This study was conducted to determine reciprocal effects of low to high doses of nitrogenous fertilizer (N30, N40, N50, N60 and N70 — 30, 40, 50, 60 and 70 kg ha−1 respectively) and CO2 enriched environment on C and N partitioning in soybean (Glycine max (L.) Merril cv JS-335). Plants were grown from seedling emergence to maturity inside open top chambers under ambient, AC (350±50 mol mol−1) and elevated, EC (600±50 mol mol−1) CO2 and analyzed at seedling, vegetative, flowering, pod setting and maturity stages. Soybean responded to both CO2 enrichment and N supply. Leaves, stem and root reserves at different growth stages were analyzed for total C and N contents. Consistent increase in the C contents of the leaf, stem and root was observed under EC than in AC. N contents in the different plant parts were found to be decreased under EC-grown plants specially at seedling and vegetative stage despite providing N doses to the soil. Significant increase observed for C to N dry mass ratio under EC in the root, stems and leaves at seedling and vegetative stage was decreased in the middle and later growth stages possibly due to combined impact of N doses to the soil and increased N2 fixing activities due to EC conditions. Critical analysis of our findings reveals that the composition and partitioning of C and N of soybean under variable rates of N supply and CO2 enrichment alter according to need under altered metabolic process. These changes eventually may lead to alteration in uptake of not only N but other essential nutrients also under changing atmosphere.  相似文献

Mammalian cell line generation typically includes stable pool generation, single cell cloning and several rounds of clone selection based on cell growth, productivity and product quality criteria. Individual clone expansion and phenotype-based ranking is performed initially for hundreds or thousands of mini-scale cultures, representing the major operational challenge during cell line development. Automated cell culture and analytics systems have been developed to enable high complexity clone selection workflows; while ensuring traceability, safety, and quality of cell lines intended for biopharmaceutical applications. Here we show that comprehensive and quantitative assessment of cell growth, productivity, and product quality attributes are feasible at the 200–1,200 cell colony stage, within 14 days of the single cell cloning in static 96-well plate culture. The early cell line characterization performed prior to the clone expansion in suspension culture can be used for a single-step, direct selection of high quality clones. Such clones were comparable, both in terms of productivity and critical quality attributes (CQAs), to the top-ranked clones identified using an established iterative clone screening approach. Using a complex, multi-subunit antigen as a model protein, we observed stable CQA profiles independently of the cell culture format during the clonal expansion as well as in the batch and fed-batch processes. In conclusion, we propose an accelerated clone selection approach that can be readily incorporated into various cell line development workstreams, leading to significant reduction of the project timelines and resource requirements.  相似文献

The present paper presents results of the study in removal of iron, arsenic and total coliform from drinking water using single-pass constructed soil filter (CSF). Results indicated that arsenic levels ranged from 0.5 to less than 10 μg l?1 levels; iron from 5 to less than 0.3 mg l?1 and coliform from 10?5 to less than 5 CFU/100 ml. The results revealed very high removal efficiency, i.e., over 99% and water quality as per WHO standard.  相似文献

An experiment was conducted to study the potential of chromium (Cr) phytoaccumulatory capabilities of four tree species viz., Anogeissus latifolia, Terminalia arjuna, Tecomella undulata, and Salvadora persica Possibility of enhancement of Cr uptake by citric acid and vesicular arbuscular mycorrhizal fungi (VAM) amendments were also tried. Cr is a major pollutant of the environment. Chromium can exist in oxidation states from III to VI, but the most stable and common forms of Cr are trivalent and hexavalent species. Cr(VI) was more toxic to the tree growth in terms of collar diameter (CD) increment in all the tree species than Cr(lll). Roots accumulated more Cr than shoots in all the tree species. There was more than 10 fold increase in root Cr content in comparison with shoot Cr content in all the trees at all the concentration of Cr and all sources of Cr. Citric acid significantly increased the Cr content in the tissues of roots in all the species under both speciation of Cr. The highest increase in Cr content brought by 20 mM citric acid addition was in A. latifolia Results suggest that Anogeissus latifolia is a potential Cr accumulator with citric acid as soil amendment.  相似文献

