder, resulting in high morbidity and mortality worldwide. Neuropathy, ocular diseases, kidney diseases, and cardiovascular disease are common complications of diabetes that threats the patient’s life. Chrysin (CH), a natural flavonoid, has several pharmacological effects, including antioxidant, antiapoptotic, anti-inflammatory, and anticancer. Strong scientific documents indicated that chrysin may be effective for preventing and treating complications of diabetes in experimental models. The present study was designed to review the association between chrysin administration and diabetes complications by focusing on the possible molecular pathway. The findings indicate that chrysin has protective effects against diabetes outcomes by modulating oxidative stress, inflammation, and apoptosis in animal models. However, more clinical trial study should be done to clear the protective effects of chrysin in diabetic patients.  相似文献

HIV-1-specific CD4+ and CD8+ T lymphocytes are important for HIV-1 replication control. F4/AS01 consists of F4 recombinant fusion protein (containing clade B Gag/p24, Pol/RT, Nef and Gag/p17) formulated in AS01 Adjuvant System, and was shown to induce F4-specific polyfunctional CD4+ T-cell responses in humans. While replication-incompetent recombinant HIV-1/SIV antigen-expressing human adenoviral vectors can elicit high-frequency antigen-specific CD8+ T-cell responses, their use is hampered by widespread pre-existing immunity to human serotypes. Non-human adenovirus serotypes associated with lower prevalence may offer an alternative strategy. We evaluated the immunogenicity of AdC7-GRN (‘A’), a recombinant chimpanzee adenovirus type 7 vector expressing clade B Gag, RT and Nef, and F4/AS01 (‘P’), when delivered intramuscularly in homologous (PP or AA) and heterologous (AAPP or PPAA) prime-boost regimens, in macaques and mice. Vaccine-induced HIV-1-antigen-specific T cells in peripheral blood (macaques), liver, spleen, and intestinal and genital mucosa (mice) were characterized by intracellular cytokine staining. Vaccine-specific IgG antibodies (macaques) were detected using ELISA. In macaques, only the heterologous prime-boost regimens induced polyfunctional, persistent and balanced CD4+ and CD8+ T-cell responses specific to each HIV-1 vaccine antigen. AdC7-GRN priming increased the polyfunctionality of F4/AS01-induced CD4+ T cells. Approximately 50% of AdC7-GRN-induced memory CD8+ T cells exhibited an effector-memory phenotype. HIV-1-specific antibodies were detected with each regimen. In mice, antigen-specific CD4+ and CD8+ T-cell responses were detected in the mucosal and systemic anatomical compartments assessed. When administered in heterologous prime-boost regimens, AdC7-GRN and F4/AS01 candidate vaccines acted complementarily in inducing potent and persistent peripheral blood HIV-1-specific CD4+ and CD8+ T-cell responses and antibodies in macaques. Besides, adenoviral vector priming modulated the cytokine-expression profile of the protein-induced CD4+ T cells. Each regimen induced HIV-1-specific T-cell responses in systemic/local tissues in mice. This suggests that prime-boost regimens combining adjuvanted protein and low-seroprevalent chimpanzee adenoviral vectors represent an attractive vaccination strategy for clinical evaluation.  相似文献

Background

Domestic goats (Capra hircus) have been selected to play an essential role in agricultural production systems, since being domesticated from their wild progenitor, bezoar (Capra aegagrus). A detailed understanding of the genetic consequences imparted by the domestication process remains a key goal of evolutionary genomics.

Results

We constructed the reference genome of bezoar and sequenced representative breeds of domestic goats to search for genomic changes that likely have accompanied goat domestication and breed formation. Thirteen copy number variation genes associated with coat color were identified in domestic goats, among which ASIP gene duplication contributes to the generation of light coat-color phenotype in domestic goats. Analysis of rapidly evolving genes identified genic changes underlying behavior-related traits, immune response and production-related traits.

Conclusion

Based on the comparison studies of copy number variation genes and rapidly evolving genes between wild and domestic goat, our findings and methodology shed light on the genetic mechanism of animal domestication and will facilitate future goat breeding.

