DNA 5' to the human myelin basic protein (MBP) gene, mapped to 18q22----qter, is known to manifest multiallelic DNA length variation with heterozygosity of at least 45%. Isolation of genomic DNA containing the MBP gene first exon and its 5' flanking region reveals that this polymorphism arises from a 994-bp region of the diverged tandem repeat (TGGA)249. This sequence is located from 1082 to 2075 bp upstream of the MBP initiator methionine. The repetitive sequence is 18% diverged from (TGGA)249 and from analysis of higher order subsequence reiterations appears to have undergone extensive recombination. The pattern of higher order repetition suggests that multiple crossover and gene conversion events have occurred within a 1.0-kb region. Molecular clones of this sequence represent essentially the longest allelic form of this region seen in Southern transfer analysis. This repetitive DNA is similar to a sequence 5' to the human myoglobin gene.  相似文献

Previous studies have analysed the process of Toxoplasma gondii egress with the aid of inducers, such as calcium ionophores. Although calcium transients have been successful in triggering T. gondii egress, the structural panorama of “natural” and artificial events should match. The present study approaches the natural egress of this parasite using super-resolution and electron microscopy and reveals lytic and non-lytic events of individual egress; this corroborates the use of calcium ionophore as a reliable tool to trigger parasite egress. Altogether, our data suggest that different signalling routes can converge to similar structural aspects in natural and induced egress.  相似文献

 The Waxy gene (Wx) encodes the granule-bound starch synthase responsible for the synthesis of amylose in rice (Oryza sativa). Recently, a polymorphic microsatellite sequence closely linked to the Wx gene was reported. To determine whether polymorphism in this sequence correlates with variation in apparent amylose content, we tested an extended pedigree of 92 current and historically important long-, medium- and short-grain US rice cultivars representing the efforts of many breeders over more than 80 years. Seven Wx microsatellite alleles were identified which together explained 82.9% of the variation in apparent amylose content of the 89 non-glutinous rice cultivars tested. Similar results were also obtained with 101 progenyof a cross between low- and intermediate-amylose breeding lines. An additional, unique microsatelliteallele, (CT)16, was detected in one glutinous cultivar,CI 5309. However, the other glutinous cultivars,Calmochi 101 and Tatsumi mochi, were in the (CT)17 class along with three other cultivars that contained15–16.5% amylose. We sequenced a 200-bp PCR-amplified fragment containing the CT microsatellite and the putative 5′ splice site of the Wx leader intron from a subset of 42 cultivars representing all eight microsatellite alleles. All of the cultivars with 18% or less amylose had the sequence AGTTATA at the putative leader intron 5′ splice site, while all cultivars with a higher proportionof amylose had AGTTATA. This single nucleotidesubstitution could also be assayed by AccI digestion of the amplified fragment. Overall, this single nucleotide polymorphism could explain 79.7% of the variation in the apparent amylose content of the 89 non-glutinous cultivars tested. Interestingly, cultivars in the (CT)19 microsatellite classes that differed substantially in amylose content still showed the correlation between this G-T polymorphism and apparent amylose content. The G-T polymorphism at this site was not, however, able to explain the very low amylose contents of the three glutinous cultivars tested, all of which had the sequence AGTTATA. The major purpose of this article is to construct a plausible emissions profile for the European chemical industry from process data and mass balance considerations.' In it we describe this industry and its major conversion processes and emissions. Four major process chains, beginning with methane, ethylene, propylene, and benzene are analyzed, along with five important stand-alone processes. A self-consistent version of the industry is constructed for 1992, based on data from a variety of sources.
In 1992 Europe consumed 9,297 metric kilotons as measured by weight of chlorine (kMT[CI]) of salt and 2 I I kMT(CI) of recycled hydrochloric acid (HCI) to produce 86 I0 kMT of virgin elemental chlorine, plus 278 kMT(CI) of virgin by-product HCI. Total chlorine input to the industry was 8,689 kMT including I2 kMT(CI) of recycled chlorinated hydrocarbons (CHCs) and (net) 79 kMT(CI) of HCI. Shipments of chlorine and HCI to other sectors was 1,367 kMT(CI), while 7,322 kMT(CI) was embodied in products or lost within the sector: Of this subtotal, 350 kMT(CI) was used to manufacture identified inorganic chemicals, 5,694 kMT(CI) for identified organic chemicals, and 1,278 kMT(CI) for "other unspecified" chemicals.
We estimate that products account for 41.6% of inputs (measured at the "fence"), while wastes account for 24.7%  相似文献

