人体小卫星DNA探针的制备

根据人体小卫星DNA核心顺序,化学合成长23碱基寡核苷酸探针,筛选人体基因组文库,旨在获得能用作遗传分析探针的小卫星顺序。结果得到15个含小卫星的阳性重组子。随机取其一(C_(35.9))作探针,试做群体分析。所有个体均可检出多条杂交带。其中某些带具有多态性。在一定检测条件下,检出的DNA图谱在有限的个体内具有个体特异性。结果表明筛选文库得到的小卫星顺序可用于小卫星多态性的检测。其它小卫星探针的筛选和应用性研究正在进行。     

Recognition of the P1 plasmid centromere analog involves binding of the ParB protein and is modified by a specific host factor.

The P1 plasmid partition system is responsible for segregation of daughter plasmids during division of the Escherichia coli host cell. The P1-encoded elements consist of two essential proteins, ParA and ParB, and the cis-acting incB region. The incB region determines partition-mediated incompatibility and contains the centromere-like site parS. We have isolated and purified the two proteins. ParB binds specifically to the incB region in vitro. DNase I footprinting assays place a strong binding site over the 35-bp parS sequence previously shown to be sufficient for partition when the Par proteins are supplied in trans. A weaker site lies within the incB region in sequences that are important for specifying incompatibility, but are not essential for partition. Gel band retardation assays show that a host factor binds specifically to the incB sequence. The factor strongly stimulates binding of ParB. Cutting the region at a site between the two ParB binding sites yields two fragments that can bind ParB but not host factor. Thus, information for host-factor binding lies in the region determining the specificity of plasmid incompatibility. The roles of parB and the host factor in partition and the specificity of plasmid incompatibility are discussed.     

c-kit protein, a transmembrane kinase: identification in tissues and characterization.

The proto-oncogene c-kit encodes a transmembrane kinase which is related to the receptors for colony-stimulating factor type 1 and platelet-derived growth factor, as well as to the immunoglobulin superfamily. Antibodies specific for the kinase domain of the P80 gag-kit protein of the Hardy-Zuckerman 4 feline sarcoma virus were prepared. These kit-specific antibodies were used to identify and characterize the c-kit protein in cat brain tissue. The c-kit protein product displays an autophosphorylating activity in immune complex kinase assays, and, in turn, this activity was used to identify the c-kit protein in different tissues. In cat brain, a single 145-kilodalton (kDa) glycoprotein was detected. Its N-linked carbohydrates were found to be sensitive to digestion with the endoglycosidases (neuraminidase, endoglycosidase F, and endoglycosidase H), indicating hybrid and/or complex and high-mannose structures. A partial purification of the c-kit protein was achieved by wheat germ agglutinin affinity chromatography, and the autophosphorylating activity of the partially purified c-kit protein was characterized and found to be specific for tyrosine. The kit antibodies cross-react with the murine c-kit protein product, and variant c-kit proteins in different mouse tissues were identified, with sizes of about 145 kDa (brain), 160 kDa (spleen), and 150 kDa (testis).     

Fate of bacteria ingested by larvae of the freshwater mayfly,Ephemera danica

The fate of bacteria in the food of a common freshwater invertebrate has been studied both in controlled laboratory experiments and in a stream sediment. The animal chosen was the larva of the burrowing mayfly,Ephemera danica. It ingested all available bacteria nonselectively. More bacteria were found associated with the hindgut than with the mesenteron despite continuous plug flow of food through the alimentary canal. Species of bacteria were affected in different ways.Aeromonas hydrophila andCitrobacter freundii were both digested, the former selectively.Flavobacterium sp. and other unidentified species appeared to attach to the hindgut wall. Digestion of bacteria was not due to a sudden change in pH.     

Dinocrocuta gigantea头骨的发现

本文记述了甘肃和政县晚中新世地层中发现的一个完整的巨鬣狗头骨;讨论了它的分类地位,修正了 Schlosser 1903年建种时的一些鉴定错误.巨鬣狗是鬣狗科中一个十分特化的成员,应代表一个独立的属.根据命名规则,我们采用 N. schmidt-Kittler 1976年所创的 Dinocrocuta. 因此,这个种应订正为 Dinocrocuta gigantea (schlosser, 1903).     

广西大戟科植物资料

<正> 广西的大戟科植物种类众多。作者对广西中草药原植物研究过程,将已采集的标本及华南植物所标本室(SCBI)和广东、广西主要标本室(均有注明)收藏的广西及其邻近地区的大戟科标本进行整理;现报道(一)、三宝木属植物摘记;有5种和3个新变种,其中广西产5种、1变种。(二)、广西白大凤属植物;1种。     

鲥鱼的驯养与生物学研究Ⅰ.O+龄幼鱼的生长与食性

本文主要描述池塘养殖条件下鲥鱼幼鱼的生长与食性。1)1987-1989年8月至11月幼鱼生长迅速,快于同期珠江天然个体的生长速度;2)体长26.6-94.9mm的幼鱼主要摄食浮游动物,轮虫和浮游植物出现率随个体增大而减少。食物共有18个属种,摄食频度为100%;3)幼鱼的平均饱满指数为75.70‰。各类食物重量百分比的顺序为:桡足类>枝角类>无节幼体>虾类溞状幼体>轮虫>藻类。     

咽喉部高频喷射通气调节各参数对狗肺通气效率的影响

用相关和回归处理方法,研究了8条正常狗咽喉部高频喷射通气时,调节驱动压、呼吸比和频率对喷气量、吸入气氧浓度、动脉血气及气道内压的作用。结果显示,驱动压和呼吸此对各观察指标几乎有同等重要的作用,频率的影响很小,喷气量与吸入气氧浓度、动脉血气、气道内压间存在显著的正相关关系。说明调节参数的意义主要在于改变了喷气量。     

大果木姜子的甘油三酯组成

采用光谱法和胰酯酶分解法对大果木姜子油酯中分得的结晶C—I组成进行了分析,结果表明C—I是由11种甘油三酯组成。其中2-位为月硅酸的甘油三酯占91.80％,1,3-位脂肪酸主要为癸酸和月桂酸,甘油三酯的组成中,CLC占62.24％,LLC占24.50％。
