Human plasma low density lipoprotein (LDL) that had been rendered polycationic by coupling with N, N-dimethyl-1, 3-propanediamine (DMPA) was shown by electron microscopy to bind in clusters to the surface of human fibroblasts. The clusters resembled those formed by polycationic ferritin (DMPA-feritin), a visual probe that binds to anionic site on the plasma membrane. Biochemical studies with (125)I-labeled DMPA-LDL showed that the membrane-bound lipoprotein was internalized and hydrolyzed in lysosomes. The turnover time for cell bound (125)I-DMPA-LDL, i.e., the time in which the amount of (125)I-DMPA-LDL degraded was equal to the steady-state cellular content of the lipoprotein, was about 50 h. Because the DMPA-LDL gained access to fibroblasts by binding nonspecifically to anionic sites on the cell surface rather than by binding to the physiologic LDL receptor, its uptake failed to be regulated under conditions in which the uptake of native LDL was reduced by feedback suppression of the LDL receptor. As a result, unlike the case with native LDL, the DMPA-LDL accumulated progressively within the cell, and this led to a massive increase in the cellular content of both free and esterified cholesterol. Studies with (14)C-oleate showed that at least 20 percent of the accumulated cholesteryl esters represented cholesterol that had been esterified within the cell. After 4 days of incubation with 10 μg/ml of DMPA-LDL, fibroblasts had accumulated so much cholesteryl ester that neutral lipid droplets were visible at the light microscope level with Oil Red O staining. By electron microscopy, these intracellular lipid droplets were observed to lack a tripartite limiting membrane. The ability to cause the overaccumulation of cholesteryl esters within cells by using DMPA-LDL provides a model system for study of the pathologic consequences at the cellular level of massive deposition of cholesteryl ester.  相似文献

A perennial ryegrass sward was established in the autumn of 1989 in an enclosed 0.3 ha site and was exposed to captive wild rabbit (Oryctolagus cuniculus) grazing. Rabbit numbers were varied from 16 rabbits ha-1 in winter to 55 rabbits ha-1 corresponding to natural fluctuations in the field. The original sward was grown for three seasons (1989/1990 to 1990/1991) and a range of grazing regimes imposed. In 1989/1990 four grazing regimes were arranged in a replicated split block design in the experimental area. These were ungrazed, winter grazed, spring grazed and totally grazed. In the second year of the experiment all plots were exposed to grazing with the exception of the originally protected plots. This was to examine the effects of longer-term grazing damage on a ryegrass sward. In the final year half of all treatments were protected to study recovery of a sward which had been damaged previously. The remainder of the plots were exposed to grazing. Three cuts were harvested in each year and the productivity assessed in terms of yield and botanical composition. There was a significant reduction in ryegrass proportion in grazed swards following the first winter of grazing, while clover, other grasses and weeds were enhanced. The promotion of clover content in grazed swards was a feature throughout the 3 yr of the study. In the second year, protection of grazed swards led to a restoration of yields, although the botanical composition remained altered. The exposure of previously ungrazed plots in the final year of the experiment showed that these swards were particularly attractive to rabbits and they suffered the greatest yield losses relative to the protected plots at the first cut in 1991. In this year dry weather conditions were experienced following the first cut and demonstrated that yield losses are exacerbated when rabbit grazing is compounded with adverse growing conditions. The results overall indicate that protection of ryegrass swards at the establishment phase is important, but that a degree of recovery is possible by providing protection at a later stage, although the ryegrass component is still impaired in swards which suffer early damage.  相似文献

Abstract Decapods associated with two species of seagrass (Zostera capricorni and Posidonia australis) were sampled at several sites within estuaries on the New South Wales coast for 3 years. Two species of decapod (Macrobrachium intermedium and Penaeus plebejus) were abundant in Z. capricorni. Four species (M. intermedium, P. plebejus, Chlorotocella leptorhynchus and Nectocarcinus tuberculosus) were abundant in P. australis. Abundance of all these species fluctuated greatly among sites and through time. Some seasonal patterns in abundance were evident, and appeared to be caused by recruitment and subsequent mortality or migration of individuals. Similarly, there were some consistent spatial patterns in abundance of most species. In particular, some sites supported consistently more juveniles during the same period of separate years. Within Jervis Bay, P. australis was also sampled in shallow (1–2m) and deep (7–8 m) water. Three species were abundant (C. leptorhynchus, N. tuberculosus and Processa australiense), but showed no consistent differences between the two depths. Abundances of these species did fluctuate greatly among the sites within Jervis Bay and through time. Such observations of the natural rates of change in abundance of populations are essential in order to be able to detect the effects of human disturbances.  相似文献

Bot. 38: 26–41. Growth, demography and N economy of 1–6-year-old standsof Acacia pulchella, A. alata, A. extensa and Bossiaea aquifoliumwere examined using population sampling to assess annual incrementsof N as living biomass, and returns of N as litter, seed anddead plants. Dependence on nitrogen fixation was assessed fromseasonal profiles of acetylene reduction, employing data fromprevious calibrations to convert C2H2 reduced to N2 fixed. After2 years of slow growth and minimal reproduction all speciesgrew rapidly to achieve maximum or near maximum size and seedproduction. Intense self-thinning of stands occurred in thesecond and third years, especially in the highly dense standsof the smallest species, A. alata. Annual turnover in standsranged from 0?3 to 127 kg N ha–1 year–1, dependingon species current age and density of a stand. Returns of Nas litter and shed seed comprised small proportions of the annualbudgets, but returns due to plant death equalled or exceededincrements of living biomass in years when stands were thinningrapidly. Proportional dependencies of the species on fixed N2were relatively high (13–61%) in first year seedlings,and then declined markedly to 1?1–3?4percnt; in the second,0?3–1?6% in the third, and, with one exception, to wellbelow 1% in the fourth and sixth year stands.
