Properties of the Escherichia coli RuvA and RuvB proteins involved in DNA repair, recombination and mutagenesis

The ruvA and ruvB genes constitute an operon, which is regulated by the SOS system and involved in DNA repair, recombination and mutagenesis. RuvA protein binds to both single-stranded and double-stranded DNA. RuvB protein has weak ATPase activity. RuvA bound to DNA greatly enhances ATPase activity of RuvB. UV-irradiation to supercoiled DNA further enhances the stimulatory effect of RuvA on the RuvB ATPase activity. In the presence of ATP the RuvA-RuvB complex has an activity that renatures cruciform structures formed by heating and gradually cooling supercoiled DNA with an inverted repeat. These findings suggest that the RuvA-RuvB complex interacts with an irregular conformation in damaged DNA and induces conformational changes in DNA using energy provided by ATP hydrolysis, so that it facilitates DNA repair, recombination and error prone replication.     

Chondrosarcoma of the trachea. A case report and literature review

The cytologic presentation of a case of chondrosarcoma of the trachea in a 72-year-old man is described. A mass detected on routine chest roentgenogram and defined by CT scan was used to make a touch imprint smear during partial tumor resection. The cytologic findings included round or polygonal cells with occasional binucleation, round hyperchromatic nuclei and prominent nucleoli, present in an amorphous pink-violet or light-blue background containing fragments of chondroid tissue. The histopathology was interpreted as a low-grade chondrosarcoma. Cartilaginous tumors of the trachea should be considered in the differential diagnosis of upper airway obstruction.     

Effects of hypergravity environment of the ultrastructure of the golden hamster parathyroid gland

The fine structure of the parathyroid glands of golden hamsters exposed to 2, 5 or 10 g environment for 5 h was studied. In the centrifuged hamsters, many secretory granules are located in a peripheral position just beneath the plasma membrane of chief cells, and the Golgi complexes and cisternae of the granular endoplasmic reticulum are significantly increased compared with those of control animals. There are no significant differences between the control and centrifuged animals with regard to secretory granules, large secretory granules, lysosomes, vacuolar bodies and lipid droplets. These findings suggest that the secretory activity of the parathyroid gland may be stimulated in response to hypergravity environment.     

Comparative studies of lipoteichoic acids from several Bacillus strains.

Structural studies were carried out on lipoteichoic acids obtained from defatted cells of 10 Bacillus strains by phenol-water partition followed by chromatography on DEAE-Sephacel and Octyl-Sepharose columns. A group of the tested bacteria (group A), Bacillus subtilis, Bacillus licheniformis, and Bacillus pumilus, was shown to have a diacyl form of lipoteichoic acids which contained D-alanine, D-glucose, D-glucosamine, fatty acids, and glycerol in molar ratios to phosphorus of 0.35 to 0.69, 0.07 to 0.15 to 0.43, 0.06 to 0.11, and 0.95 to 1.18, respectively, whereas the other group (group B), Bacillus coagulans and Bacillus megaterium, had diacyl lipoteichoic acids which contained D-galactose, fatty acids, and glycerol in molar ratios to phosphorus of 0.05 to 0.42, 0.06 to 0.12, and 0.96 to 1.07, respectively. After treatment with 47% hydrogen fluoride, the lipoteichoic acids obtained from group A strains commonly gave a hydrophobic fragment, gentiobiosyl-beta (1----1 or 3)diacylglycerol, in addition to dephosphorylated repeating units, glycerol, 2-D-alanylglycerol, N-acetyl-D-glucosaminyl-alpha (1----2)glycerol, and D-alanyl-N-acetyl-D-glucosaminyl-alpha (1----2)glycerol, whereas the lipoteichoic acids from group B strains yielded diacylglycerol in addition to glycerol and D-galactosyl-alpha (1----2)glycerol. The results together with data from Smith degradations indicate that in the lipoteichoic acids of group A strains the polymer chains, made up of partially alanylated glycerol phosphate and glycosylglycerol phosphate units, are joined to the acylglycerol anchors through gentiobiose. However, in the lipoteichoic acids of group B strains, the partially galactosylated poly(glycerolphosphate) chains are believed to be directly linked to the acylglycerol anchors.     

A new hereditary single band variant of the Gc system

Summary A new single band variant (Gc Ar) or the Gc subtypes not identical with the known Gc variants has been detected in the plasma of a healthy blood donor by isoelectric focusing. Using this technique the variant is represented by a single band which has a similar isoelectric point to the Gc 1C2 anodal band. It is well known that the single band Gc phenotypes remain unaltered after neuraminidase treatment. Nevertheless, the new single band variant (Gc Ar) is altered after neuraminidase treatment as is Gc 2A3. After neuraminidase treatment, the Gc Ar band is affected and moved to the nearby position of the Gc 2 band. Investigation of the proband's family shows that the variant occurs combined with the common alleles Gc 1F, Gc 1S and that it has an autosomal dominant inheritance.     

Formation of Chlorophyll-Protein Complexes in Greening Cucumber Cotyledons in Light and then in Darkness

The changes in chlorophyll-protein complexes (CPs) in cucumbercotyledons during illumination and subsequent dark incubationwere studied by SDS-polyacrylamide gel electrophoresis. Whenetiolated cucumber seedlings were illuminated, chlorophyll wassynthesized and CPs were formed. In the early phase of greening(6 h of illumination), light-harvesting chlorophyll a/b-proteincomplex (LHCP) was the main GP. As the greening proceeded, P700chlorophyll a-protein complex (CP1) accumulated. When 6-h illuminatedseedlings were transferred to darkness, CP1 accumulated concomitantlywith a decrease in LHCP without new chlorophyll synthesis. Thechanges in the amounts of CPs in the dark became smaller withthe progress of greening and were not observed after 72 h ofillumination. These changes were confirmed by examining thechlorophyll/P700 ratio and the low temperature absorption spectrumof cotyledons. These results suggest that in the early phaseof greening, CPs were unstable and their chlorophyll moleculeseasily exchanged with those of other kinds of CPs. Sodium- and Calcium-Dependent Spike Potentials in the Secretory Neuron Soma of the X-Organ of the Crayfish

Membrane characteristics of neuron somata in the medulla terminalis ganglionic X-organ of crayfish have been investigated with intracellular glass microelectrodes. The soma membrane developed action potentials with 10–20 mv of overshoot. Delayed rectification appeared at 10–20 mv above resting membrane potential. In 50% of the neuron somata examined, action potentials were observed in Na-free medium or TTX medium. The peak potential level of the spike in these media depended on the extracellular concentration of Ca ion. It increased with the Ca concentration. In low calcium media, the peak potential level of the spike varied with Na concentration. Action potentials of the X-organ-sinus gland tract disappeared after bathing in Na-free or TTX medium, suggesting that the conductive action potential was dependent on Na ions. From these results, it is concluded that there are two systems in the neuron soma, one of which responds to the Na ion and the other, to the Ca ion. Inhibitory innervation of the X-organ by the cerebral ganglion was manifested by IPSP's when the optic peduncle was stimulated. A postulated connection between the Ca-dependent spike and the release of hormone in X-organ neuron somata is discussed.