S ummary . Membrane filtration has become an accepted method for enumerating Escherichia coli in water, but little published evidence could be found to judge the specificity of the method to assess faecal contamination in either fresh or saline waters. The method is used in our laboratory to monitor the extent and degree of sewage pollution in coastal areas, but there is need for information on what proportion of lactose-fermenting colonies from seawater, developing at 44° on a 4% enriched Teepol medium, are E. coli type I. A total of 1352 colonies from seawater was tested for production of indole and for gas from lactose at 44°. In addition, 46% of the colonies were screened by the IMVEC series of tests. The proportion of colonies tested ranged from 10–100%, depending on the number of colonies on the membrane. Many of the colonies (81.9%) to which IMVEC tests were applied were E. coli type I; a further 10.9% were Irregular type I. The practical implications of these findings are discussed.  相似文献

Seven strains of Penicillium viridicatum isolated from country-cured ham produced citrinin in potato dextrose broth and on country-cured ham. None of the strains produced detectable amounts of citrinin at 10 C. The optimal temperature range for citrinin production was 25 to 30 C.  相似文献

The chromosome complement and patterns of heterochromatin distribution (as demonstrated by the DNA d-r method) were studied from three different guinea pigs. Karyotype analyses showed that one of the females had a heteromorphic sex pair formed by a submetacentric X chromosome and a subterminal X chromosome originated by a shortening of the short arm (x-chromosome). The heterochromatin was mainly found in the pericentromeric areas of the autosomes and X chromosomes and in the short arm of pair 7. The Y chromosome exhibited a degree of heterochromatinization different from that of pericentromeric areas.—The analysis of the heterochromatin distribution in the X chromosomes showed that the smaller size of the heteromorphic x-chromosoine was probably due to a lack of heterochromatin in its short arm. Moreover, two out of the three animals studied had a heteromorphic pattern of heterochromatinization in the pair 21 characterized by heterochromatinization of the pericentromeric area in one chromosome and almost complete heterochromatinization of the other homologue.—It is suggested that most of the heterochromatin disclosed by the DNA d-r method is formed by repetitious DNA; and that the Y chromosome and perhaps some autosome regions in guinea pigs are formed by a type of heterochromatin with properties different from those of the constitutive and facultative heterochromatin (intermediate heterochromatin).Supported in part by NIH Grant 5-501-RR05672-02 and by NIH contract 70-2299.  相似文献

稻米直链淀粉是在由蜡质基因Wx编码的颗粒结合淀粉合成酶(GBSS)的催化下合成的。最近,在Wx基因的区段内发现了一段多态性微卫星序列(CT)n。对74个非糯籼稻和粳稻材料的(CT)n多态性进行了分析,并探讨了其与直链淀粉含量之间的关系。在74个品种(系)中共发现7种(CT)n片段(Wx等位基因),即(CT)8,(CF)10,(CT)11,(CT)16,(CT)17,(CT)18,(Ch)19。在籼粳亚种间,不同(CT)n的分布存在差异较大:在籼稻中,以(CT)11和(CT)18为主,占92.6%,另有(CT)10和(CT)8各2份,(CT)17型1份;在粳稻中,以(CT)16、(CT)17为主,共占20份材料中的90.0%。在上述74个品种(系)中,以(CT)n表示的Wx基因型对稻米直链淀粉含量的决定系数R2达0.912,也即Wx基因型差异可解释这些材料直链淀粉含量变异的91.2%。还发现6份籼稻材料Wx座位上为杂合的(CT)18/(CT)11,其中2份为推广早籼优质品种浙9248和舟优903,并对其在遗传和育种研究中的意义作了探讨。  相似文献
