Candida antarctica Lipase B (CALB), a metal-free enzyme, was successfully employed as catalyst for ring-opening copolymerization of omega-pentadecalactone (PDL) with p-dioxanone (DO) under mild reaction conditions (<80 degrees C, atmospheric pressure). Poly(PDL-co-DO) with high molecular weight (Mw > 30 000) and a wide range of comonomer contents was synthesized using various PDL/DO feed ratios. During the copolymerization reaction, large ring PDL was found to be more reactive than its smaller counterpart DO, resulting in higher PDL/DO unit ratios in polymer chains than the corresponding PDL/DO monomer ratios in the feed. The copolymers were typically isolated in 50-90 wt % yields as the monomer conversion was limited by the equilibrium between monomers and copolymer. 1H and 13C NMR analysis on poly(PDL-co-DO) formed by CALB showed that the copolymers contain nearly random sequences of PDL and DO units with a slight tendency toward alternating arrangements. Copolymerization with PDL was found to remarkably enhance PDO thermal stability. Differential scanning calorimetry (DSC) and wide-angle X-ray scattering (WAXS) results demonstrate high crystallinity in all copolymers over the whole range of compositions. Depending on copolymer composition, the crystal lattice of either PDO or PPDL hosts units of the other comonomer, a behavior typical of an isodimorphic system. In poly(PDL-co-DO), both melting temperature and melting enthalpy display a minimum at 70 mol % DO, that is, at the pseudoeutectic composition. WAXS diffractograms show one crystal phase (that of either PPDL or PDO) on either side of the pseudoeutectic and coexistence of PPDL and PDO crystals at the pseudoeutectic.  相似文献

G12 rotaviruses were first detected in diarrheic children in the Philippines in 1987, but no further cases were reported until 1998. However, G12 rotaviruses have been detected all over the world in recent years. Here, we report the worldwide variations of G12 rotaviruses to investigate the evolutionary mechanisms by which they managed to spread globally in a short period of time. We sequenced the complete genomes (11 segments) of nine G12 rotaviruses isolated in Bangladesh, Belgium, Thailand, and the Philippines and compared them with the genomes of other rotavirus strains. Our genetic analyses revealed that after introduction of the VP7 gene of the rare G12 genotype into more common local strains through reassortment, a vast genetic diversity was generated and several new variants with distinct gene constellations emerged. These reassortment events most likely took place in Southeast Asian countries and spread to other parts of the world. The acquirement of gene segments from human-adapted rotaviruses might allow G12 to better propagate in humans and hence to develop into an important emerging human pathogen.  相似文献

Recognizing the severity of road effects and need for developing a natural and self-sustained roadside vegetation cover, this study aimed to provide an ecological basis for selecting desirable native plants based on their autecological attributes by floristic analysis of naturally colonized plants in roadside microhabitats. We hypothesized that (i) vegetation zonation along roadsides is a function of the different microtopography and substrate types (microhabitats) created by road construction and (ii) plant colonization in these microhabitats is dependant upon the presence of suitable regeneration traits adapted to the specific microhabitats. We identified four distinct microhabitats namely shoulder, side slope, ditch and back slope from the edge of the road to the edge of the forest. We conducted vegetation and soil analyses in these microhabitats along 34 random transects running perpendicular to the road to the edge of forest in a 14 km section of the Trans Canada Highway (TCH) in Terra Nova National Park (TNNP), Newfoundland, Canada. The multi-response permutation procedure (MRPP) confirmed that the plant communities of the four roadside microhabitats were significantly different from each other. Canonical correspondence analysis (CCA) showed that composition of roadside plant communities was related to gradients of soil moisture content, bulk density, organic matter depth and pH. Several indicator plants, determined by indicator species analysis (ISA), were abundant in their respective microhabitats, indicating their affinity to particular sets of environmental conditions. The root–shoot analysis of selected dominant plants across the roadside habitats revealed that plants of a particular microhabitat had similar above and below ground spread and biomass allocation patterns.We found that floristic zonation along roadsides is a function of roadside microtopography, substrate type and environmental gradients created by the road building process. The similarities in above and below ground spread and biomass allocation patterns of dominant plants of respective microhabitats could be used as a basis of interpreting the role of autecological attributes of the species that enable them to establish in specific microhabitats. Several native plants, such as Empetrum nigrum, Juniperus communis, Vaccinium angustifolium, Trifolium repens, and Anaphalis margaritaceae are naturally abundant in side slopes and possess autecological attributes such as low stature, widespread above- and below-ground parts, and drought tolerance. Presence of these desirable properties and their perennial habit make them excellent candidates for roadside revegetation.  相似文献

