Summary We have developed a procedure for determining the rates of mitotic recombination of an interrupted duplication created by integration of transforming plasmid sequences at the benA, beta-tubulin, locus of Aspergillus nidulans. Transformation of a strain carrying a benomyl-resistant benA allele with plasmid AIpGM4, which carries the wild-type benA allele and the pyr4 (orotidine-5-phosphate decarboxylase) gene of Neurospora crassa, creates an interrupted duplication with plasmid sequences flanked by two benA alleles, one wild type and one benomyl resdistant. Such transformants will not grow in the presence of high levels of benomyl. Mitotic recombination causes the loss of the wild-type benA allele or conversion of the wild-type to the mutant allele resulting in nuclei carrying only the benomylresistant allele. Conidia containing such nuclei can be selected on media with high benomyl allowing easy quantitation of mitotic recombination. We found that the rate of recombination giving rise to benomyl-resistant conidia was 4.6×10-4. Reciprocal recombination leading to benomyl-resistant conidia lacking plasmid sequences occurred at a rate of 2.0×10-4 and gene conversion leading to benomylresistant conidia occurred at a rate of 2.6×10-4. We selected for reciprocal recombination leading to loss of pyr4 sequences on 5-fluoro-orotic acid and used this selection for two-step gene replacement of a mutant benA allele with the wild-type allele.  相似文献

A crucial premise in many recent arguments against the moral permissibility of surrogate motherhood arrangements is the claim that a woman cannot autonomously consent to gestating and relinquishing a child to another couple, because she cannot be fully informed about what her future emotional responses will be to the foetus developing within her, and to the giving up of the newborn infant to its social parents. When conjoined with some moral principle about the justifiable limits on the ways others can be expected to exercise their autonomy on our behalf, this claim is often taken to establish that various forms of surrogate motherhood arrangements are morally wrong. In this paper I want to show that there is a serious non sequitur in this kind of argument. That is, I want to show that even if women cannot in fact have this kind of information about what their future emotional responses to pregnancy and relinquishment will be, nothing follows about the wrongness or otherwise of surrogacy. For, when we consider what counts as informed consent in the context of other important ventures with uncertain consequences, it becomes clear that informed consent does not require having this kind of information about one's future emotional states. In putting these arguments, I also hope to clarify some of the connections which might be thought to hold between informed consent and autonomous decision-making generally.  相似文献

Summary Rats were used in this study to determine the time course of conversion of muscle fiber types. The right or left gastrocnemius muscle was removed thereby causing an overload on the ipsilateral soleus and plantaris muscles. The contralateral limb served as a control. The type II to type I fiber conversion was followed histochemically in the soleus and plantaris muscles for one to six weeks following surgery. Muscle sections were stained for myofibrillar actomyosin ATPase and NADH tetrazolium reductase. The type I population in the soleus muscle was 99.3% six weeks after synergist removal. The plantaris muscle underwent a two fold increase in the percentage of type I fibers after six weeks. Transitional fibers were prominent in the plantaris muscle and reached their peak at 4% (P<0.05) of the total population, four weeks after surgery.This research was funded in part by grants from The Graduate School at Washington State University, and The Society of the Sigma Xi  相似文献

Relationships among the less derived genera in the subfamily Salmoninae were investigated by phylogenetic analysis of data from restriction maps of the ribosomal DNA using Osmerus mordax (rainbow smelt), Coregonus hoyi (bloater), and Thymattus arcticus (Arctic grayling) as outgroups. Analysis of the results suggests that Hucho h. taimen is more closely related to Brachymystax lenok (lenok) than to Hucho perryi (Sakhalin taimen) and that B. lenok and Hucho h. taimen (Eurasian taimen) form the most ancestral clade of the Salmoninae.  相似文献

