ABSTRACT. Leishmania donovani promastigotes were collected, washed, resuspended in buffer, and assayed for sucrase activity. No activity was observed in the intact washed cells, but activity was measurable when the cells were permeabilized with Triton X-100. Intracellular sucrase activity was highest in promastigotes grown at pH 7.4, somewhat lower in promastigotes grown at pH 5.5, and significantly lower in "amastigotes" grown at pH 5.5. No trehalase, lactase, or maltase activities were observed. Assay of the medium in which the cells had grown showed that most the sucrase activity was extracellular, i.e. was secreted into the medium during growth. 相似文献

The hypothesis that heterosis in biomass production of sorghum(Sorghum bicolor L. Moench) may be ascribed to stability incarbon exchange rate (CER) over a wide range of environmentalconditions was evaluated. This hypothesis was based on previousresults from detached leaves that hybrids sustained greaterCER over a wider temperture range than their parents. Two grain sorghum hybrids (ATx378/RTx430 and ATx378/RTx434)and their parental lines were grown in the greenhouse in a gradientof ambient temperatures under two water regimes (well-irrigatedand drought up to heading). Plant water-use (estimated by weighingpots), leaf area, leaf gas exchange, grain yield, and above-groundbiomass were determined. Significant heterosis was found for biomass, grain yield perplant, and grain number per panicle. No heterosis occurred forharvest index, indicating that heterosis in grain yield wasdue to heterosis in biomass. Neither growth duration nor leafarea could explain heterosis in biomass. CER and stomatal conductancefor hybrid ATx378/RTx430 in the controls were greater than forboth its parents at leaf temperatures above 38 °C. This,however, was not observed in the other hybrid which was lessheterotic for biomass and grain yield in the controls. WhenCER data were subjected to a stability analysis by joint linearregression, the two hybrids had greater CER than their respectiveparents especially under conditions favouring high CER. ABSTRACT. Leishmania major promastigotes were grown to late-log phase and washed and resuspended in an isosmotic buffer. When osmolality was suddenly decreased by 50%, the cells rapidly became shorter and increased in width. Cell volume, calculated assuming a prolate-ellipsoidal shape, increased 1.4 times after 1 min. Over the next several minutes, the average length and width returned to control values while the volume returned to baseline, indicating the ability to regulate volume. Concomitantly with the swelling, large amounts of alanine and other ninhydrin-positive substances were released. All of the alanine pool was released within 1 min after reduction of the osmolality by 66%. Cells pre-loaded with [14 C]-aminoisobutyric acid also released it very rapidly upon hypo-osmotic stress. Release of ninhydrin-positive substances resulted from decreased osmolality rather than changes in ionic composition. The same results were obtained if osmolality was decreased by reducing only the NaCl content of the buffer instead of diluting it with water, and mannitol could substitute for the NaCl. Promastigotes were able to grow well over several days in media as low as 154 mOsm/kg. Gene expression levels were quantified after ultraviolet radiation treatment in the parental inbred lines of the maize mapping (IBM) population. This allows us to take advantage of natural variation between maize lines to analyse variation in gene expression. Using a statistically sound split‐plot experiment cDNAs were identified with differently regulated expression in B73 and Mo17 after UV treatment. Fewer genes were down‐regulated in B73; this global strain difference in the number of genes up‐ and down‐regulated does not appear to reflect general hybridization differences. Contrary to our expectation, there was a higher proportion of highly expressed genes (based on EST recovery) that were differently expressed by UV between lines. Genes affected by UV (but not significantly different between B73 and Mo17) include gene types proposed to function in UV acclimation and adaptation based on experiments in other species or other experiments in maize. 1. The influence of spatial structure on population dynamics within river–stream networks is poorly understood. Utilizing spatially explicit analyses of temporal genetic variance, we tested whether persistence of central stonerollers (Campostoma anomalum) reflects differences in habitat quality and location within a highly modified urban catchment in southwestern Ohio, U.S.A. 2. Estimates of genetic diversity did not vary with habitat quality. Nevertheless, evidence of weak but temporally stable genetic structure, location‐dependent effective population sizes and rates of immigration among sites, together suggest that persistence of central stonerollers within the catchment may be attributable to source–sink dynamics driven by habitat heterogeneity. 3. Under this scenario, migrant‐pool colonization from areas of relatively high habitat quality in the upper catchment sustains the presence of central stonerollers at degraded sites in the main stem and dampens population subdivision within the catchment. However, because intact habitat is restricted to the upper portion of the catchment, it is not possible to preclude net downstream dispersal as a mechanism contributing to source–sink dynamics. The slight genetic structure that persists appears to reflect weak isolation by distance diminished by high rates of immigration. 4. This study suggests that without a systems perspective of the conditions that sustain populations in degraded waterways, environmental assessments may underestimate levels of impairment. Conservation and management of stream fishes could be improved by maintaining habitat in areas that are net exporters of migrants or by remediation of impaired habitat. 相似文献
