Microtubules influence morphogenesis by forming distinct geometrical arrays in the cell cortex, which in turn affect the deposition of cellulose microfibrils. Although many chemical and physical factors affect microtubule orientation, it is unclear how cortical microtubules in elongating cells maintain their ordered transverse arrays and how they reorganize into new geometries. To visualize these reorientations in living cells, we constructed a microtubule reporter gene by fusing the microtubule binding domain of the mammalian microtubule-associated protein 4 (MAP4) gene with the green fluorescent protein (GFP) gene, and transient expression of the recombinant protein in epidermal cells of fava bean was induced. The reporter protein decorates microtubules in vivo and binds to microtubules in vitro. Confocal microscopy and time-course analysis of labeled cortical arrays along the outer epidermal wall revealed the lengthening, shortening, and movement of microtubules; localized microtubule reorientations; and global microtubule reorganizations. The global microtubule orientation in some cells fluctuates about the transverse axis and may be a result of a cyclic self-correcting mechanism to maintain a net transverse orientation during cellular elongation. 相似文献

Background
Selfish genetic elements that distort the sex ratio are found widely. Notwithstanding the number of records of sex ratio distorters, their incidence is poorly understood. Two factors can prevent a sex ratio distorter from invading: inability of the sex ratio distorter to function (failure of mechanism or transmission), and lack of drive if they do function (inappropriate ecology for invasion). There has been no test to date on factors causing variation in the incidence of sex ratio distorting cytoplasmic bacteria. We therefore examined whether absence of the male-killing Wolbachia infection in D. bifasciata in Hokkaido island of Japan, in contrast to the presence of infection on the proximal island of Honshu, was associated with failure of the infection to function properly on the Hokkaido genetic background. 相似文献34.
Since the initial work of Jukes and Cantor (1969), a number of procedures
have been developed to estimate the expected number of nucleotide
substitutions corresponding to a given observed level of nucleotide
differentiation assuming particular evolutionary models. Unlike the
proportion of different sites, the expected number of substitutions that
would have occurred grows linearly with time and therefore has had great
appeal as an evolutionary distance. Recently, however, a number of authors
have tried to develop improved statistical approaches for generating and
evaluating evolutionary distances (Schoniger and von Haeseler 1993;
Goldstein and Polock 1994; Tajima and Takezaki 1994). These studies clearly
show that the estimated number of nucleotide substitutions is generally not
the best estimator for use in reconstruction of phylogenetic relationships.
The reason for this is that there is often a large error associated with
the estimation of this number. Therefore, even though its expectation is
correct (i.e., on average the expected number of substitutions is
proportional to time- -but see Tajima 1993), it is not expected to be as
useful as estimators designed to have a lower variance.

Ribosomal protein S14 genes (RPS14) in eukaryotic species from protozoa to
primates exhibit dramatically different intron-exon structures yet share
homologous polypeptide-coding sequences. To recognize common features of
RPS14 gene architectures in closely related mammalian species and to
evaluate similarities in their noncoding DNA sequences, we isolated the
intron-containing S14 locus from Chinese hamster ovary (CHO) cell DNA by
using a PCR strategy and compared it with human RPS14. We found that rodent
and primate S14 genes are composed of identical protein-coding exons
interrupted by introns at four conserved DNA sites. However, the structures
of corresponding CHO and human RPS14 introns differ significantly.
Nonetheless, individual intron splice donor, splice acceptor, and upstream
flanking motifs have been conserved within mammalian S14 homologues as well
as within RPS14 gene fragments PCR amplified from other vertebrate genera
(birds and bony fish). Our data indicate that noncoding, intronic DNA
sequences within highly conserved, single-copy ribosomal protein genes are
useful molecular landmarks for phylogenetic analysis of closely related
vertebrate species.

Abstract Vertebrates living in dry or salty habitats spend significant amounts of time and energy maintaining their hydro‐mineral balance; any opportunity to drink fresh water should reduce such expenses. However, to find fresh water, individuals are generally forced to leave the safe and buffered conditions that prevail in their refuges. Therefore, they must face harsh environmental conditions and increased predation risks. We examined this trade‐off in the field, using a very large dataset gathered on amphibious sea snakes that confront periodic droughts. The study was carried out on different islets in the lagoon of New Caledonia. As expected, sea kraits remain concealed most of the time under rocks or within burrows during dry weather. They select precise time windows to move rapidly between the sea and the land, thereby minimizing the time spent in the open. Rainfall triggered massive disruptions of this rhythm: many snakes quickly forsook their shelter to drink in the open, and remained almost motionless until satiety. Interestingly, they adopted specific and unusual postures to drink. They perched on the top of rocks that were benefiting from running and less salty water during downpours. In a diverse array of avian and mammalian species, experimental manipulations of clutch size have tested the hypothesis that natural selection should adjust numbers of neonates produced so as to maximize the number of viable offspring at the end of the period of parental care. Reptiles have not been studied in this respect, probably because they rarely display parental care. However, females of all python species brood their eggs until hatching, but they do not care for their neonates. This feature provides a straightforward way to experimentally increase or reduce clutch size to see whether the mean clutch size observed in nature does indeed maximize hatching success and/or optimize offspring phenotypes. Eggs were removed or added to newly laid clutches of Ball Pythons ( Python regius ) in tropical Africa (nine control clutches, eight with 50% more eggs added, six with 42% of eggs removed). All clutches were brooded by females throughout the 2-month incubation period. Experimental manipulation of clutch-size did not significantly affect the phenotypes (morphology, locomotor ability) of hatchlings, but eggs in 'enlarged' clutches hatched later, and embryos were more likely to die before hatching. This mortality was due to desiccation of the eggs, with females being unable to cover 'enlarged' clutches sufficiently to retard water loss. Our results support the notion of an optimal clutch size, driven by limitations on parental ability to care for the offspring. However, the proximate mechanisms that generate this optimum value differ from those previously described in other kinds of animals. © 2003 The Linnean Society of London .