Photosynthetic light‐response curves of the deep‐water Laminaria abyssalis Oliveira and of the intertidal L. digitata Lamoroux were determined and related to photoinhibition phenomena as monitored by oxygen evolution and photosystem II efficiency (FV/FM). L. abyssalis has half the pigment content, number of cells and plastids, and photosynthetic capacity per unit area compared with L. digitata. L. abyssalis showed a higher in vivo Chl a absorption coefficient and higher photosynthetic efficiency on a Chl a basis, although the two algae showed somewhat similar light‐response curves on a Chl a basis. Both species showed similar Chl a/Chl c and Chl a/fucoxanthin ratios, and similar dark respiration rates and light compensation points. In addition, they also showed similar convexities in their light‐response curves and no differences in their light saturation of FV/FM. Room temperature chlorophyll fluorescence induction measurements of fronds incubated in 3‐(3,4‐dichlorophenyl)‐1,1‐dimethylurea (DCMU) suggest that both species may have a similar PSII absorption cross section. Thus, L. abyssalis appears to optimize its light absorption at very low light intensities, not by increasing the pigment content, but by absorbing light more efficiently. However, L. abyssalis was more sensitive to photoinhibition than L. digitata and showed no recovery of FV/FM and O2 evolution after a photoinhibitory treatment, even with a subsequent exposure to 24 h of dim light. L. digitata, on the other hand, recovered its photosynthetic capacity within 6 h under dim light. These results suggest that photosynthetic light‐induction curves based on Chl a are not a good indicator of either the photosynthetic capacity or the sensitivity to photoinhibition when macroalgae of different species are being compared. Based on their light‐response and photoinhibition characteristics, we suggest that L. abyssalis, a deep‐water oceanic macroalgae, is an atypical shade alga whereas L. digitata has the properties of a sun alga.  相似文献

In order to develop a method for obtaining mitotic synchrony in aspergillus nidulans, we have characterized previously isolated heat-sensitive nim mutations that block the nuclear division cycle in interphase at restrictive temperature. After 3.5 h at restrictive temperature the mitotic index of a strain carrying one of these mutations, nimA5, was 0, but when this strain was subsequently shifted from restrictive to permissive temperature the mitotic index increased rapidly, reaching a maximum of 78 percent after 7.5 min. When this strain was examined electron-microscopically, mitotic spindles were absent at restrictive temperature. From these data we conclude that at restrictive temperature nimA5 blocks the nuclear division cycle at a point immediately preceding the initiation of chromosomal condensation and mitotic microtubule assembly, and upon shifting to permissive control over the initiation of microtubule assembly and chromosomal condensation in vivo through a simple temperature shift and, consequently, nimA5 should be a powerful tool for studying these processes. Electron-microscopic examination of spindles of material synchronized in this manner reveals that spindle formation, although very rapid, is gradual in the sense that spindle microtubule numbers increase as spindle formation proceeds.  相似文献

A method is described for identifying fiber types of skeletal muscle from several mammalian species on the basis of the sequential inactivation of myofibrillar actomyosin ATPase during acid preincubation. When this method is used in combination with the standard alkaline preincubation at least 5 types of fibers can be identified. Of these, 2 are type I fibers with those of the slow twitch soleus muscle being different from those that exist in mixed muscles. The 3 subtypes of type II fibers exist independent of their metabolic properties. The need for careful standardization of histochemical methods for the visualization of myofibrillar actomyosin ATPase and the implication of the existence of different fiber types in apparently homogeneous muscle for the preparation of antibodies used for immunocytochemical methods of fiber identification are discussed.  相似文献

The effect of prolonged maternal hyperglycaemia on fetal plasma glucose and insulin concentrations was investigated in eight normal and nine diabetic patients. It was found that under fasting conditions the fetal glucose concentration in gestational diabetic pregnancies tended to be lower than in normal pregnancies. Insulin measurements suggested that this may be due to fetal hyperinsulinism in the diabetic group. During glucose infusion, regardless of the degree of maternal hyperglycaemia, the fetal glucose concentration was limited in 12 out of 13 cases to less than 200 mg/100 ml, with only small differences between normal and diabetic pregnancies. It is proposed that the placenta prevents unlimited transport of glucose to the fetus; yet in diabetic pregnancies a sequence of increased maternal-fetal glucose transport, fetal hyperinsulinism, and fetal hypoglycaemia may contribute to the observed perinatal mortality.  相似文献