In the research, the single-and dual effects of phosphate-solubilizing bacteria (PSB) (B0, Pseudomonas sp. FA1, and Bacillus simplex UT1) and silicon (Si) (0, 150, 300, and 600 mg kg−1 used as silicic acid) on P uptake by sorghum (Sorghum bicolor L.) plant fertilized with soluble or insoluble P (rock phosphate—RP) were studied via a perlite-potted experiment. Moreover, the effects of various treatments on morphological (shoot and root dry weight), nutritional (the uptake of Si and K) and physiological parameters (activity of catalase, superoxide dismutase, and peroxidase enzymes) of this plant were also measured. When grown in RP-fertilized medium compared with those grown in soluble P-fertilized medium, both shoot biomass and root biomass of sorghum plants were noticeably diminished. The PSB strains and Si levels independently improved all the aforementioned parameters. Use of Si and PSB strains to sorghum plants grown in soluble P or insoluble P medium significantly augmented P use efficiency. Silicon not only augmented the uptake of P from sparingly soluble-P source (RP), but also augmented uptake of P from water-soluble P source. Both B. simplex UT1 and Pseudomonas sp. FA1 indicated a meaningful betterment in sorghum plant dry matter and uptake of P (and K and Si) under both soluble and insoluble P fertilization conditions with Pseudomonas sp. FA1 being more efficacious than B. simplex UT1. But, the dual use of the PSB with Si resulted in the greatest increase in sorghum plant P uptake and other measured growth indices. Application of 600 mg Si kg−1 and Pseudomonas sp. FA1 significantly augmented the P shoot concentration of sorghum plant fertilized with RP to an sufficient level (> 0.3%) in the range of P-fertilized sorghum plants. Therefore, in addition to PSB utilization, Si should be considered as soil amendment in agricultural soils inadequate in plant-available Si as a means of sustainable agriculture with respect to possible savings of scarce P resources.

Purpose

Winter road maintenance in the Nordic climate is demanding due to challenging weather conditions, high precipitation, and icy conditions. As a leading country in the transition to low-emission transport, Norway must work to reduce their emissions while providing a safe level of service through winter maintenance operations. This article investigates the environmental impacts of winter road maintenance (WRM) in Norway both today and under a climate change scenario predicted for 2050.

Methods

Life cycle assessment (LCA) is used to evaluate the environmental impact of the functional unit “average winter road maintenance in Norway on national and county roads per km.lane.” The ReCiPe (hierarchy) method was used to identify and categorize emissions related to WRM to show how different factors affect the system and to reveal hidden emissions hotspots. Real-time data from WRM vehicles were used to determine how fuel consumption is affected by gradient and weather. Producers and operators provided other relevant information on WRM vehicles. Official reports supplied information on deicer quantities used and the total distance driven by WRM vehicles in Norway.

Results and discussion

The quantity of deicer used is the main source of emissions contributing toward all impact categories. The effect of deicer is likely to be even higher in certain impact categories. The environmental impact of the deicer after application is not included. The representation of WRM in existing emissions data is limited despite the considerable amount of deicer applied and the long distances that WRM vehicles travel. The results document how energy use throughout the system is another important source of emissions. Various parameters, such as road gradient, vehicle properties, driver behavior, and weather, affect the fuel consumption of WRM vehicles, with weather being the most important of these.

Conclusions

Significant potential for emissions reductions from WRM was found, and WRM operations should be included in cold-climate road LCA studies. The environmental impacts of deicer application are especially high compared to the mechanical clearing of roads and contribute strongly to impact categories such as terrestrial, freshwater, and human toxicity and to the formation of particulate matter.

