In the freshwater snail Biomphalaria glabrata, the outer mantleepithelium and the transitional epithelium from the outer mantleepithelium to the belt are characterized by apical vesiclesof different electron density and vacuoles including lipid dropletsand fibrillar structures. Wide intercellular spaces predominatein the transitional epithelium. In addition to belt desmosomes,freeze fracture studies detect septate junctions as apical intercellularjunctions. The permeability of the septate junctions is testedby injecting anaesthetized snails with solutions containingperoxidase and lanthanum-nitrate. The septate junctions appearto be impermeable for the protein and inhibit permeation ofthe ion. Alkaline phosphatase is detected at the light microscope levelin the transitional and outer mantle epithelium. At the electronmicroscope level localization of alkaline phosphatase is restrictedto the apical and basal cell membranes of the same epithelia.An ATPase with low affinity to Ca2+ and Mg2+ ions is observedin the lumen of vacuoles in the proximal belt, the transitionaland the outer mantle epithelium. The possible role of the vacuolesas a site of synthesis or modification of the intercrystallinematrix is discussed. A high affinity Ca2+/Mg2+ ATPase is detectedwith the help of Electron Spectroscopic Imaging in the cellmembranes and in membranes of the rER. Infectivity of second‐stage juvenile (J2) populations of Heterodera schachtii was assayed with radish.The numbers of J2 in three‐day‐old seedlings were proportional to the numbers of J2 in two differently textured soils.In a microplot trial with a known H.schachtii‐supprcssivc soil, half of the plots contained untreated suppressive soil, the other half contained the same soil, but methyl iodide‐fumigated and therefore conducive.Both soils were infested with cysts introducing the equivalents of 0, 30, 60 or 120 H.schachtii eggs g‐1 soil, kept moist for 2 months, and then planted to Swiss chard.The numbers of J2 in radish roots were proportional to the numbers of H.schachtii eggs introduced into the microplots, at a low level of detection in suppressive soil and at a high level in conducive soil.Growth of Swiss chard was not different at increasing infestation levels in suppressive soil, but growth was reduced in conducive soil proportionally to increasing nematode infestation level.  相似文献

This review summarizes the previous and current literature on the immunogenetics of idiopathic inflammatory myopathy (IIM) and updates the research progress that has been made over the past decade. A substantial part of the genetic risk for developing adult- and juvenile-onset IIM lies within the major histocompatibility complex (MHC), and a tight relationship exists between individual human leukocyte antigen alleles and specific serological subtypes, which in turn dictate clinical disease phenotypes. Multiple genetic regions outside of the MHC are increasingly being identified in conferring IIM disease susceptibility. We are still challenged with the task of studying a serologically and clinically heterogeneous disorder that is rarer by orders of magnitude than the likes of rheumatoid arthritis. An ongoing and internationally coordinated IIM genome-wide association study may provide further insights into IIM immunogenetics.  相似文献

High surface ozone concentrations are recognized as an emerging threat to food security in Asia. This study aimed at determining the effects of ozone on the nutritive quality of rice straw, a by‐product of rice grain production and a major feed resource for ruminant livestock. Further, the question was addressed whether negative effects of ozone can be mitigated through molecular breeding. Rice plants from three different genotypes were exposed to four different ozone treatments in fumigation chambers from transplanting to maturity. These genotypes were (i) IR64, one of the most wide spread indica varieties in the world, (ii) Nipponbare, a typical japonica variety, and (iii) SL41, an ozone tolerant breeding line that carried chromosomal inserts at the ozone tolerance QTL OzT9 in the genetic background of Nipponbare. The treatments consisted of (i) charcoal filtered air, (ii) simulated ambient ozone concentration, (iii) 2 × ambient ozone concentration, and (iv) 2.5 × ambient ozone concentration. The effects of ozone on the chemical composition of straw were clearly dependent of the ozone level, and were significant even at ambient ozone concentration. Increases in crude ash, lignin and phenolics concentration adversely affected the digestibility as demonstrated in incubation experiments simulating rumen digestion in vitro. Negative ozone effects included reductions in the rate and extent of gas production due to inhibition of microbial fermentation, reduced formation of short chain fatty acids (SCFA), and a decrease in the true organic matter digestibility. The ozone tolerant genotype SL41 was less responsive to ozone than its more susceptible recurrent parent Nipponbare in terms of lignin and phenolics formation, organic matter digestibility and SCFA production. These data demonstrate that the feed quality of rice straw is affected by ozone even at ambient concentration, and that these negative effects are mitigated by the ozone tolerance QTL OzT9.  相似文献

Owing to their role as vectors of malaria parasites, species of the Anopheles maculipennis complex (Diptera: Culicidae) Meigen were intensively studied in the past, but with the disappearance of malaria in Germany in the middle of the last century, the interest in this field of research declined. A comprehensive ecological analysis of the current species distribution for Germany is lacking. Between 2010 and 2013, a total of 1445 mosquitoes of the An. maculipennis complex were collected at 72 different sites in Germany. The samples comprise 722 single individuals as well as 723 individuals in 90 pools of up to 25 mosquitoes. All samples were analysed with newly developed species‐specific qPCR assays for the identification of the four German species using nucleotide differences within the internal transcribed spacer 2 (ITS2) ribosomal DNA. All gathered data were used for species distribution modelling. The overall prevalence of An. messeae s.l. was highest with 98.89% of all pools; An. daciae with 6.93% of all individuals and An. messeae s.s. with 69.53%. The prevalence of the other two species was relatively low: An. maculipennis s.s. with 13.30% of all individuals (6.67% of all pools) and An. atroparvus with 1.80% of all individuals (1.11% of all pools).  相似文献
