The significance of photorespiration in drought-stressed plants was studied by withholding water from wild-type barley (Hordeum vulgare L.) and from heterozygous mutants with reduced activities of chloroplastic glutamine synthetase (GS-2), glycine decarboxylase (GDC) or serine : glyoxylate aminotransferase (SGAT). Well-watered plants of all four genotypes had identical rates of photosynthesis. Under moderate drought stress (leaf water potentials between –1 and –2 MPa), photosynthesis was lower in the mutants than in the wild type, indicating that photorespiration was increased under these conditions. Analysis of chlorophyll a fluorescence revealed that, in the GDC and SGAT mutants, the lower rates of photosynthesis coincided with a decreased quantum efficiency of photosystem II and increased non-photochemical dissipation of excitation energy. Correspondingly, the de-epoxidation state of xanthophyll-cycle carotenoids was increased several-fold in the drought-stressed GDC and SGAT mutants compared with the wild type. Accumulation of glycine in the GDC mutant was further evidence for increased photorespiration in drought-stressed barley. The effect of drought on the photorespiratory enzymes was determined by immunological detection of protein abundance. While the contents of GS-2 and P- and H-protein of the GDC complex remained unchanged as drought stress developed, the content of NADH-dependent hydroxypyruvate reductase increased. Enzymes of the Benson–Calvin cycle, on the other hand, were either not affected (ribulose-1,5-bisphosphate carboxylase-oxygenase and plastidic fructose-1,6-bisphosphatase) or declined (sedoheptulose- 1,7-bisphosphatase and NADP-dependent glyceraldehyde-3-phosphate dehydrogenase). These data demonstrate that photorespiration was enhanced during drought stress in barley and that the control exerted by photorespiratory enzymes on the rate of photosynthetic electron transport and CO2 fixation was increased.  相似文献

SUMMARY: This work is part of a project that is currently applying microbiology to certain aspects of the multiple sclerosis problem. The present comparative studies were carried out in connection with atherosclerosis because this disease, like multiple sclerosis, involves deranged lipid metabolism. Twenty-one soil actinomycetes and a bacterium were isolated from agar streak plates containing homogenized human aorta as the source of nitrogen, carbon, and energy. These organisms included forms capable of digesting and growing on the ether-extractable lipids from human atherosclerotic aorta plaques, cholesterol, cholesterol esters, glycerides, phospholipids, bovine white matter and other related materials. Two actinomycetes, isolated in other studies and capable of demyelinating bovine spinal cord in vitro , produced lipoproteinase(s) which hydrolyzed lipemic serum obtained from dogs after a fatty meal.  相似文献

Cuticular melanism and innate immune parameters can share common physiological pathways in insects, and this functional connection may contribute to the maintenance of insect colour polymorphisms. However, evidence linking colouration and immune function has been equivocal, particularly when tested in wild populations. The present study investigates phenotypic links between colouration and immune function in migratory Mormon crickets (Anabrus simplex, Haldeman), in which juveniles occur in conspicuous colour variants but mature to become uniformly melanic adults. Wild‐caught insects are used to evaluate the relationship between juvenile colouration and three immune parameters: encapsulation ability, lysozyme‐like activity and phenoloxidase activity. As nymphs, brown crickets are better able to encapsulate an inert implant introduced into the haemocoel than green crickets, although the difference is slight and ceases after they all become darkly‐coloured adults. By contrast, adults that develop from brown nymphs have a higher basal phenoloxidase activity than those that develop from green nymphs, regardless of the fact that all adults are brown. Intrinsic factors other than colouration exert larger effects on immunity: males show stronger encapsulation responses but lower phenoloxidase activity than females, suggesting a sex‐specific trade‐off between these two immune parameters, and adults exhibit higher immune function than nymphs. In summary, modest support is found for a correlation between cuticular melanism and increased immune function in wild Mormon crickets. Additional intrinsic factors such as developmental stage and sex appear to interact with colouration and have a more substantial connection to immune function in the wild.  相似文献

Globally significant increases in the riverine delivery of nutrients and suspended particulate matter have occurred with deforestation. We report here significant increases in streamwater transport of dissolved silicate (DSi) following experimental forest harvesting at the Hubbard Brook Experimental Forest, NH, USA. The magnitude of the streamwater response varied with the type of disturbance with the highest DSi export fluxes occurring in the manipulations that left the most plant materials on the soil surface and disturbed the soil surface least. No measurable loss of amorphous silica (ASi) was detected from the soil profile; however, ASi was redistributed within the soil profile after forest disturbance. Mass‐balance calculations demonstrate that some fraction of the DSi exported must come from dissolution of ASi and export as DSi. Land clearance and the development of agriculture may result in an enhanced flux of DSi coupled with enhanced erosion losses of ASi contained in phytoliths.  相似文献

