The objective of this study was to develop single‐step PCR species‐specific primers that reliably discriminate four economically important Xiphinema species (X. brevicolle, X. elongatum, X. ifacolum and X. longicaudatum) and X. diffusum that is taxonomically very similar to X. brevicolle. Each species‐specific reverse primer was located in the ITS‐1 rDNA region and was used in combination with a universal forward primer located in the 18S rDNA gene. Primer reliability was confirmed by screening seven and 11 populations, respectively of X. diffusum and X. elongatum. Potential species‐specific primers were also identified for X. brevicolle, X. longicaudatum and X. ifacolum, however too few populations of these species were available to thoroughly assess their reliability. For all species‐specific primers, specificity was demonstrated by the absence of cross‐reactions with 14 non‐target Xiphinema species. Multiplex PCR was effective and reproducible for two (X. longicaudatum and X. ifacolum) or three (X. brevicolle, X. diffusum and X. elongatum) of the target nematode species, thus improving the applicability of the diagnostic primers.  相似文献

  • 1 To investigate the failure of DDT to interrupt malaria transmission in parts of the British Solomon Islands Protectorate, regular all-night man-biting catches of malaria vectors were made before and after DDT house spraying on San Cristobal Island.
  • 2 Changes were observed in the man-biting behaviour of Anopheles farauti. There was a reduction in the degree of entry into houses and a shifting of the times of peak biting. Whereas before spraying the indoor and outdoor biting cycles differed, after spraying there was no difference although both the indoor and outdoor cycles had altered.
  • 3 DDT was found to have a deterrent effect on An.farauti but this effect decreased with time.
  • 4 DDT also appears to eliminate a dominant indoor feeding fraction of the farauti population. Following this there can be an increase in an outdoor feeding fraction which can be responsible for a resumption of malaria transmission.
  • 5 Biting cycles obtained before spraying are also shown for An.koliensis and An.punctulatus.

  • 1 A total of 7063 specimens of Sphaeroceridae representing forty species were caught in four light traps, two in woodland and two in the open, at Rothamsted Experimental Station between December 1980 and December 1983.
  • 2 A complete list of species and the numbers of individuals of each is provided.
  • 3 The catches in the two traps in woodland were less diverse than the catches in the two traps in the open.
  • 4 Two-way analysis of variance demonstrated a significant difference in catch from trap to trap.
  • 5 When their similarities were compared, the four light trap catches formed two groups, corresponding with habitat type.
  • 6 The majority of the catch were Copromyza similis (Collin) (5555 specimens or 78.64% of the total). The species was most abundant in winter from mid-December to early February, although there were slight differences in times of maximum abundance between open and woodland habitats.

The numbers of honeyeaters present at particular sites in the Jarrah forest varied significantly from month to month, with peak abundance occurring between May and September. Numbers also varied from site to site, depending upon the major plant species present. Honeyeater abundance was not limited by arthropod availability, but in many instances was closely correlated with the availability of nectar, particularly that produced by Dryandra sessilis. Large honeyeaters, such as Anthochaera chrysoptera and Phylidonyris novaehollandiae, were generally most abundant at times and sites of greatest nectar production. Small honeyeaters, such as Acanthorhynchus superciliosis, were never abundant but were present for most of the year. The production of nectar between October and December was such that more honeyeaters could have been supported than were actually present. Low numbers at these times can be explained in terms of reduced foraging efficiency that would have resulted from more widely dispersed flowers, and the possible availability of more rewarding nectar resources at other sites.  相似文献

