To characterize the carbohydrate moieties of the insulin receptor on IM-9 lymphocytes, the cells were surface iodinated and solubilized, and the insulin receptors were precipitated with anti-receptor antibody. The precipitates were resuspended, subjected to either enzymatic digestion or chemical treatment with trifluoromethanesulfonic acid and the relative mobilities of the alpha and beta subunits before and after treatment were analyzed by polyacrylamide gel electrophoresis and autoradiography. The results indicate that the alpha subunit possesses primarily N-linked carbohydrate which is both complex (Endoglycosidase F sensitive) and polymannose (Endoglycosidase H sensitive). The beta subunit also contains polymannose oligosaccharide units and has, in addition, a substantial amount of carbohydrate which is removed by chemical treatment but is not susceptible to Endoglycosidase F, suggesting the presence of O-linked saccharides. The apparent molecular weights of the core protein of the mature alpha and beta subunits as determined by gel electrophoresis following complete deglycosylation are 98 kDa and 80 kDa, respectively.  相似文献

  1. Pollination syndromes refer to stereotyped floral characteristics (flower colour, shape, etc.) that are associated with a functional group of pollinators (bee, bird, etc.).
  2. The trumpet creeper Campsis radicans, endemic to the southeast and mid‐west United States, has been assigned to the hummingbird‐pollination syndrome, due mainly to its red, trumpet‐shaped flowers.
  3. Previous studies demonstrated that the ruby‐throated hummingbird Archilochus colubris is C. radicans' primary pollinator, but anecdotal data suggest various bee species may provide pollination service when hummingbirds are absent.
  4. This study characterised C. radicans nectar volume and concentration by time of day. Nectar volume was suitable for hummingbirds, but concentration was higher than typical hummingbird‐pollinated plants (~20% w/w); at ~30% w/w, it approached the concentration expected in bee‐pollinated plants (~50% w/w). We also found substantial amounts of nectar at night.
  5. Two C. radicans populations received virtually no hummingbird visits, but the number of bees were markedly higher than in the populations previously described. Interestingly, there were no night‐time visitors despite the large quantity of nocturnal nectar.
  6. Based on previously published pollen delivery per visit by various species, this study estimated that cumulative deposition by bees routinely reached pollen deposition thresholds for setting fruit in C. radicans. They are, unequivocally, the predominant pollinators in these populations, thus providing pollination service in the absence of hummingbirds.
  7. These results highlight C. radicans as a food source for native bees and add to the understanding of how floral phenotypes can facilitate pollination by disparate functional groups.

Surface waters from paired agricultural watersheds under controlled tile drainage (CTD) and uncontrolled tile drainage (UCTD) were monitored over 7 years in order to determine if there was an effect of CTD (imposed during the growing season) on occurrences and loadings of bacterial and viral pathogens, coliphages, and microbial source tracking markers. There were significantly lower occurrences of human, ruminant, and livestock (ruminant plus pig) Bacteroidales markers in the CTD watershed in relation to the UCTD watershed. As for pathogens, there were significantly lower occurrences of Salmonella spp. and Arcobacter spp. in the CTD watershed. There were no instances where there were significantly higher quantitative loadings of any microbial target in the CTD watershed, except for F-specific DNA (F-DNA) and F-RNA coliphages, perhaps as a result of fecal inputs from a hobby farm independent of the drainage practice treatments. There was lower loading of the ruminant marker in the CTD watershed in relation to the UCTD system, and results were significant at the level P = 0.06. The odds of Salmonella spp. occurring increased when a ruminant marker was present relative to when the ruminant marker was absent, yet for Arcobacter spp., the odds of this pathogen occurring significantly decreased when a ruminant marker was present relative to when the ruminant marker was absent (but increased when a wildlife marker was present relative to when the wildlife marker was absent). Interestingly, the odds of norovirus GII (associated with human and swine) occurring in water increased significantly when a ruminant marker was present relative to when a ruminant marker was absent. Overall, this study suggests that fecal pollution from tile-drained fields to stream could be reduced by CTD utilization.  相似文献

The binding characteristics of the oligopeptide chemoattractant receptor on guinea pig macrophages and macrophage membrane preparations were characterized using detailed binding studies and computer analysis. Viable macrophages bound the radiolabeled chemoattractant N-formyl-methionyl-leucyl-[3H]phenylalanine with single dissociation constant (KD) of 18.4 +/- 4.6 nM with 15,300 +/- 1,800 sites per cell. Binding data from membrane preparations indicated the presence of two classes of binding sites with KD of 1.5 +/- 0.4 nM and 25.5 +/- 11.0 nM. Approximately 23% of the receptors were in the high affinity state. In the presence of added guanine nucleotide di- or triphosphates, the high affinity receptors in the membrane preparations were converted to low affinity states with no change in the total receptor number. Nonhydrolyzable derivatives of GTP were most potent in converting the receptor from its high to low affinity state. These data suggest that the affinity state of the oligopeptide chemoattractant receptor in macrophages is regulated by guanine nucleotides and GTPase, implying that the transduction mechanisms of this receptor may be controlled by a guanine nucleotide regulatory unit.  相似文献

Intraspecific phenotypic variation between populations separated by large geographic distances is common. Differences in the mean and variance of traits among populations can be used to infer the relative strength, direction, and type of selection on traits. Patterns in the mean provide information on the type of selection, and patterns in variance provide information on the strength of selection. However, interpretation of mean/variance patterns is difficult when two traits are linked and strongly correlated to fitness because it is unlikely that each trait will reach phenotypic optima. In amphibians time to metamorphosis and size at metamorphosis are positively related both phenotypically and genetically. Using a common-garden experiment we investigated whether selection favours shorter time to metamorphosis or increased mass at metamorphosis between two populations which differ in the length of the post-metamorphic growing season by 2–4 weeks. Animals from the population a shorter growing season took longer to reach and metamorphosed at a greater mass, while animals from the population with a longer period for post metamorphic growth reached metamorphosis faster, but at a smaller mass. Greater phenotypic variance was observed in both traits in the population with the shorter growing season. These data suggest that animals from the population with a restricted growth period maximise mass at metamorphosis at the expense of longer larval periods while animals from population with the longer post-metamorphic growth period sacrifice mass at metamorphosis to shorten the larval period and maximise larval survival. Differences in phenotypic variance among populations suggest either directional or diversifying selection has acted on both traits.  相似文献

A strain of Tetranychus urticae from pears was evaluated for cyhexatin and fenbutatin-oxide resistance after field control failure to the latter occurred. High-level fenbutatin-oxide resistance was confirmed in the field strain. Fenbutatin-oxide resistance was >1,500 fold (based on flowable powder) and 127 fold (based on emulsifiable concentrate). However, the level of cyhexatin resistance was only 9.8 fold which was similar to levels previously found. This provides a stark warning against the overuse of fenbutatin-oxide.  相似文献
