Until recently, Tetrahymena thermophila has rarely been isolated from nature. With improved sampling procedures, T. thermophila has been found in ponds in many northeastern states. The availability of resident populations makes possible both population and ecological genetic studies. All seven known mating types have been recovered; no eighth mating type has been found. Crosses among whole-genome homozygotes derived from Pennsylvania isolates reveal a spectrum genotypes with mating type alleles resembling traditional A (IV- and VII-) and B(I-) categories. The genotypes differ significantly with respect to mating type frequency, both among themselves and from previously described genotypes. One A-category genotype appears to lack mating type II, while one A-category and all B-category genotypes have low frequencies of mating type III, thus accounting for the low frequency of III in the pond. The low frequency of III in all five B-category genotypes examined suggests that the founding allele in this region was low for III. These and other differences are discussed both in terms of mating type frequencies in the pond and in terms of the possible molecular structure of mat alleles. By contrast, numerous variants of the cell surface immobilization antigen are found in addition to the previously described i-antigens. Variants of the known SerH alleles include those with restriction fragment length polymorphisms and temperature sensitivity as well as alleles with new antigenic specificity. Multiple alleles are present in single ponds. Genes exhibiting serially dominant epistasis over SerH genes also are found. In two instances (K and C), families of antigenically similar polypeptides are expressed in place of H i-antigen. Molecular weight differences suggest that these paralogous i-antigen genes evolve by gene duplication and unequal crossing over within central repeats. The existence of complex patterns of epistasis together with seasonal changes in i-ag frequencies suggest that i-ag play an important, but as yet unknown, ecological role related to the occurrence of frequent conjugation.  相似文献

ABSTRACT Weekly counts of western Atlantic red knots (Calidris canutus rufa) at their Delaware Bay migration stopover site have suggested a major decline since the 1980s. We estimated red knot spring passage population size in the New Jersey Coast-Delaware Bay region (DENJ; 2004 and 2006) and Virginia (VA; 2006 and 2007), USA, by correcting weekly aerial counts for mean daily residence probability between counts in a Monte-Carlo simulation. We used daily telemetry relocations in mark-resight models to estimate mean daily residence probability. Average daily residence probability was approximately 1.0 in mid-May, 0.96–0.97 in the week of 22 May, and 0.64–0.77 after May 28 in DENJ in 2004 and 2006 and in VA in 2006. Average daily residency was approximately 0.88 in VA in 2007 from 22 May to 5 June. No birds moved from VA to DENJ in 2006 and only 2 birds (5.5%) moved in 2007. Stopover population sizes (±SE) in DENJ were 17,108 ± 1,322 in 2004 and 19,555 ± 831 in 2006, and in VA were 7,224 ± 389 in 2006 and 8,332 ± 718 in 2007, significantly greater than peak aerial counts. Years with similar peak counts had different residence probabilities; hence, adjustments for turnover should be used in the future to assess annual population changes. Our results suggest that VA can support a significant portion of this red knot subspecies during migration in at least some years. Managing red knots for recovery should entail improving our understanding of the use of other Atlantic Coast sites and protecting key coastal habitat from disturbance and development.  相似文献

1. Terrestrial plant litter entering two forest and two pasture sites on upland streams in south-eastern Australia was measured over a 2-year period using traps suspended above the streams. Laterally transported terrestrial plant litter was also determined in one stream, but found to be only 10% of the total. 2. Litter accession to traps suspended above the stream was significantly lower at both forested sites than was litter fall in the adjacent riparian vegetation. 3. Litter input was high (600–700g DW m?2yr?1) and seasonal, with 30–50% of the annual total entering the stream during December—February (summer). However, seasonality was not as great as that recorded in Northern Hemisphere deciduous forest streams. 4. Leaves comprised the largest litter component, but represented a relatively smaller proportion of the litter than is usually the case in Northern Hemisphere deciduous forest streams; in contrast bark was a relatively large proportion of the litter. 5. At the sites flowing through pasture, litter accession was about two orders of magnitude lower than at the forested sites.  相似文献

