High densities of introduced herbivores can damage sensitive ecosystems, increase the risk of extinction of native biota, and host and spread disease. An essential step in managing large ‘feral’ animal populations is to quantify how they use habitats so that management interventions, such as culling, can be targeted to reduce densities and to minimize migration into areas from which animals have been removed. An effective method to quantify animal movements is by measuring landscape‐scale genetic population structure. We describe the genetic population structure of one of Australia's more destructive introduced mammals – the Asian swamp buffalo (Bubalus bubalis). We collected 524 skin samples from buffalo across their range in the Northern Territory of Australia. Allelic diversity in the Northern Territory population was low compared to those reported from populations in their native Asian habitats. The Australian population is tentatively made of three subpopulations; Melville Island, Eastern Arnhem and Central‐Western Arnhem populations. The Melville Island population is represented by a single cluster, while the Eastern Arnhem population has three clusters and the Central‐Western Arnhem population seven clusters. We found some support for isolation by distance across all the sampled populations, but little evidence for this relationship when comparing the two well‐mixed mainland meta‐populations. Despite their small founder populations and limited genetic variation, the persistence of buffalo in Australia has likely been aided by release from high predation, parasitism and disease typical of their native habitats. 相似文献

1. To assess changes in stoichiometric constraints on stream benthos, we measured elemental composition of epilithon and benthic macroinvertebrates in intrinsically P‐limited mountain rivers, upstream and downstream of low‐level anthropogenic nutrient enrichment by effluents of municipal wastewater treatment plants. 2. While there was a broad range in the elemental composition of epilithon (C : P ratios of 200–16 500, C : N ratios of 8–280, N : P ratios of 8–535) and heptageniid mayfly scrapers (C : P ratios of 125–300, C : N ratios of 5.1–7.2, N : P ratios of 20–60), the average C : P ratio of epilithon was 10‐fold lower and the average C : N ratio twofold lower at more nutrient‐rich downstream sites. Nutrient ratios in benthic macroinvertebrates were lower than in epilithon and varied little between relatively nutrient‐poor and nutrient‐rich sites. 3. We modified the existing definition of producer‐consumer elemental imbalance to allow for variation in consumer nutrient content. We defined this ‘non‐homeostatic’ imbalance as the perpendicular distance between the producer and consumer C : P, C : N, or N : P ratios, and the 1 : 1 line. 4. At P‐limited sites, the estimated mayfly N : P recycling ratio was higher than the N : P ratio in epilithon, suggesting nutrient recycling by consumers could accentuate P‐limitation of epilithon. 5. Measuring the degree of producer–consumer nutrient imbalance may be important in predicting the magnitude of effects from nutrient enrichment and can help elucidate the causes and consequences of ecological patterns and processes in rivers. 相似文献

ABSTRACT As human development continues in coastal areas, shoreline properties adjacent to expansive tidal marsh habitat are increasingly used for access to coastal waterways via long piers (>30 m) over marsh habitat. These tidal wetlands provide breeding and foraging habitat for many marsh birds, which may be affected by the human disturbance associated with long piers. Our objectives were to determine the effect of long piers over vegetated tidal marshes on the relative abundance and species richness of marsh birds. We completed combined passive and callback surveys in tidal marsh habitat at 22 sites with long piers and 24 sites without long piers, May-July 2005–2006 in Worcester County, Maryland, USA. Pier sites had lower relative abundance and species richness of obligate marsh birds than nonpier sites. Pier sites had a greater relative abundance of gulls, terns, herons, and egrets than nonpier sites. Pier sites had fewer species of herons and egrets than at nonpier sites. The presence of long piers had no affect on facultative marsh birds. Long pier density was negatively related to obligate marsh bird relative abundance and species richness, and facultative marsh bird species richness, whereas it was positively related to the relative abundance and species richness of gulls and terns. Herons and egrets relative abundance and species richness were not related to long pier density. Obligate marsh birds were negatively affected by long piers, whereas herons, egrets, gulls, and terns appeared to benefit from perching opportunities. Based on the negative effects of long piers on obligate marsh birds, management should focus on reducing the presence and density of long piers in tidal marshes by requiring the removal of existing long piers, or reducing or eliminating permits for construction of new long piers. 相似文献

Antigen/antibody complexes can efficiently target antigen presenting cells to allow stimulation of the cellular immune response. Due to the difficulty of manufacture and their inherent instability complexes have proved inefficient cancer vaccines. However, anti-idiotypic antibodies mimicking antigens have been shown to stimulate both antibody and T cell responses. The latter are due to T cell mimotopes expressed within the complementarity-determining regions (CDRs) of antibodies that are efficiently presented to dendritic cells in vivo. Based on this observation we have designed a DNA vaccine platform called ImmunoBody™, where cytotoxic T lymphocyte (CTL) and helper T cell epitopes replace CDR regions within the framework of a human IgG1 antibody. The ImmunoBody™ expression system has a number of design features which allow for rapid production of a wide range of vaccines. The CDR regions of the heavy and light chain have been engineered to contain unique restriction endonuclease sites, which can be easily opened, and oligonucleotides encoding the T cell epitopes inserted. The variable and constant regions of the ImmunoBody™ are also flanked by restriction sites, which permit easy exchange of other IgG subtypes. Abstract An unusually high diversity of macropods inhabit the rocky areas in the monsoon tropics of the Northern Territory, Australia, yet the mechanisms that allow their niche separation are not clear. Previous studies suggest that the nabarlek, Petrogale concinna, may have a more grazing diet than the short‐eared rock‐wallaby, Petrogale brachyotis, with whom it coexists. Thus, diet may be an important mechanism of niche separation between these species. We examined the diet of the four sympatric species (the black wallaroo Macropus bernardus, common wallaroo Macropus robustus, P. brachyotis and P. concinna) to determine whether there are differences in the dominant plant groups eaten by the species across the landscape and with season. Diets were determined with a macroscopic analysis of the seed and fruit content of scats and an analysis of the 12C to 13C isotope ratios of scats using mass spectrometry. In the dry season the rock‐wallaby species predominantly consumed browse and/or forbs, and the larger wallaroos predominantly consumed grass. However, there was large variation across the landscape in the dry season diets of P. brachyotis, M. bernardus and M. robustus; including high proportions of grass eaten at some sites and high proportions of browse at other sites. In the wet season, greater proportions of grass were eaten by P. brachyotis and M. bernardus than in the dry season. Generally, there was little evidence to support the previous suggestion that P. concinna is more of a grazer than P. brachyotis, but there was some evidence than M. bernardus consumes greater amounts of browse and/or forbs than M. robustus. 相似文献
