The Bcl-2 family of proteins: regulators of cell death and survival   总被引:15,自引:0,他引:15  
The Bcl-2 protein inhibits apoptosis induced by a variety of signals, in a range of cell types and in diverse organisms, and it is implicated in both normal development and oncogenesis. Despite this central role, the mechanism of action of Bcl-2 is not yet clear. Recent studies have uncovered a number of Bcl-2-related gene products that regulate apoptosis either negatively or positively, and Bcl-2 forms heterodimers with at least one of these proteins, Bax. This article discusses the role of the Bcl-2 family of proteins in the light of these findings.  相似文献

Amphibians and reptiles are sensitive to changes in the thermal environment, which varies considerably in human-modified landscapes. Although it is known that thermal traits of species influence their distribution in modified landscapes, how herpetofauna respond specifically to shifts in ambient temperature along forest edges remains unclear. This may be because most studies focus on local-scale metrics of edge exposure, which only account for a single edge or habitat patch. We predicted that accounting for the combined effect of multiple habitat edges in a landscape would best explain herpetofaunal response to thermally mediated edge effects. We (1) surveyed herpetofauna at two lowland, fragmented forest sites in central Colombia, (2) measured the critical thermal maximum (CTmax) of the species sampled, (3) measured their edge exposure at both local and landscape scales, and (4) created a thermal profile of the landscape itself. We found that species with low CTmax occurred both further from forest edges and in areas of denser vegetation, but were unaffected by the landscape-scale configuration of habitat edges. Variation in the thermal landscape was driven primarily by changes in vegetation density. Our results suggest that amphibians and reptiles with low CTmax are limited by both canopy gaps and proximity to edge, making them especially vulnerable to human modification of tropical forest. Summary Candida utilis var. major NRRL-Y-1084 was grown in a defined medium without a phosphorous (P) source. During the exponential phase, cells divided according to a specific growth rate of 0.32 h-1, which is lower than the usual rate for a balanced medium (0.4–0.6 h-1). The relative P content of the biomass decreased from 2.70% to 0.75% over a period of 6 h, including 2 h of cell division arrest. At the end of this period there was another interruption of cell division. After that, multiplication restarted at a considerably lower rate and it deviated slightly from the exponential pattern. The stationary phase began when biomass P content reached 0.4%–0.5%, slowly decreasing afterwards to 0.25–0.20%. Biomass synthesis was less affected than cell division by the relative decrease of endogenous P, the two processes differing partially in their kinetics. Cell lysis started shortly before the stationary phase and affected about 20% of the population by the end of the assay. RNA and P content of the resulting biomass were 2.4% and 0.25% respecitvely, P being mainly incorporated to RNA.The relationship of biomass production to glucose uptake was very low, probably because the marked P deficiency called for an increase in energy consumption for growth and specially for maintenance. Ferritin as a source of iron was consiered. A good iron absorption rate appears in normal rats with an in vivo absorption technique. The same absorption appears in iron-deficient animals. The iron stored in intestinal wall is lower in anemic rats than in normal ones, suggesting a higher draw of iron from lumen to blood.  相似文献

Summary The effect of sampling conditions on the levels of adenine nucleotides, pyridine nucleotides, glycolytic intermediates and related metabolites in yeast has been studied. A systematic examination of the conditions for harvesting has shown that it can be best accomplished by rapid filtration. Delays in the handling for removal of the medium, as is usual in the process of obtaining a number of data reported in the literature, lead to important changes in some of the metabolites examined. It is also shown that when a washing is imperative it can be carried out with a methanol-water mixture (50/50, v/v) cooled at –40° without loss of intracellular concentrations of non-readily diffusible metabolites.On the basis of this experience the outline of a generally applicable procedure is presented.  相似文献

Permeabilization induced by lacticin 3147, lactococcins A, B and M, enterocin AS-48 and nisin, bacteriocins described as cell membrane-pore forming and lytic agents, enhanced in all cases aldehyde formation by Lactococcus lactis IFPL730. Nevertheless, the conversion of isoleucine into 2-methylbutyraldehyde depended not only on the degree of permeabilization but also on the bacteriocin that caused the cell membrane damage. The highest values of 2-methylbutyraldehyde corresponded to cell suspensions containing lacticin 3147 and lactococcins, treatments that provoked further lysis in addition to induced permeabilization.  相似文献

Immunoelectrotransfer blot assay in acute and chronic human trichinellosis   总被引:3,自引:0,他引:3  
An immunoelectrotransfer blot assay (IETB) using excretory secretory products of muscle larvae of Trichinella spiralis (ML-ESP) and the avidin biotin system was developed in order to characterize reactivity against ML-ESP in sera from patients with acute and chronic trichinellosis. A complete pattern of up to 13 bands was developed by sera from individuals with trichinellosis where doublets, triplets, or single bands were shown to have molecular weights of roughly 66, 55, 45, 36, 29, 24, and 14 kDa. The bands at approximately 55, 36, 29, and 14 kDa proved specific for T. spiralis. The band at approximately 55 kDa was present in all trichinellosis sera, whereas the approximately 14-kDa band was present in only a small percentage of sera. The development of approximately 36- and 29-kDa bands suggests a modulation of the reactivity against ML-ESP over time. IETB proved more sensitive for the population of chronic trichinellosis under study than a conventional diagnostic enzyme-linked immunosorbent assay, allowing negative or borderline serum samples to be determined. Thus, this technique, when applied for human trichinellosis surveillance, should provide a useful tool in endemic areas.  相似文献
