Herpetomonas roitmani , a trypanosomatid containing a bacterial endosymbiont, was cured by high doses of chloramphenicol. Wild-type and cured flagellates were compared as to polysaccharide composition, nutritional requirements and cellular differentiation. Fucose (18.0%), xylose (15.7%), mannose (38.9%), galactose (10.8%), glucose (16.4%) and inositol (< 1.0%) were identified as polysaccharide components of cured H. roitmani as assessed by gas-liquid chromatography. However, the wild-type strain displayed a markedly different sugar profile, in that xylose was absent and inositol preferentially synthesized, whereas the other monosaccharide components remained unchanged. Variations in nutritional pattern also occurred between both strains. The bacterial endosymbiont seems to provide the flagellates with nutritional factors, including usual amino acids, vitamins, purine (as adenine) and hemin. The process of cell differentiation was also significantly influenced by the endosymbiont. Opisthomastigote forms predominate (72.0%) in cured as compared with wild-type H. roitmani (37.0%).  相似文献

The amino acid composition of organic matrices associated with calcium carbonate precipitates in microbialites built by different Phormidium species (cyanobacteria) has been compared for samples recovered in lagoonal settings from two regions of the Southern Tropical Pacific separated by more than 4000 km: New Caledonia (Nouméa lagoon) and French Polynesia (Tikehau atoll). Calcium carbonate precipitation in these microbial structures was observed mainly in the interior of the domes and clearly separated from the photosynthetically active surface layer. This study focuses on the hydrolysable amino acid composition of the associated organic matrices that are typically rich in cysteine, leucine, alanine and arginine in New Caledonia, whereas they are particularly rich in dicarboxylic amino acids in French Polynesia. This striking difference is seemingly related to different environmental conditions that characterize the two reef settings. The high cysteine content suggests an origin from metallothioneins produced by the cyanobacteria and/or by epiphytic diatoms that were observed on the top layer, as the result of the input of metals from terrestrial origin in the Nouméa lagoon. In addition, we analysed the bulk organic matter of the photosynthetically active surface layer and of the interior of the domes. The former showed remarkable variations of amino acid composition throughout the year 2001, which may potentially reflect the impact of climatological events (e.g. cyclones) and/or a much stronger seasonality in New Caledonia than in French Polynesia. Although the mechanisms behind the differences remain elusive, our study clearly shows that environmental conditions can be reflected by amino acid compositions, particularly for the organic matrices associated with carbonate precipitates.  相似文献

The effect of some fresh‐cut carrot label attributes on consumer purchase intention was evaluated using conjoint analysis. The following three attributes were chosen and manipulated: price (high and low), packaging type (polyethylene bags and expanded polystyrene trays) and production process (organic with certification stamp, organic without certification stamp and no information). A complete factorial design was used, and 12 hypothetical packages of fresh‐cut carrot were developed and evaluated by 96 consumers based on their purchase intention. The consumers were divided into four groups with different opinions. Low price and organic with certification label were the favorite choices of the groups. Only one group preferred expanded polystyrene trays. No interactions were exhibited among the attributes in all groups.  相似文献

Abstract.  1. Recent research has addressed the function of herbivore-induced plant volatiles in attracting natural enemies of feeding herbivores. While many types of insect herbivory appear to elicit volatile responses, those triggered by gall insects have received little attention. Previous work indicates that at least one gall insect species induces changes in host-plant volatiles, but no other studies appear to have addressed whether gall insects trigger plant indirect defences.
2. The volatile responses of wheat to feeding by larvae of the Hessian fly Mayetiola destructor (Say) (Diptera: Cecidomyiidae) were studied to further explore indirect responses of plants to feeding by gall insects. This specialist gall midge species did not elicit a detectable volatile response from wheat plants, whereas a generalist caterpillar triggered volatile release. Moreover, Hessian fly feeding altered volatile responses to subsequent caterpillar herbivory.
3. These results suggest that Hessian fly larvae exert a degree of control over the defensive responses of their host plants and offer insight into plant-gall insect interactions. Also, the failure of Hessian fly larvae to elicit an indirect defensive response from their host plants may help explain why natural enemies, which often rely on induced volatile cues, fail to inflict significant mortality on M. destructor populations in the field.  相似文献

Young potato plants in pots exposed in the open near plots of potatoes for limited periods at intervals during the summer, became infested with large numbers of winged aphids only during warm, calm and dry weather. Although visited by aphids during May and June, when much of the spread of viruses occurred in nearby potato crops, few of the potted plants became infected. Most potted plants became infected in July when alate aphids were leaving neighbouring potato crops. Widely different proportions of the exposed plants became infected in different years; in two of the three years, many more plants were infected with virus Y than with leaf roll virus.  相似文献

Field experiments with Majestic potatoes were made over six years at Rothamsted to test the effects of varying date of planting and manuring on the yield of tubers and the incidence of the aphid-transmitted leaf roll and Y (rugose mosaic) viruses. Yield was increased by early planting, and by all the manures, especially dung. Early planting also usually increased the incidence of virus disease. Different manures had different effects on disease incidence; the average results from all comparisons showed the largest increase in incidence of both viruses from the use of dung; sulphate of ammonia increased the incidence of leaf roll, and muriate of potash that of rugose mosaic. Counts in two years showed that aphid populations were highest on the earlier planted potatoes, and were increased by dung, sulphate of ammonia and superphosphate, but were reduced by muriate of potash.  相似文献

Copulation and/or cohabitation with a pregnant female facilitate paternal behavior in male mice. However, their contribution to the rearing of the offspring is still not well understood. Our aims were to investigate the behavior of sires toward own or alien pups; the immediate consequences of the presence of fathers on the offspring and the behavior of the mother; and whether the exposure of juvenile males to newborn siblings, in an overlapping litters context, facilitates paternal behavior in C57BL/6 mice. We found that sires behaved paternally toward alien pups at both postpartum days 3 and 7; did not affect the behavior of the mother (e.g., licking and grooming, retrieval behavior, time in the nest, and crouching postures); and reduced the time offspring stayed alone in the nest. The exposure to newborn siblings did not promote paternal behavior in juvenile males. Therefore, sires are more paternal than usually described in the literature for laboratory mice, suggesting a facultative role in the rearing of the offspring. However, juvenile male mice, in contrast to juvenile females, could be adapted to leave the nest earlier without major contribution to the offspring.  相似文献
