The activity described in this article engages students in learning about proper meat storage procedures by using the Internet. They apply their knowledge practically through creation of a pamphlet to be displayed in their kitchen at home and a follow-up report on their parents’ kitchen procedures; they may also participate in a classic food storage experiment. This activity is well suited for elementary school children in the second through fourth grades and promotes all basic STEM (science, technology, engineering, and math) concepts. Kitchen activities require all these concepts for successful and safe outcomes. The examination of food storage devices references engineering, and technology is used for the research and pamphlet creation. The section on bacteria introduces life sciences, and physical sciences come into play through the discussion of heat inducing change in the meats. The evolutionary interplay between myxoma virus (MYXV) and the European rabbit (Oryctolagus cuniculus) following release of the virus in Australia in 1950 as a biological control is a classic example of host-pathogen coevolution. We present a detailed genomic and phylogeographic analysis of 30 strains of MYXV, including the Australian progenitor strain Standard Laboratory Strain (SLS), 24 Australian viruses isolated from 1951 to 1999, and three isolates from the early radiation in Britain from 1954 and 1955. We show that in Australia MYXV has spread rapidly on a spatial scale, with multiple lineages cocirculating within individual localities, and that both highly virulent and attenuated viruses were still present in the field through the 1990s. In addition, the detection of closely related virus lineages at sites 1,000 km apart suggests that MYXV moves freely in geographic space, with mosquitoes, fleas, and rabbit migration all providing means of transport. Strikingly, despite multiple introductions, all modern viruses appear to be ultimately derived from the original introductions of SLS. The rapidity of MYXV evolution was also apparent at the genomic scale, with gene duplications documented in a number of viruses. Duplication of potential virulence genes may be important in increasing the expression of virulence proteins and provides the basis for the evolution of novel functions. Mutations leading to loss of open reading frames were surprisingly frequent and in some cases may explain attenuation, but no common mutations that correlated with virulence or attenuation were identified. 相似文献

Tentatively dated, the Plio‐/Pleistocene origin of the ancient Lake Ohrid on the Balkan Peninsula makes it the oldest ancient lake in Europe. Given the surface area of the lake and the adjusted endemicity rate, it may be also defined as the most diverse of all the ancient lakes in the world. From all the animal groups endemic to this lake, gammarids are amongst the most scarcely known in terms of their diversity and phylogenetic relationships. Partial DNA sequences of two mitochondrial genes, cytochrome oxidase subunit I (cox1) and 16S ribosomal RNA (16S rRNA) of eight known endemic Gammarus species from the Lake Ohrid valley were analysed. Phylogenetic analyses showed that endemic Gammarus species comprise an ancient species flock, with Gammarus sketi from the feeder springs being their sister taxon outside the lake. Amongst the species inhabiting the lake, Gammarus solidus and Gammarus salemaai are morphologically and molecularly well defined. By contrast, Gammarus ochridensis, Gammarus parechiniformis, Gammarus lychnidensis, and Gammarus stankokaramani revealed high discrepancy between morphological and genetic data. None of these morphospecies form a monophyletic clade and a significant degree of apparent gene flow occurs between them. This could be caused by incomplete lineage sorting and/or hybridization events. Two novel mtDNA lineages were found within the lake, possibly constituting two new species (Gammarus sp. 1 and Gammarus sp. 2). Molecular clock analysis showed that the split between G. sketi and the Gammarus species flock from the lake occurred approximately 5–7 Mya, whereas within the flock there were at least two intralacustrine radiations: one estimated at 2–3 Mya and the second at less than 1 Mya. Nucleotide polymorphism at 12 nuclear loci and two mitochondrial gene fragments was studied in three closely related pine species from the Pinus mugo complex in populations across the species distributional range in Europe. Despite large differences in the census sizes of the populations, high and similar levels of nucleotide diversity (θsil = ~0.013–0.017) were found at nuclear loci in the three pine species. More rapid decay of overall linkage disequilibrium (LD) and recombination to diversity ratio (ρ/θ) was observed across the species distributional range in P. mugo (ρ = 0.0369 ± 0.0028; ρ/θ = ~2.2) than in P. uncinata (ρ = 0.0054 ± 0.0011; ρ/θ = ~0.4) and P. uliginosa (ρ = 0.0051 ± 0.0010, ρ/θ = ~0.4). However, regional groups of P. mugo showed similar levels of LD and ρ/θ ratio to the other species. An excess of rare nucleotide variants was found in P. mugo at four loci, but, overall, the allelic frequency spectrum in the three species did not deviate significantly from neutrality (multilocus Tajima's D = ?0.681, D = ?0.118 and D = ?0.266, P > 0.05, respectively). Bayesian clustering methods showed no clear correspondence of clusters to species or geographical regions. Some differences between populations and species were found in a hierarchical analysis of molecular variance (AMOVA) and in the distribution of the mitochondrial DNA haplotypes, suggesting rather limited gene flow between the taxa and ongoing divergence. In the present study, we have demonstrated that membrane-free extracts of etiolated shoots of Phaseolus coccineus seedlings show tocopherol oxidase activity. For this reaction, presence of membrane lipids, such as lecithin and mixture of plant lipids was required. The rate of the reaction was the highest for α-tocopherol and decreased in the order α ? β > γ > δ tocopherols. In the case of α-tocopherol, the main oxidation product was α-tocopherolquinone, while for the other tocopherol homologues the dominant products were other derivatives. When the enzyme activity was measured in leaves, hypocotyls and roots of etiolated seedlings of P. coccineus, the oxidase activity was the highest in extracts of leaves and decreased towards the roots where no activity was detected. The effect of hydrogen peroxide and of different inhibitors on the reaction suggest that tocopherol oxidase does not belong to peroxidases or flavin oxidases but rather to multi-copper oxidases, such as polyphenol oxidases or laccases. On the other hand, catechol, the well-known substrate of polyphenol oxidases and laccases, was not oxidized by the enzyme, indicating a high substrate specificity of the tocopherol oxidase. 相似文献

