The three-dimensional capture organs of the nematodetrapping fungus Arthrobotrys oligospora are presented in scanning electron micrographs. The fungus was grown directly on agar substrates or on millipore filters or glass cover slips placed on agar substrates. To avoid shrinkage of the soft-walled mycelial material, a fixation in OsO4-vapour was performed, before freeze-drying and coating with Au-Pd alloy. When capture organs were prepared in this way, it was possible to visualize them in their erect position in the scanning electron microscope. Young capture organs, both spontaneously formed and nematode-induced, remained standing better than old ones. An adhesive substance on the capture organs, with the aid of which nematodes are trapped, seems to be visible in these pictures.  相似文献

Ongoing genetic transfer from mitochondria and plastids into the nucleus is a well‐documented fact. While in metazoan molecular ecology the need for surveillance against pseudogene‐mediated artefacts when analysing mtDNA sequences is commonly accepted, no comparable measurements have been established for plastid‐based studies. We highlight the impact and management of nuclear mitochondrial insertions, argue that nuclear plastid sequences represent an underestimated but major factor in plant molecular ecology, and discuss potential avenues of remedy in chloroplast studies.  相似文献

Noccaea camlikensis sp. nov. (Brassicaceae) is described and illustrated from a localized serpentine range in southern Turkey. Its close relationship to N. cariensis , from which it differs in leaf and carpological characters, is discussed. The protologue of this fairly unknown species can now be amended to cover better the morphological variability of this taxon. A comparative study of seed-coat anatomy advocates its move from Thlaspi to Noccaea ; the relevant combination N. cariensis (Carlström) Parolly, Nordt & Aytaç is made. Growing seasons are getting longer, a phenomenon partially explained by increasing global temperatures. Recent reports suggest that a strong correlation exists between warming and advances in spring phenology but that a weaker correlation is evident between warming and autumnal events implying that other factors may be influencing the timing of autumnal phenology. Using freely rooted, field‐grown Populus in two Free Air CO2 Enrichment Experiments (AspenFACE and PopFACE), we present evidence from two continents and over 2 years that increasing atmospheric CO2 acts directly to delay autumnal leaf coloration and leaf fall. In an atmosphere enriched in CO2 (by ~45% of the current atmospheric concentration to 550 ppm) the end of season decline in canopy normalized difference vegetation index (NDVI) – a commonly used global index for vegetation greenness – was significantly delayed, indicating a greener autumnal canopy, relative to that in ambient CO2. This was supported by a significant delay in the decline of autumnal canopy leaf area index in elevated as compared with ambient CO2, and a significantly smaller decline in end of season leaf chlorophyll content. Leaf level photosynthetic activity and carbon uptake in elevated CO2 during the senescence period was also enhanced compared with ambient CO2. The findings reveal a direct effect of rising atmospheric CO2, independent of temperature in delaying autumnal senescence for Populus, an important deciduous forest tree with implications for forest productivity and adaptation to a future high CO2 world.  相似文献

The xylem hydraulic efficiency and safety is usually measured on mechanically unstressed samples, although trees may be exposed to combined hydraulic and mechanical stress in the field. We analysed changes in hydraulic conductivity and vulnerability to drought‐induced embolism during static bending of Picea abies and Pinus sylvestris branches as well as the effect of dynamic bending on the vulnerability. We hypothesized this mechanical stress to substantially impair xylem hydraulics. Intense static bending caused an only small decrease in hydraulic conductance (?19.5 ± 2.4% in P. abies) but no shift in vulnerability thresholds. Dynamic bending caused a 0.4 and 0.8 MPa decrease of the water potential at 50 and 88% loss of conductivity in P. sylvestris, but did not affect vulnerability thresholds in P. abies. With respect to applied extreme bending radii, effects on plant hydraulics were surprisingly small and are thus probably of minor eco‐physiological importance. More importantly, results indicate that available xylem hydraulic analyses (of conifers) sufficiently reflect plant hydraulics under field conditions.  相似文献

Amino acid transporters in plants are crucial for distributing amino acids between plant organs and cellular compartments. The H+‐coupled plasma membrane transporter CAT1 (cationic amino acid transporter 1) facilitates the high‐affinity uptake of basic amino acids. The uptake of lysine (Lys) via the roots was not altered in loss‐of‐function mutants, in accordance with the minor expression of CAT1 in roots, but plants ectopically overexpressing CAT1 incorporated Lys at higher rates. Exogenous Lys inhibited the primary root of Arabidopsis, whereas lateral roots were stimulated. These effects were augmented by the presence or absence of CAT1. Furthermore, the total biomass of soil‐grown plants ectopically overexpressing CAT1 was reduced and the time to flowering was accelerated. These effects were accompanied by only minor changes in the overall amino acid profile. Interestingly, CAT1 belongs to a specific small cluster of nitrogen‐containing metabolite transporter genes that are rapidly up‐regulated upon infection with Pseudomonas syringae and that may participate in the systemic response of plants to pathogen attack. The overexpression of CAT1 indeed enhanced the resistance to the hemibiotrophic bacterial pathogen P. syringae via a constitutively activated salicylic acid (SA) pathway, which is consistent with the developmental defects and the resistance phenotype.  相似文献

1. The growth pattern of Namibian fairy circles was examined in relation to environmental, termite, and plant variables to provide support for the sand termite (Psammotermes allocerus Silvestri) hypothesis of circle origin. 2. New and young circles were associated with the highest number of sand termites and their foraging grass sheetings which were both considerably lower in mature and senescent circles. 3. Circles increased in size with age, and apart from the senescent stage had higher soil moisture levels than the matrix. 4. In laboratory trials sand termites browsed seedling roots, negatively impacting root and leaf number. 5. This provides a potential mechanism for circle formation through central‐based foraging by P. allocerus which eliminates Stipagrostis grass clumps around its nest system. 6. The resulting bare disc is postulated to be maintained through a combination of a depleted seed bank, termite herbivory on seedlings, and excavations by pugnacious ants (Anoplolepis steingroeveri Forel).  相似文献

Ex situ cultivation in botanic gardens could be one possibility to preserve plant species diversity and genetic variation. However, old ex situ populations are often sparsely documented. We were able to retrieve three different ex situ populations and their source in situ populations of the endangered plant species Silene otites after 20–36 years of isolation. Furthermore, three additional wild populations were included in the analysis. Population genetic diversity and differentiation were analysed using AFLP markers. Genetic variation in the ex situ populations was lower than the variation found in the in situ populations. Strong differentiation (FST = 0.21–0.36) between corresponding in situ and ex situ populations was observed. Bayesian clustering approach also showed a distinct genetic separation between in situ and ex situ populations. The high genetic differentiation and loss of genetic diversity during spatial and temporal isolation in the ex situ populations can be attributable to small population sizes and unconscious selection during cultivation. Therefore, adequate sampling prior to ex situ cultivation and large effective population sizes are important to preserve genetic diversity. Near‐natural cultivation allowing for generation overlap and interspecific competition without artificial selection is recommended as being best for the maintenance of the genetic constitution. © 2011 The Linnean Society of London, Botanical Journal of the Linnean Society, 2011, ??, ??–??.  相似文献
