Biotic threats to trees often arise from interactions among two or more species, frequently insects and fungi, that function together to defeat host defenses, secure resources and colonize new hosts. Feedbacks among plant enemies can have large effects on host population and disease dynamics, either by promoting stabilizing negative feedbacks or contributing to positive feedbacks that can destabilize populations and permit outbreaks. Feedbacks can be rapid and direct (e.g. within trees or among years) or can arise from slowly developing changes in host resource quantity or quality at the scale of forest stands or landscapes. Climate may also influence system dynamics by altering feedbacks within or among species or through density independent effects. We evaluated major drivers of population dynamics of beech bark disease (BBD), an important forest disease in eastern deciduous forests of North America, using data from 28 study sites in the eastern United States monitored for up to 14 yr between 1979 and 1992. Both primary causal agents of BBD – the introduced felted beech scale Cryptococcus fagisuga and native fungi Neonectria spp. – showed strong simple density dependence in all study populations. Surprisingly, densities of scale insects and fungi had little or no effects on population growth rates of the other, despite their habit of living in close physical relationships. For both insects and fungi, ecologically important features of the density dependent functions (slope, carrying capacity and density independent variance) were variable across sites. Climatic effects on density‐dependent functions (and scatter around them) were evident but generally weak and variable. The most striking predictor of patterns in density dependence was duration since establishment of BBD in the region. Apparently BBD alters forests over decades in ways that strengthen self‐regulation among causal agents without eliminating or even dramatically reducing host populations.  相似文献

The ecological literature abounds with studies of environmental effects on plant antiherbivore defences. While various models have been proposed (e.g. plant stress, optimal allocation, growth-differentiation balance), each has met with mixed support. One possible explanation for the mixed results is that constitutive and induced defences are differentially affected by environmental conditions. In this study, constitutive oleoresin flow from Pinus tadea was least during periods of rapid tree growth and most when drought conditions limited growth; this is as expected if constitutive secondary metabolism is a function of the carbohydrate pool size after growth has been maximised. Induced increases in resin flow, however, were greatest in the fastest growing trees during the season of greatest growth. Apparently, resin production becomes an allocation priority after wounding but not before. Understanding environmental effects on plant antiherbivore defences requires physiological and evolutionary models that account for the differences between constitutive and induced secondary metabolism.  相似文献

Background  

Development, differentiation and physiology of metazoans all depend on cell to cell communication and subsequent intracellular signal transduction. Often, these processes are orchestrated via sites of specialized cell-cell contact and involve receptors, adhesion molecules and scaffolding proteins. Several of these scaffolding proteins important for synaptic and cellular junctions belong to the large family of membrane-associated guanylate kinases (MAGUK). In order to elucidate the origin and the evolutionary history of the MAGUKs we investigated full-length cDNA, EST and genomic sequences of species in major phyla.  相似文献

In populations subject to positive density dependence, individuals can increase their fitness by synchronizing the timing of key life history events. However, phenological synchrony represents a perturbation from a population's stable stage structure and the ensuing transient dynamics create troughs of low abundance that can promote extinction. Using an ecophysiological model of a mass-attacking pest insect, we show that the effect of synchrony on local population persistence depends on population size and adult lifespan. Results are consistent with a strong empirical pattern of increased extinction risk with decreasing initial population size. Mortality factors such as predation on adults can also affect transient dynamics. Throughout the species range, the seasonal niche for persistence increases with the asynchrony of oviposition. Exposure to the Allee effect after establishment may be most likely at northern range limits, where cold winters tend to synchronize spring colonization, suggesting a role for transient dynamics in the determination of species distributions.  相似文献

Hepatitis B virus (HBV)-specific T-cell responses are important in the natural history of HBV infection. The number of known HBV-specific T-cell epitopes is limited, and it is not clear whether viral evolution occurs in chronic HBV infection. We aimed to identify novel HBV T-cell epitopes by examining the relationship between HBV sequence variation and the human leukocyte antigen (HLA) type in a large prospective clinic-based cohort of Asian patients with chronic HBV infection recruited in Australia and China (n = 119). High-resolution 4-digit HLA class I and II typing and full-length HBV sequencing were undertaken for treatment-naïve individuals (52% with genotype B, 48% with genotype C, 63% HBV e antigen [HBeAg] positive). Statistically significant associations between HLA types and HBV sequence variation were identified (n = 49) at 41 sites in the HBV genome. Using prediction programs, we determined scores for binding between peptides containing these polymorphisms and associated HLA types. Among the regions that could be tested, HLA binding was predicted for 14/18 (78%). We identified several HLA-associated polymorphisms involving likely known anchor residues that resulted in altered predicted binding scores. Some HLA-associated polymorphisms fell within known T-cell epitopes with matching HLA restriction. Enhanced viral adaptation (defined as the presence of the relevant HLA and the escaped amino acid) was independently associated with HBeAg-negative disease (P = 0.003). Thus, HBV appears to be under immune pressure in chronic HBV infection, particularly in HBeAg-negative disease.  相似文献

Germline stem cells (GSCs) can be used for large animal transgenesis, in which GSCs that are genetically manipulated in vitro are transplanted into a recipient testis to generate donor‐derived transgenic sperm. The objectives of this study were to explore a non‐viral approach for transgene delivery into goat GSCs and to investigate the efficiency of nucleofection in producing transgenic sperm. Four recipient goats received fractionated irradiation at 8 weeks of age to deplete endogenous GSCs. Germ cell transplantations were performed 8–9 weeks post‐irradiation. Donor cells were collected from testes of 9‐week‐old goats, enriched for GSCs by Staput velocity sedimentation, and transfected by nucleofection with a transgene construct harboring the human growth hormone gene under the control of the goat beta‐casein promoter (GBC) and a chicken beta‐globin insulator (CBGI) sequence upstream of the promoter. For each recipient, transfected cells from 10 nucleofection reactions were pooled, mixed with non‐transfected cells to a total of 1.5 × 108 cells in 3 ml, and transplanted into one testis (n = 4 recipients) by ultrasound‐guided cannulation of the rete testis. The second testis of each recipient was removed. Semen was collected, starting at 9 months after transplantation, for a period of over a year (a total of 62 ejaculates from four recipients). Nested genomic PCR for hGH and CBGI sequences demonstrated that 31.3% ± 12.6% of ejaculates were positive for both hGH and CBGI. This study provides proof‐of‐concept that non‐viral transfection (nucleofection) of primary goat germ cells followed by germ cell transplantation results in transgene transmission to sperm in recipient goats.
