While hydroxyl radical cleavage is widely used to map RNA tertiary structure, lack of mechanistic understanding of strand break formation limits the degree of structural insight that can be obtained from this experiment. Here, we determine how individual ribose hydrogens of sarcin/ricin loop RNA participate in strand cleavage. We find that substituting deuterium for hydrogen at a ribose 5′-carbon produces a kinetic isotope effect on cleavage; the major cleavage product is an RNA strand terminated by a 5′-aldehyde. We conclude that hydroxyl radical abstracts a 5′-hydrogen atom, leading to RNA strand cleavage. We used this approach to obtain structural information for a GUA base triple, a common tertiary structural feature of RNA. Cleavage at U exhibits a large 5′ deuterium kinetic isotope effect, a potential signature of a base triple. Others had noted a ribose-phosphate hydrogen bond involving the G 2′-OH and the U phosphate of the GUA triple, and suggested that this hydrogen bond contributes to backbone rigidity. Substituting deoxyguanosine for G, to eliminate this hydrogen bond, results in a substantial decrease in cleavage at G and U of the triple. The scale morphology of pleuronectiforms in the Gulf of Oman remains insufficiently known. This study used light microscopy and morphological analysis to examine scale variation across the flank of four Tonguefishes species; Cynoglossus arel, C. bilineatus, C. lingua, and C. puncticeps. Scales were extracted from six flank regions, three on the eyed and blind sides, respectively. The most differentiated species was C. arel, which showed significant differences in four size variables in five regions. In Cynoglossus arel and C. lingua, the scales of the eyed side were ctenoid, and those scales from the blind side were cycloid; C. puncticeps have ctenoid scales on both flank sides and C. bilineatus has cycloid scales on both sides. All species' scales on the blind side have fewer ctenial spines (except in C. bilineatus). This study indicated that scale morphology demonstrated considerable variation among the flank regions of the examined species. As a result, the scales from the head and the trunk regions of the eyed side and the scales from the head region of the blind side have a good power of species separation in this family. 相似文献

This study was performed to investigate the effect of ondansetron, a serotonin receptor (5‐HT3) antagonist, in the alleviation of diclofenac‐induced kidney injuries. NMRI mice were randomly divided into six groups and treated with (A) untreated control group, (B) diclofenac (100 mg/kg), (C) ondansetron (1 mg/kg), (D to F) ondansetron (0.1, 0.5, and 1 mg/kg, respectively) and diclofenac (100 mg/kg) for last 3 days of experiment. The oxidative stress tests strongly demonstrated the negative synergistic effects of diclofenac and ondansetron, regarding the observation of dose‐dependent enhancement of malondialdehyde concentration, and reduction of glutathione content, and superoxide dismutase and catalase activity. Histopathological analyses revealed dose‐dependent tubular epithelial cells degeneration, outstanding mononuclear cells infiltration, clear necrosis at the papillary region of kidney, dilation, and vascular hyperemia in mice kidney tissues treated with ondansetron and diclofenac. Conclusively, these findings suggested the possible ondansetron‐diclofenac interaction through the induction of oxidative stress. 相似文献

An improved method has been developed for the large-scale purification of covalently closed circular (CCC) plasmid DNA molecules of sizes ranging from 4·3 to 73 kb. This protocol uses an alkaline-lysis procedure followed by acid-phenol extraction but with several modifications to previously reported methods. The principal modification is the replacement of NaCl by MgCl2 in the extraction buffer to improve yield and to remove chromosomal and other non-CCC plasmid DNA. Plasmid DNA can be purified in less than 1 h and used successfully in restriction enzyme analysis and cloning experiments. 相似文献

An Iranian National Quince collection containing 40 quince genotypes, originating from six distinct geographic areas, was screened using 15 SSR markers developed originally for apple and pear genomes. Overall, 13 markers exhibited polymorphism, with an average of 5.36 putative alleles per locus and a mean PIC value of 0.76. An UPGMA analysis divided the quince genotypes into five major clusters. The same results were obtained when the principal coordinates were plotted. The assignment test successfully allocated 83% of individuals into their place of origin. These results agree somewhat with the geographic origin of the quince accessions, and we conclude that geographic isolation leads to considerable genetic differentiation among Iranian quince collections. A significant ratio of transferability with a mean of 87.86% was measured, and we deduced that STMS markers derived from pear and apple have enough potential to detect polymorphism and differentiation in quince. 相似文献

AbstractMicrobial community structure reflects the surrounding natural environment and changes to that environment. Although the subsurface at 5–100?m depth is important for human activities and there are potential risks of environmental pollution in this region, there have been only a few reports of subsurface microbial community structures in terrestrial areas. We investigated the diversity and community compositions of Bacteria and Archaea in boring cores collected from various depths at three different sites in the southern Kanto Plain, Japan. The results of 16S rRNA gene amplicon sequencing using MiSeq showed that the microbial community composition varied with the geological unit. Proteobacteria (Alphaproteobacteria and Gammaproteobacteria) were dominant members within sediments accumulated during the Pleistocene in the Musashino Upland. In contrast, Acidobacteria and Chloroflexi characteristically appeared in the Holocene layers of the Arakawa Lowland. These data suggest that the subsurface microbial composition is controlled by the geological features of the sediments. 相似文献

It is usually assumed that new beneficial mutations are extremely rare. Yet, few experiments have been performed in multicellular
organisms that measure the effect of new beneficial mutations on viability and other measures of fitness. In most experiments,
it is difficult to clearly distinguish whether adaptations have occurred due to selection on new beneficial mutations or on
preexisting genetic variation. Using a modification of a Dobzhansky and Spassky (Evolution 1:191–216, 1947) assay to study change in viability over generations, we have observed an increase in viability in lines homozygous for the
second and third chromosomes of Drosophila melanogaster in 6–26 generations due to the occurrence of new beneficial mutations in population sizes of 20, 100 and 1,000. The lines
with the lowest initial viability responded the fastest to new beneficial mutations. These results show that new beneficial
mutations, along with selection, can quickly increase viability and fitness even in small populations. Hence, new advantageous

Plasmodium berghei ookinete kinetics and densities were examined in the blood meals of 3 species of Anopheles mosquitoes fed simultaneously from a gametocytemic mouse. Simple techniques were developed for estimating relative and absolute ookinete densities within individual mosquito blood meals. The kinetics of ookinete formation were similar in all 3 species, with peak ookinete densities occurring from 12 to 24 hr postingestion. Ookinete densities consistently were lower in Anopheles stephensi than in Anopheles albimanus or Anopheles freeborni and could not be accounted for by species differences in blood meal volumes or gametocyte densities. Likely explanations involve species differences in blood meal hemolysis or sampling error as the result of ookinete emigration from the blood meal. 相似文献