Rubinstein-Taybi syndrome (RSTS), a developmental disorder comprising abnormalities that include mental retardation, an unusual facial appearance, broad thumbs and big toes is frequently associated with molecular lesions in the CREB-binding protein gene, CREBBP. The objective of the present study was to identify and analyse CREBBP mutations in Indian RSTS patients on which there are no data. Direct sequencing of CREBBP performed in 13 RSTS patients identified the three zinc fingers (CH1, CH2, CH3) and HAT domain as mutational hotspots in which ten novel pathogenic mutations were localized. Functional analysis revealed that three of these mutations affecting amino acids Glu1459, Leu1668 and Glu1724 were critical for histone acetyltransferase activity. Twenty-eight novel CREBBP single-nucleotide polymorphisms (SNPs) were also identified in the Indian population. Linkage disequilibrium studies revealed associations between (i) SNP (rs129974/c.3836-206G>C) and mutation (p.Asp1340Ala); (ii) (rs130002) with mutation (p.Asn435Lys) and (iii) SNPs rs129974, rs130002 and SNP (c.3836-206G>C) signifying a disease affection status. In conclusion, the present study reports the highest detection rate of CREBBP mutations (76.9%) in RSTS patients to date, of which ten are predicted to be pathogenic and three critical for histone acetyltransferase activity. Moreover, identification of the association of CREBBP polymorphisms with disease susceptibility could be an important risk factor for the pathogenesis of RSTS.  相似文献

Cyclic nucleotides are well-known second messengers involved in the regulation of important metabolic pathways or virulence factors. There are six different classes of nucleotide cyclases that can accomplish the task of generating cAMP, and four of these are restricted to the prokaryotes. The role of cAMP has been implicated in the virulence and regulation of secondary metabolites in the phylum Actinobacteria, which contains important pathogens, such as Mycobacterium tuberculosis, M. leprae, M. bovis and Corynebacterium, and industrial organisms from the genus Streptomyces. We have analysed the actinobacterial genome sequences found in current databases for the presence of different classes of nucleotide cyclases, and find that only class III cyclases are present in these organisms. Importantly, prominent members such as M. tuberculosis and M. leprae have 17 and 4 class III cyclases, respectively, encoded in their genomes, some of which display interesting domain fusions seen for the first time. In addition, a pseudogene corresponding to a cyclase from M. avium has been identified as the only cyclase pseudogene in M. tuberculosis and M. bovis. The Corynebacterium and Streptomyces genomes encode only a single adenylyl cyclase each, both of which have corresponding orthologues in M. tuberculosis. A clustering of the cyclase domains in Actinobacteria reveals the presence of typical eukaryote-like, fungi-like and other bacteria-like class III cyclase sequences within this phylum, suggesting that these proteins may have significant roles to play in this important group of organisms.  相似文献

The acidic amino acids (Asp, Glu) and their amides (Asn, Gln) support rapid growth of a variety of Pseudomonas strains when provided as the sole source of carbon and nitrogen. All key enzymes of glutamate metabolism were detected in P. fluorescence, with glutaminase and asparaginase showing the highest specific activities. A periplasmic glutaminase/asparaginase activity (PGA) was found in all pseudomonads examined, including a number of root-colonizing biocontrol strains. The enzyme was purified and shown to be identical with the ansB gene product described previously. In addition to PGA, P. fluorescens contains a cytoplasmic asparaginase with marked specificity for Asn. PGA is strongly and specifically induced by its substrates (Asn, Gln) but also by the reaction products (Asp, Glu). In addition, PGA is subject to efficient carbon catabolite repression by glucose and by citrate cycle metabolites. A mutant of P. putida KT2440 with a disrupted ansB gene was unable to utilize Gln, whereas growth of the mutant on other amino acids was normal.  相似文献