Oocysts of Octosporella hystrix n. sp., Eimeria tachyglossi n. sp., and E. echidnae n. sp. are described from the feces of the echidna Tachyglossus aculeatus (Monotremata: Tachyglossidae) from Australia. Eimeria tachyglossi has subspherical oocysts, 26.4 × 23.7 μm in size, with a single oocyst wall; no micropyle; four ellipsoidal sporocysts 13.2 × 9.7, slightly pointed at one end, each containing two sporozoites. Eimeria echidnae has subspherical oocysts, 19.4 × 17.8 in size, with a single oocyst wall; no micropyle; four ellipsoidal sporocysts 9.8 × 7.8, blunt at both ends, each containing two sporozoites. Octosporella hystrix has ovoid or subspherical oocysts 32.9 × 29.7 in size with a thick outer and thin inner oocyst wall; no micropyle; eight sporocysts spherical or slightly subspherical 11.3 × 11.2 each containing two sporozoites lying in embrace, with an extensive granular sporocyst residuum about the equator of the sporocyst. Endogenous stages considered to be of E. tachyglossi at least, were recognized in the lamina propria and epithelium on villi in the small intestine of three echidnas.  相似文献

Measurements of annual litterfall rate and mean annual litter mass for various indigenous open-forests are listed, along with the consequent decomposition constant (k) and mean annual rainfall. Using data only from sites considered valid for comparison (i.e. all those with a sclerophyllous plant understorey, not recently burnt, grazed or invaded by an exotic decomposer, and where for accuracy of assessment the litter layer has been sampled on several occasions) a quadratic relationship between litterfall rate and litter mass was significant, with decomposition rates at sites increasing with increasing litterfall rate. The existence of such a relationship may be useful for future studies: if litter data obtained lie appreciably outside the values on the line of quadratic regression, it would be worthwhile checking the accuracy of litterfall and mass estimates, site past history and the validity of the decomposition constant estimate. There was a significant positive linear relationship between the decomposition constant and mean annual rainfall at sites where rainfall was within the range 600–1800 mm y-1, the general limits for open-forest site development. However, more data are required to increase the predictive value of relationship.  相似文献

The Signature Series Symposium “Cellular Therapies for Orthopaedics and Musculoskeletal Disease Proven and Unproven Therapies—Promise, Facts and Fantasy” was held as a pre-meeting of the 26th International Society for Cellular Therapy (ISCT) annual congress in Montreal, Canada, May 2, 2018. This was the first ISCT program that was entirely dedicated to the advancement of cell-based therapies for musculoskeletal diseases. Cellular therapies in musculoskeletal medicine are a source of great promise and opportunity. They are also the source of public controversy, confusion and misinformation. Patients, clinicians, scientists, industry and government share a commitment to clear communication and responsible development of the field. Therefore, this symposium convened thought leaders from around the world in a forum designed to catalyze communication and collaboration to bring the greatest possible innovation and value to patients with musculoskeletal conditions.  相似文献

1. Periphyton chlorophyll a (chl a), ash-free dry mass, taxonomic composition, and cellular and water-column nutrients were analysed every 4 weeks for a year at sixteen stream sites in New Zealand. The hypothesis was investigated that broad-scale differences in mean monthly periphyton development are defined primarily by the frequency of flood disturbances and the periphyton's interaction with the nutrients. it us of the streams as determined by catchment geology and land use. 2. Overall, mean monthly chl a concentration declined with increasing flood frequency (r= -0.711, P < 0.001), and seasonality in chl a was better defined at sites with a low frequency of floods. Chlorophyll a concentration was generally low throughout the year at sites with frequent floods (> 15 yr?1). 3. No relationship existed between inorganic nutrient concentrations and catchment geology or land development. However, conductivity declined significantly as a function of the percentage of the catchment underlain by nutrient-poor, hard rocks (plutonic and fine-grained metamorphic rocks) (r= -0.515, P < 0.05), but increased significantly with the percentage of the catchment in intensive agricultural land use (r= 0.799, P < 0.001). 4. Cellular nutrient concentrations suggested that nitrogen was the nutrient most commonly limiting periphyton production. In turn, cellular N concentrations declined significantly with increasing percentage of the catchment in hard rock (r= -0.5M, P < 0.05) and increased with percentage of the catchments in intensive agricultlural land use (r= 0.948, P < 0.001). 5.The sites were classified into three enrichment groups (high, moderate and low) based on their land use and underlying geology. Cellular N concentrations varied significantly among these enrichment groups (ANOVA F= 14.661, P < 0.001). 6. Log chl a decreased significantly with increases in the annual 80th percentile velocity. However, the relationship was significantly different among the enrichment groups. 7. A stepwise multiple regression on the full dataset identified that the frequency of floods, proportion of the catchment in high-intensity agricultural land use and proportion in alkaline rocks were the most significant factors explaining variation in mean monthly chl a among the sites (r2= 89%). 8. Overall, the results showed that flood disturbance and catchment enrichment regimes are probably the principal axes of the habitat template of periphyton among the study streams, and could be used to explain and predict broad-scale differences in periphyton development among other temperate stream ecosystems.  相似文献