The malate extractedwas labelled to 35–40 per cent in the C-1 position andthe citric acid was labelled to 70–75 per cent in theC-6 position. A new species of Bellevalia Lapeyr. from the vilayet of Edirne in European Turkey is described which is hexaploid; 2 n = 24. This is a new chromosome number for Turkish material of the genus. Intact chloroplasts from young pea leaves were able to incorporate[3H]thymidine into DNA at relatively high rates (50 pmol mg–1chlorophyll h–1 or more), using light as the sole energysource. The intact plastids were also able to synthesize DNAin darkness, but only if ATP and MgCl2 (MgATP) were both present.The rates of MgATP-driven assimilation in darkness were equalto or greater than light-driven activity. Neither light nordithiothreitol pretreatments enhanced thymidine incorporationin darkness, suggesting that enzymes of chloroplast DNA (ctDNA)biosynthesis are not regulated via a thioredoxin-type system.Although exogenous nucleosides (other than [3H]thymidine) werenot an absolute requirement, dramatically elevated rates ofincorporation (over 300 pmol mg–1 chlorophyll h–1)were seen when adenosine, cytidine, guanosine and thymidinewere supplied in combination (500 mmol m–3 each). RadiolabelledDNA synthesized by the isolated chloroplasts was prepared usinga new heat extraction method. After digestion by restrictionendonucleases, ctDNA synthesized in organello was found to givetypical autoradiography patterns for chloroplast DNA. ExonucleaseIII studies suggested that 5% to 15% of the newly synthesizedDNA might be in a closed circular form. MgATP-driven synthesisin darkness was highly age-dependent. Chloroplasts from young(6 to 8-d-old) plants, or alternatively the youngest leavesof more mature plants, were 4–10 times more active thanthose from older tissues. Although these data do not establishconclusively that replication-type synthesis was occurring inthe isolated chloroplasts, they are consistent with this suggestion. Tipuloidea, the crane flies, are a diverse lineage of true flies (Insecta: Diptera) whose phylogenetic classification and taxonomy remain a challenge. Here we present the results of a quantitative phylogenetic analysis of Tipuloidea based on combined morphological characters (adult, larvae and pupae) and nuclear gene sequence data (28S rDNA and CAD). Forty‐five species, from 44 genera and subgenera, were sampled, representing the four putative families of Tipuloidea (Cylindrotomidae, Limoniidae, Pediciidae and Tipulidae sensu stricto). Analyses of individual datasets, although differing in overall topology, support the monophyly of several major lineages within Tipuloidea. Parsimony and Bayesian analyses using individual morphological and molecular datasets resulted in incongruent topologies. Increased resolution and tree support was obtained when both datasets (morphology and genes) were combined, in both combined evidence parsimony and Bayesian analyses, than when analysed separately. The recovered consensus phylogeny was not consistent with any previously proposed Tipuloidea classification, with previous importance assigned to character states shown here to represent losses and reversals seen as a major factor influencing erroneous classification. The results provided here, together with evidence from previous analyses, were used to append the Tipuloidea classification to supported evolutionary lineages. Tipuloidea is presented as two families: Pediciidae and Tipulidae. Pediciidae is recovered as the sister group to all remaining Tipuloidea. Our current phylogenetic hypothesis is not consistent with the existing subfamilial classification of the ‘Limoniidae’, which is paraphyletic with respect to a well‐supported Tipulinae + Cylindrotominae clade, whereas the three ‘limoniid’ subfamilies are para‐ or polyphyletic. The recognition of ‘Limoniidae’ as a valid monophyletic family is discouraged and the subfamilies of ‘Limoniidae’ are amended and placed within Tipulidae. A revised phylogenetic classification is proposed for the crane flies based on a synthesis of evidence from multiple genes and morphology.  相似文献

Boreal peatland ecosystems occupy about 3.5 million km2 of the earth's land surface and store between 250 and 455 Pg of carbon (C) as peat. While northern hemisphere boreal peatlands have functioned as net sinks for atmospheric C since the most recent deglaciation, natural and anthropogenic disturbances, and most importantly wildfire, may compromise peatland C sinks. To examine the effects of fire on local and regional C sink strength, we focused on a 12 000 km2 region near Wabasca, AB, Canada, where ombrotrophic Sphagnum‐dominated bogs cover 2280 km2 that burn with a fire return interval of 123±26 years. We characterized annual C accumulation along a chronosequence of 10 bog sites, spanning 1–102 years‐since‐fire (in 2002). Immediately after fire, bogs represent a net C source of 8.9±8.4 mol m−2 yr−1. At about 13 years after fire, bogs switch from net C sources to net C sinks, mainly because of recovery of the moss and shrub layers. Subsequently, black spruce biomass accumulation contributes to the net C sink, with fine root biomass accumulation peaking at 34 years after fire and aboveground biomass and coarse root accumulation peaking at 74 years after fire. The overall C sink strength peaks at 18.4 mol C m−2 yr−1 at 75 years after fire. As the tree biomass accumulation rate declines, the net C sink decreases to about 10 mol C m−2 yr−1 at 100 years‐since‐fire. We estimate that across the Wabasca study region, bogs currently represent a C sink of 14.7±5.1 Gmol yr−1. A decrease in the fire return interval to 61 years with no change in air temperature would convert the region's bogs to a net C source. An increase in nonwinter air temperature of 2 °C would decrease the regional C sink to 6.8±2.3 Gmol yr−1. Under scenarios of predicted climate change, the current C sink status of Alberta bogs is likely to diminish to the point where these peatlands become net sources of atmospheric CO2‐C.  相似文献