Across a large mountain area of the western Swiss Alps, we used occurrence data (presence‐only points) of bird species to find suitable modelling solutions and build reliable distribution maps to deal with biodiversity and conservation necessities of bird species at finer scales. We have performed a multi‐scale method of modelling, which uses distance, climatic, and focal variables at different scales (neighboring window sizes), to estimate the efficient scale of each environmental predictor and enhance our knowledge on how birds interact with their complex environment. To identify the best radius for each focal variable and the most efficient impact scale of each predictor, we have fitted univariate models per species. In the last step, the final set of variables were subsequently employed to build ensemble of small models (ESMs) at a fine spatial resolution of 100 m and generate species distribution maps as tools of conservation. We could build useful habitat suitability models for the three groups of species in the national red list. Our results indicate that, in general, the most important variables were in the group of bioclimatic variables including “Bio11” (Mean Temperature of Coldest Quarter), and “Bio 4” (Temperature Seasonality), then in the focal variables including “Forest”, “Orchard”, and “Agriculture area” as potential foraging, feeding and nesting sites. Our distribution maps are useful for identifying the most threatened species and their habitat and also for improving conservation effort to locate bird hotspots. It is a powerful strategy to improve the ecological understanding of the distribution of bird species in a dynamic heterogeneous environment.  相似文献

Background: Nowadays, there is an increasing interest in noninvasive methods to diagnose Helicobacter pylori infection. Indeed, they can profitably replace endoscopy in predicting the diagnosis. The stool antigen test for H. pylori is a noninvasive immunoassay to diagnose active infection with this bacterium in human fecal samples. The aim of this study was detection of alkyl hydroperoxide reductase protein (AhpC) antigen by immunoblotting in stool samples for diagnosis of H. pylori. Materials and Methods: Chromosomal DNA from H. pylori was isolated. AhpC gene was amplified by PCR, These amplicons were cloned into pTZ57R/T cloning vector then subcloned into pQE30 expression vector and overexpressed using isopropyl‐beta‐D‐thiogalactopyranoside in E. coli M15. AhpC protein was purified by affinity chromatography. Rabbits were immunized with the purified AhpC protein for the production of antibodies. To determine the accuracy of the test for diagnosing H. pylori infection from stool, we evaluated 84 patients (6–81 years old) using Western blot analysis by rabbit anti‐AhpC antibody. Positive rapid urease test on biopsy samples was considered as the gold standard. Results: AhpC gene was overexpressed, and AhpC protein was purified. Rabbit anti‐AhpC antibody produced after immunization with the purified AhpC protein. By immunoblotting, we detected AhpC protein in the positive stool samples. The test showed a 83.3% sensitivity (95% CI: 69.8–92.5%) and a 91.7% specificity (95% CI: 77.5–98.2). Among the children, the sensitivity was 88.2% (95% CI: 63.6–98.5) and the specificity was 100% (95% CI: 69.2–100); in adults, the sensitivity and specificity were 80.6% (95% CI: 62.5–92.5) and 88.5% (95% CI: 69.8–97.6), respectively. Conclusions: Using of AhpC antigen for diagnosis of H. pylori infection is a useful noninvasive method, accurate in adolescents and children, and can be used for the development of a stool antigen detection kit for H. pylori.  相似文献

Staphylococcus aureus is considered one of the most important food borne pathogens. A total of 111 isolates of S. aureus were cultured from raw milk samples during January 2009 to June 2009 from Tehran and Mashhad. The coagulase gene polymorphism and the prevalence of classical enterotoxin genes of S. aureus strains were determined by PCR-RFLP (restriction fragment length polymorphism) and Multiplex-PCR. Disk diffusion method was used to determine the susceptibility of isolates to antimicrobial agents as instructed by Clinical and Laboratory Standards Institute. Sixty-seven % of the isolates harboured one or more enterotoxin genes. The most prevalent gene was sec, found in 59 % of the isolates. Approximately 8% of the isolates were positive for sea, seb and sed genes. Only one isolate had see gene. The rate of coexistence of enterotoxin genes was 14%. All S. aureus isolates were susceptible to ciprofloxacin, gentamicin, imipenem, minocycline, oxacillin and vancomycin. They were resistant to ampicillin (64%), penicillin (56%), clindamycin (22%), tetracycline (22%), doxycycline (19%), teicoplanin (13%), rifampin (2%) and trimethoprim-sulfamethoxazole (2%). On the basis of coagulase gene analysis of 111 S. aureus isolates, the PCR products of 56 isolates were digested with Alu I that produced three distinct patterns. These data indicate the high prevalence of enterotoxigenic S. aureus in raw bovine milk in Tehran and Mashhad, and highlight the importance of proper quality control of dairy products for public health.  相似文献