A rice alginate prill formulation of isolate EN - 4 of Fusarium oxysporum f . sp . erythroxyli, pathogenic to Erythroxylum coca var . coca (coca) , was evaluated in greenhouse and field studies for its ability to enhance pathogen populations in the soil and cause disease in coca . The formulation was applied to four different soil types in the greenhouse at 33 . 6 kg ha 1 . It enhanced the population of EN - 4 in each soil and most ( > 90%) of the fungal population remained in the upper 5 cm of soil during the 49 - day experiment . When applied in field experiments , the formulation enhanced the population of EN - 4 in the soil . Isolate EN - 4 was present in the upper 7 . 6 cm of soil 28 days after application at populations similar to those in the greenhouse studies (1 103 to 1 104 colony - forming units (CFUs) / g of soil) . Elevated populations of the pathogen (1 102 CFUs / g of soil) were still present in treated soils 229 days after application of the formulation . The areas used for field studies were already infested with the pathogen and typically developed high levels of fusarium wilt within 2 years of planting with coca . The formulated F. oxysporum began having a significant effect on plant death 100 - 200 days after application based on repeated measures analysis . These data suggest that a formulation of F. oxysporum f . sp . erythroxyli which enhances the incidence of fusarium wilt in coca fields can be produced using established techniques .  相似文献

The genus Kawanaphila (Tettigoniidae: Zaprochilinae) is unusual among the Tettigoniidae in the possession of sexually dimorphic auditory organs. We examined the auditory system and acoustic behaviour of two previously unstudied species in this genus to test whether reduced hearing in males is consistently associated with reduced male–male competition. Kawanaphila yarraga (Rentz, 1993) and K. mirla (Rentz, 1993) are both sexually dimorphic with respect to their auditory system, but to different degrees. Males of both species produce songs consisting of trains of brief (< 1 ms) pure-tone sound pulses at ultrasonic frequencies (K. yarraga, 40 kHz;K. mirla, 70 kHz). In both species, female hearing is more sensitive than that of males by 10 dB. In addition, male K. mirla are most sensitive at lower frequencies than females. Male and female K. yarraga differed only in sensitivity, not in tuning. The two species also differ in their degree of sexual dimorphism in auditory anatomy. Kawanaphila mirla males lack some auditory specializations of the prothoracic tracheal system, which are present in the normal tettigoniid condition in females. In K. yarraga males these structures are present, but reduced in size relative to females. The acoustic behaviour of males of the two species is consistent with this pattern of relative auditory sensitivity. Males of both species interact acoustically by altering the timing of their sound output to synchronize with neighbouring males. However, K. mirla males only interact in this way over very short distances (< 5 m), whereas K. yarraga males interact with neighbours up to at least 10 m distant. These results indicate that, although males of the two species differ in hearing sensitivity, the nature of their responses to conspecific calls are similar to one another and to those of other acoustic insects. This suggests that acoustically mediated male–male competition may be maintained even while selection favours a reduction in male auditory sensitivity.  相似文献

Abstract This paper describes the first results for an alternative approach to the development of sediment quality criteria in the nearshore areas of the Laurentian Great Lakes. The approach is derived from methods developed in the United Kingdom for establishing predictive relationships between macroinvertebrate fauna and the physico-chemistry of riverine environments. The technique involves a multivariate statistical approach using (i) data on the structure of benthic invertebrate communities, (ii) functional responses (survival, growth and reproduction) in four sediment toxicity tests (bioassays) with benthic invertebrates; and (iii) selected environmental variables at 96 reference (‘clean’) sites in the nearshore areas of all five Great Lakes (Lakes Superior, Huron, Erie, Ontario and Michigan). Two pattern recognition techniques (using the computer software package PATN) are employed in the analysis: cluster analysis and ordination. The ordination vector scores from the original axes of the pattern analysis are correlated (using CORR in SAS) with environmental variables which are anticipated to be least affected by anthropogenic activities (e. g. alkalinity, depth, silt, sodium etc.). Multiple discriminant analysis (MDA) is used to relate the site groupings from the pattern analysis to the environmental variables and to generate a model that can be used to predict community assemblages and functional responses at new sites with unknown but potential contamination. The predicted community assemblages and functional responses are then compared with the actual benthic communities and responses at a site, and the need for remedial action is determined. The predictive capability of the discriminant model was confirmed by performing several validation runs on subsets of the data. An example of the use of the model for sediment in Collingwood Bay (an area of concern designated by the IJC in Georgian Bay, Lake Huron) is presented and the technique is shown to be more precise in determining the need for remediation than the currently used provincial sediment quality criteria based on Screening Level Concentration (SLC) and laboratory toxicity tests. The ultimate goal of the study is the development of a method to determine the need for, and the success of, remedial action and to predict what benthic communities should look like at a site if it were clean and what responses of organisms in sediment toxicity tests constitute an acceptable end-point.  相似文献

Antibodies specific to the granulosis virus (GV) of the codling moth (Cydia pomonella) were detected by indirect ELISA in sera of woodmice (Apodemus sylvaticus) trapped in an apple orchard sprayed by tractor-drawn mist blower with 2 × 1013 virus granules C. pomonella GV/ha. Such antibodies were absent from sera of mice trapped before virus had been applied. Virus antigens were detected by double antibody sandwich ELISA on the ground in and around the orchard and also in the faeces of woodmice inhabiting the orchard, but did not persist from one year to another. Some, but not all, of the samples containing virus antigens were found to be infective.  相似文献
