This study aimed to investigate potential methane production through anaerobic digestion of dairy manure and co‐digestion with maize silage. Two different anaerobic reactor configurations (single‐stage continuously stirred tank reactor [CSTR] and hybrid anaerobic digester) were used and biogas production performances for each reactor were compared. The HR was planned to enable phase separation in order to improve process stability and biogas production under higher total solids loadings (≥4%). The systems were tested under six different organic loading rates increased steadily from 1.1 to 5.4 g VS/L.d. The CSTR exhibited lower system stability and biomass conversion efficiency than the HR. The specific biogas production of the hybrid system was between 440 and 320 mL/gVS with 81–65% volatile solids (VS) destruction. The hybrid system provided 116% increase in specific biogas production and VS destruction improved by more than 14%. When MS was co‐digested together with dairy manure, specific biogas production rates increased about 1.2‐fold. Co‐digestion was more beneficial than mono‐material digestion. The hybrid system allowed for generating methane enriched biogas (>75% methane) by enabling phase separation in the reactor. It was observed that acidogenic conditions prevailed in the first two compartments and the following two segments as methanogenic conditions were observed. The pH of the acidogenic part ranged between 4.7 and 5.5 and the methanogenic part was between 6.8 and 7.2.  相似文献

Proton conducting biopolymer networks have potential use for bio-sensors. The cost-effective, non-hazardous and environmentally safe biopolymer, such as chitosan, is an attractive feature for bio-sensors. Cholesterol oxidase was immobilized in conducting network via complexation of chitosan with alginic acid. A method for the preparation of the complex along with characterization by elemental analysis, FTIR spectroscopy, TGA and DSC were reported. The proton conductivity chitosan–alginic acid network was studied via impedance spectroscopy under humidified condition. The complex polymer electrolyte with x = 1 exhibited maximum proton conductivity of 1.4 × 10?3 S/cm at RT, RH  50%. The potential use of this network in enzyme immobilization was studied by manufacturing cholesterol oxidase entrapped polymer networks. Additionally, the maximum reaction rate (Vmax) and Michaelis–Menten constant (Km) were investigated for the immobilized cholesterol oxidase. Also, temperature and pH optimization studies were performed, and operational stability and shelf life of the polymer network were examined.  相似文献

Bacteria-induced calcite precipitation (BICP) is a promising technique that utilizes bacteria to form calcite precipitates throughout the soil matrix, leading to an increase in soil strength and stiffness. This research investigated BICP in two types of sands under sterile and nonsterile conditions. Bacteria formation and BICP in the sterilized sand specimens are higher than those in the nonsterilized sand specimens. The development of calcite with time is initially greater for the sand specimens containing calcite. Scanning electron microscope imaging allowed the detection of cementation from calcite precipitation on the surface and pores of the sand matrix. The effects of injecting nutrient mediums and bacteria into the specimens, as well as pH of soil samples on BICP were investigated. The bearing capacity of biologically treated vs. untreated sand specimens were determined especially by laboratory foundation loading tests.  相似文献

Essential thrombocythemia (ET) is an entity of classic Philadelphia chromosome-negative myeloproliferative neoplasms (MPNs), characterized by thrombocytosis with megakaryocytic hyperplasia and thrombocytes are increased with abnormal functions. Discovery of the protein tyrosine kinase JAK2 V617F allele contributed to better understanding of the pathogenetic mechanisms of MPNs. Acquired single point mutation in the JAK2 V617F was determined approximately 50–60 % of patients with ET. In this study we aimed to investigate the relationship between JAK2 V617F gene mutation, hematologic, biochemical markers and the complications in the ET patients. A total of 268 patients diagnosed with ET and 219 of those studied for JAK2 gene mutation were followed at the hematology clinics of three major hospitals between 2008 and 2013 were screened retrospectively. Laboratory, clinical and hematologic parameters were compared for JAK2 V617F positive and JAK2 V617F negative patients with ET. 102 (46 %) patients were positive with the JAK2 V617F mutation. The complications were observed in 61 (28 %) patients and 38 (62 %) of them had JAK2 V617F mutation. The levels of white blood cells, neutrophil, basophil, red blood cells, hemoglobin, hematocrit, mean platelet volume, thrombocytes, eosinophil; urea, creatinine were significantly different in patients with the JAK2 V617F mutation (P < 0.05). Presence of the JAK2 V617F mutation supports the diagnosis of ET. It would be useful to investigate the JAK2 V617F mutation and the hematologic and biochemical markers at diagnosis with respect to consider the risk of developing complications and to take the precautions against these complications.  相似文献

It was shown by gel filtration and viscosity measurements that N‐terminal fragment (FA) of diphtheria toxin (DT) can interact with both G‐ and F‐actin (filamentous actin). Elution profiles on Sephadex G‐100 indicated the formation of a binary complex of fragment A (FA) with globular actin monomer (G‐actin), which was inhibited by gelsolin. Deoxyribonuclease I (DNase I) in turn appeared to interact with this complex. Tritiated FA was found to bind to F‐actin stoichiometrically. This binding was inhibited again by gelsolin and G‐actin, but not by DNase I. The binding of FA inhibited polymerization of G‐actin and induced a time‐dependent breakdown of F‐actin under polymerization conditions. Inhibition of its ADP‐ribosyltransferase activity did not have any effect on the interactions of FA with actin. FA interacted with actin also in the cell. After treatment of human umbilical vein endothelial cells (HUVEC) with biotin‐labeled DT, Western blot analysis revealed predominantly the presence of actin in affinity‐isolated complexes of the labeled FA.