MicroRNAs (miRNAs) are a class of 17-25 nt non-coding RNAs that have been shown to have critical functions in a wide variety of biological processes during development. Recently developed miRNA microarray techniques have helped to accelerate research on miRNAs. However, in some instances there is only a limited amount of material available for analysis, which requires more sensitive techniques that can preferably work on single cells. Here we demonstrate that it is possible to analyse miRNA in single cells by using a real-time PCR-based 220-plex miRNA expression profiling method. Development of this technique will greatly facilitate miRNA-related research on cells, such as the founder population of primordial germ cells where rapid and dynamic changes occur in a few cells, and for analysing heterogeneous population of cells. In these and similar cases, our method of single cell analysis is critical for elucidating the diverse roles of miRNAs.  相似文献

Nitric oxide (NO), a potent vasodilator, plays a pivotal role in blood pressure regulation. Endothelial NO synthase gene (NOS3) polymorphisms influence NO levels. Here, we investigated the role of the – 922A/G, – 786T/C, 4b/4a, and 894G/T polymorphisms of the NOS3 and NOx levels in 800 consecutive unrelated subjects comprising 455 patients of essential hypertension and 345 controls. The polymorphisms were investigated independently and as haplotypes. Plasma NOx levels (nitrate and nitrite) were estimated by the Griess method. Genotype frequencies for the –786T/C, 4b/4a, and 894G/T polymorphisms differed significantly (P < 0.001) between patients and controls and were associated with an increased risk of hypertension (OR = 2.0, OR = 3.8, OR = 1.6, respectively). The 4-locus haplotypes ATaG (H1), ATaT (H2), and GCaG (H3) were significantly associated with essential hypertension and served as susceptible haplotypes (P ≤ 0.0001). On the other hand, haplotypes ATbG (H4) and GTbG (H5) were negatively associated with hypertension and served as protective haplotypes (P < 0.0001). NOx levels were significantly lower in patients than controls (P < 0.0001). The individual polymorphisms showed marginal association with NOx level; however, the susceptible haplotype H2 associated significantly with lower NOx levels in patients (P < 0.001) and conversely the haplotype H4 with higher NOx levels in controls (P < 0.001). In conclusion, the 4b/4a and likely – 786T/C polymorphisms were identified as the determinants modifying the risk of hypertension. This study identifies the NOS3 variants and haplotypes as genetic risk factors and as useful markers of increased susceptibility to the risk of essential hypertension.  相似文献

To investigate the potential functional cooperation between p27Kip1 and p130 in vivo, we generated mice deficient for both p27Kip1 and p130. In p27Kip1-/-; p130-/- mice, the cellularity of the spleens but not the thymi is significantly increased compared with that of their p27Kip1-/- counterparts, affecting the lymphoid, erythroid, and myeloid compartments. In vivo cell proliferation is significantly augmented in the B and T cells, monocytes, macrophages, and erythroid progenitors in the spleens of p27Kip1-/-; p130-/- animals. Immunoprecipitation and immunodepletion studies indicate that p130 can compensate for the absence of p27Kip1 in binding to and repressing CDK2 and is the predominant CDK-inhibitor associated with the inactive CDK2 in the p27Kip1-/- splenocytes. The finding that the p27Kip1-/-; p130-/- splenic B cells are hypersensitive to mitogenic stimulations in vitro lends support to the concept that the hyperproliferation of splenocytes is not a result of the influence of their microenvironment. In summary, our findings provide genetic and molecular evidence to show that p130 is a bona fide cyclin-dependent kinase inhibitor and cooperates with p27Kip1 to regulate hematopoietic cell proliferation in vivo.  相似文献
