Bovine collagen alpha-1 is a naturally occurring extracellular matrix protein found in tendons and other connective tissues. It plays
a vital role in cell growth, differentiation, attachment, and migration. Recent findings have established that collagen alpha-1 is
involved in osteogenesis imperfecta phenotype in cattle but deep information about other members of this large family is not
available so far. So with a view to finding a new edge and attempt to figure out a correlation among the well attributed Bovine
alpha-1 collagen sequences are executed and analyzed. To do so, comparative analysis among the 28 members of collagen family
has been carried out using Computational tools. Consequently, based on the physico-chemical, secondary structural, functional
and phylogenetic classifications, we have selected collagen 12, 14 and 20 as targets for pathological conditions. These proteins
belong to the FACIT family and significantly showed low glycine and proline content, high instability and aliphatic index.
Moreover, FACIT family collagens contain multiple triple helical domains and being members of the FACIT family, bovine
collagen 12, 14, 20 do not form fibrils by themselves but they are associated to collagen 1 associated fibrils. These collagen
molecules might be crucial candidates to detect and understand the process of matrix remodeling in diseases especially in the arena

Mycobacterium is gram positive, slow growing, disease causing Actinobacteria. Beside potential pathogenic species, Mycobacterium also contains opportunistic pathogens as well as free living non-pathogenic species. Disease related various analyses on Mycobacterium tuberculosis are very widespread. However, genomic study of overall Mycobacterium species for understanding the selection pressure on genes as well as evolution of the organism is still illusive. MLSA and 16s rDNA based analysis has been generated for 241 Mycobacterium strains and a detailed analysis of codon and amino acid usage bias of mycobacterial genes, their functional analysis have been done. Further the evolutionary features of M. avium complex also have been revealed. Mycobacterial genes are moderately GC rich showed higher expression level in PPs and significant negative correlation with biosynthetic cost of proteins. Translational selection pressure was observed in mycobacterial genes. MAC showed close relationship with NPs and higher evolutionary rate in MAC revealed their constant evolving nature. 相似文献

Rheumatoid arthritis (RA) is a systemic inflammatory autoimmune disorder wherein the contributory role of oxidative stress has been established in the synovial fluid. As availability of synovial fluid is limited, this study aimed to evaluate in the peripheral blood of patients with RA, the relationship if any, between the extent of oxidative stress in terms of generation of reactive oxygen species (ROS) in neutrophils, plasma NADPH oxidase and myeloperoxidase activity with markers of oxidative damage, circulating cytokines and disease activity score (DAS28). In patients with RA, neutrophils in peripheral blood demonstrated an enhanced generation of ROS, coupled with depletion of free radical scavenging activity. Furthermore, the NADPH oxidase and myeloperoxidase activity was enhanced as were markers of damage. There was a positive correlation between the DAS 28 and generation of ROS, NADPH oxidase and myeloperoxidase activity as also with oxidative stress mediated protein carbonylation. Patients with RA demonstrated an increase in proinflammatory (IL-17, IL-23, and IFN-γ) and some anti-inflammatory (IL-4, IL-5, and TGF-β) cytokines. Although the levels of IL-17 correlated positively with generation of ROS, myeloperoxidase, markers of protein damage and DAS28, IL-23 correlated positively only with protein damage, and negatively with free radical scavenging activity. Importantly, incubation of neutrophils from healthy donors with plasma or SF from patients with RA translated into an enhanced generation of ROS, along with an elevation of intracellular proinflammatory cytokines. Taken together, in patients with RA, circulating neutrophils mediated a shift in the oxidant/antioxidant balance favouring the former, which translated into protein damage and contributed towards disease progression. 相似文献

Species of the genusHypencum are of considerable interest worldwide because of their medicinal properties.In- vitro culture is a useful tool for both multiplication of the genus and studying its economically important secondary metabolites.
Here, we present an effectivein- vitro propagation method forH. bupleuroides. Leaf and internodal expiants excised from 9-week-old,in vitro-germinated seedlings were cultured on a Murashige and Skoog (MS) medium supplemented with benzyladenine (BA; 1.0 or 0.1 mg
L-1) and 2,4-dichlorophenoxyacetic acid (2,4-D; 1.0 or 0.1 mg L1). Depending on the BA and 2,4-D combination used, these cultures produced adventitious shoot buds directly on the surfaces
of both types of explants as well as excessive calli. Numerous shoots were obtained when the calli from both expiant types
were cultured on an MS medium supplemented with 2 mg L-1 BA. Internodal expiants were more responsive than leaf tissues to direct and indirect plant regeneration. After shoots that
regenerated from either the calli or the expiant surface were excised, rooting was best on an MS medium lacking any growth
hormones. These rooted plants were then acclimatized under greenhouse conditions, and 90% of regenerants had survived. Ours