A case‐study is presented of two Los Angeles‐based cross‐cultural coalitions, the Black‐Korean Alliance [BKA] and the Latino‐Black Roundtable [LBR]. An analysis of the development and eventual dissolution of these two coalitions is presented against the backdrop of the history of relations between the African‐American, Korean‐American and Latino communities of Los Angeles. The central issues leading to the breakup of the two coalitions were: lack of resources, internal power dynamics between coalition partners, nationalistic factions within the broader communities, cross‐cultural dynamics between coalition members, the use of a dialogue model to frame coalition activities, and the analysis of intergroup relations underlying the formation of the coalitions. In this study, the seedlings of two wheat cultivars were used: drought-resistant Chinese Spring (CS) and drought-susceptible (SQ1). Seedlings were subjected to osmotic stress in order to assess the differences in response to drought stress between resistant and susceptible genotype. The aim of the experiment was to evaluate the changes in physiological and biochemical characteristics and to establish the optimum osmotic stress level in which differences in drought resistance between the genotypes could be revealed. Plants were subjected to osmotic stress by supplementing the root medium with three concentrations of PEG 6000. Seedlings were grown for 21 days in control conditions and then the plants were subjected to osmotic stress for 7 days by supplementing the root medium with three concentrations of PEG 6000 (D1, D2, D3) applied in two steps: during the first 3 days of treatment ?0.50, ?0.75 and ?1.00 and next ?0.75, ?1.25 and ?1.5 MPa, respectively. Measurements of gas exchange parameters, chlorophyll content, height of seedlings, length of root, leaf and root water content, leaf osmotic potential, lipid peroxidation, and contents of soluble carbohydrates and proline were taken. The results highlighted statistically significant differences in most traits for treatment D2 and emphasized that these conditions were optimum for expressing differences in the responses to osmotic stress between SQ1 and CS wheat genotypes. The level of osmotic stress defined in this study as most suitable for differentiating drought resistance of wheat genotypes will be used in further research for genetic characterization of this trait in wheat through QTL analysis of mapping population of doubled haploid lines derived from CS and SQ1. 相似文献

Like other species of the genus Taxus, European yew trees contain taxanes, including paclitaxel (T) and its precursor 10-deacetylbaccatin III (10-DAB). Taxanes are one of the most effective anticancer drugs. This study was undertaken to describe the levels and patterns of taxane variation in the Sudetian region (SW Poland). Paclitaxel (T) and 10-deacetylbaccatin III (10-DAB) concentrations were analysed in five populations. Needles and twigs were analysed from 60 individuals (30 males and 30 females) in each population. In addition, morphometric measurements were taken in the populations to obtain light intensity coefficients (specific leaf area, SLA). High variability in the taxane contents at both intra and interpopulational levels was found. Nevertheless, females had a significantly higher taxane content compared to males. Because taxanes are carbon-based secondary metabolites, females have higher rate of gas exchange of females compared to males. This was probably an adaptation to greater reproductive effort incurred by females. In this regard, female individuals seem to be better for selecting elite cultivars with a higher taxane production. The relationship between light intensity and taxane content was not significant. Shading, important for optimizing crop production, should not reduce the concentration of taxanes. 相似文献

Measuring ethnicity in any society is a challenge. Given world immigration patterns, many countries face a growing dilemma in determining the cultural antecedents of their populations. A further complication is the reality that such determination occurs within the political and nationalistic settings where ethnic‐cultural groups may be potent forces in their own right. As societies mature and evolve, there is an increasing tendency for populations, especially those with many generations of residence in the country, to see themselves as ‘indigenous’ to the society in which they live. Canada is not alone in having to deal with the fluidity of the concept, ‘Canadian’, ‘American’, ‘Australian’, ‘Yugoslav’, and ‘Soviet’ are parallel concepts in other countries of multiple ethnic composition. Using 1991 National Census Test results, the article explores some of the parameters of the indigenous category ‘Canadian’. In particular, the location in Canada and mother tongue of respondents reporting ‘Canadian’ as the ethnic origin of their parents and grandparents or as their own ethnic identity are important indicators for this emerging ethnic category. 相似文献

The article examines the educational and employment experiences and aspirations of young Pakistani and Bangladeshi people living in Oldham, in Great Manchester. Many young people demonstrated high aspirations and high levels of participation, particularly in relation to the educational and occupational level of their parents. Explanatory factors include the cultural value of education among Asian groups, the desire by parents to ensure success for their children and the ethnic penalty which these young people incur in the labour market. However, not all Pakistani and Bangladeshi young people have these aspirations. Girls who wished to continue their education faced a more complex situation than boys; for girls it was important to avoid jeopardizing the family honour. Nonetheless, national statistics show a marked increase in the numbers of young Pakistani and Bangladeshi women in full-time undergraduate courses in recent years. Women with degree level qualifications showed considerable determination to combine paid employment with family life. 相似文献