A quantitative study of the soil and litter fauna of three South Australian low open forests was made for two years. The soil and vegetation of the sites were similar but the mean annual rainfall of the areas differed; 635, 690 and 1050 mm/y, respectively for the ‘dry’, ‘medium’ and ‘wet’ sites. Monthly samples were taken of litter, and the upper 0–4 cm and lower 4–8 cm soil layers, and the method used to estimate the numbers of active animals is also described. Mean annual population densities of Collembola and Acari were greatest at the medium site (9.5 and 48.6 × 103/m2, respectively) and least at the dry site (4.9 and 35.1 × 103/m2, respectively) and values are discussed in relation to site organic carbon content. Over all the sites, between 11–21, 67–75 and 10–15% of the collembolan populations, and 10–13, 66–72 and 16–21% of the acarine populations inhabited the litter layer and upper and lower soil layers respectively. Isotomidae were the most abundant collembolan family in each level at each site, but the overall proportion of Entomobryidae increased from the dry to wet site. Prostigmata were the most abundant acarine order in each level at two sites. Cryptostigmata were dominant in the litter layer of the wet site, and their overall proportion increased from the dry to wet site. Seasonal density changes were similar on all sites with minima in the summer and maxima in the winter months. The results obtained here combined with those from other published investigations indicate a trend of faunal population change over various ecosystems in Australia. Prostigmatid mites and iso-tomid Collembola dominate at arid sites. As sites become less arid and the amount of soil organic matter increases, the proportion of cryptostigmatid and mesostigmatid mites increases, and other groups of Collembola, particularly Entomobryidae, become more frequent.  相似文献

It has previously been suggested from physical evidence thatvessels in twigs of sycamore (Acer pseudoplatanus L.) are connectedby small pores in intervessel pit membranes. From results obtainedusing a combination of physical techniques and electron microscopyit is clear that this is an oversimplification: almost all intervesselpit membranes are absent in sycamore twig and stem wood, yetphysical tests still suggest that vessels are connected by poressmaller than pit apertures. Where these pores are is less certain.They may be in plugs observed to fill vessel lumens. They maypossibly be in a small number of intact intervessel pit membranesin the terminal elements of vessels, but such membranes couldnot be found under the microscope. In either case, the resultsof physical tests suggest that vessels connected by membranelessintervessel pits must effectively end together at a particularpoint in the tree. The surface proteins and glycoproteins on red cells from normal and Babesia bovis-infected calf blood have been compared. Several radiolabeling probes were used to label specifically external membrane molecules which were then separated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and identified by autoradiography or fluorography. No differences were observed among the Coomassie Blue-stained membrane proteins of erythrocytes from individual uninfected calves. Comparison of red cells from these animals also indicated no qualitative differences in the surface proteins with accessible tyrosyl residues labeled by lactoperoxidase-catalyzed radioiodnation, although some quantitative variation in the uptake of radioactivity into particular proteins was observed. the major radioiodinated bands on normal bovine erythrocytes had Mr of 165, 130, 90, and 45 kiloDaltons. However, labeling of surface glycoproteins by the periodate/[3H]NaBH4 and galactose oxidase (± neuraminidase)/[3H]NaBH4 methods showed significant differences in the surface proteins of red cells from individual uninfected calves. of 14 animals tested, 5 had major labeled glycoproteins of unique Mr. No changes were observed in radioiodinated surface proteins of total red cell samples from infected calves with 0.5-6% parasitemia. Radioiodination of concentrated infected red cells from the same samples (concentrated by selective hypotonic lysis of uninfected erythrocytes in KC1) resulted in the labeling of 3 new surface proteins, with Mr of 118, 115, and 60 kiloDaltons. the same new 125I-labeled bands were identified on infected cells from 3 avirulent strains of B. bovis used in vaccine production. Furthermore, in concentrated infected cells there was very poor radiolabeling of major bands strongly labeled on uninfected cells (Mr 165, 130, and 90 kiloDaltons), suggesting parasite-induced loss of these proteins. Although there were some differences in 3H-labeled surface glycoproteins of red cells from normal and. B. bovis -infected blood, they were restricted to minor labeled bands and were not seen consistently. the labeled surface glycoproteins of concentrated infected cells were very similar to those of the uninfected red blood cells from infected blood.  相似文献
