The herbicides amiprophos-methyl (APM) and oryzalin disrupt mitosis and cytokinesis in plant cells by causing the depolymerization of microtubules. These drugs have also been shown to affect calcium sequestration by mitochondria. Controversy thus exists as to whether microtubule depolymerization occurs as a result of direct interaction between the drug and tubulin, or because of elevated intracellular calcium levels resulting from drug interference with calcium regulation. In order to clarify this issue we have directly measured the effect of these herbicides and other cell-motility-altering drugs on intracellular calcium levels in stamen-hair cells of Tradescantia. The results indicate that low levels (1–3 M) of APM and oryzalin can act within 3–7 min causing disorganization of mitosis. Studies using the calcium indicator indo-1 injected into stamen-hair cells to monitor internal levels of calcium, show that at drug concentrations where inhibitory effects on mitosis and-or cytokinesis are clearly seen, APM, oryzalin, isopropyl-N-phenyl carbamate, caffeine and cytochalasin D produce no change in intracellular calcium levels. Furthermore, except for cytochalasin D, these drugs do not inhibit cytoplasmic streaming, a calcium-sensitive process. Male Sprague-Dawley rats were treated by intratracheal instillation with 1 mg/animal of refractory ceramic fibers. Intratracheal exposure to ceramic fibers led to significant changes of immune response. Results of proliferative activity of spleen lymphocytes showed significantly decreased proliferative activity of T-cells in response to mitogens phytohemagglutinin and concanavalin A in animals given ceramic fibers in comparison with control rats. Similarly, T-dependent B-cell response to pokeweed mitogen was significantly suppressed. Spontaneous proliferative activity of lymphocytes in non-stimulated spleen cell cultures did not differ in exposed and control rats. No significant changes were found among groups in percentage of phagocytic blood polymorphonuclear leukocytes and percentage of cells with respiratory burst.  相似文献

Respiratory syncytial virus (RSV) causes respiratory disease in young children, the elderly, and immunocompromised individuals, often resulting in hospitalization and/or death. After more than 40 years of research, a Food and Drug Administration-approved vaccine for RSV is still not available. In this study, a chimeric bovine/human (b/h) parainfluenza virus type 3 (PIV3) expressing the human PIV3 (hPIV3) fusion (F) and hemagglutinin-neuraminidase (HN) proteins from an otherwise bovine PIV3 (bPIV3) genome was employed as a vector for RSV antigen expression with the aim of generating novel RSV vaccines. b/h PIV3 vaccine candidates expressing native or soluble RSV F proteins were evaluated for efficacy and immunogenicity in a nonhuman primate model. b/h PIV3 is suited for development of pediatric vaccines since bPIV3 had already been evaluated in clinical studies in 1- and 2-month-old infants and was found to be safe, immunogenic, and nontransmissible in a day care setting (Karron et al., Pediatr. Infect. Dis. J. 15:650-654, 1996; Lee et al., J. Infect. Dis. 184:909-913, 2001). African green monkeys immunized with b/h PIV3 expressing either the native or soluble RSV F protein were protected from challenge with wild-type RSV and produced RSV neutralizing and RSV F-protein specific immunoglobulin G serum antibodies. The PIV3-vectored RSV vaccines evaluated here further underscore the utility of this vector system for developing safe and immunogenic pediatric respiratory virus vaccines.  相似文献

Vibration is a common mechanical phenomenon and possesses mechanical energy in ambient environment, which can serve as a sustainable source of power for equipment and devices if it can be effectively collected. In the present work, a novel soft and robust triboelectric nanogenerator (TENG) made of a silicone rubber‐spring helical structure with nanocomposite‐based elastomeric electrodes is proposed. Such a spring based TENG (S‐TENG) structure operates in the contact‐separation mode upon vibrating and can effectively convert mechanical energy from ambient excitation into electrical energy. The two fundamental vibration modes resulting from the vertical and horizontal excitation are analyzed theoretically, numerically, and experimentally. Under the resonant states of the S‐TENG, its peak power density is found to be 240 and 45 mW m?2 with an external load of 10 MΩ and an acceleration amplitude of 23 m s?2. Additionally, the dependence of the S‐TENG's output signal on the ambient excitation can be used as a prime self‐powered active vibration sensor that can be applied to monitor the acceleration and frequency of the ambient excitation. Therefore, the newly designed S‐TENG has a great potential in harvesting arbitrary directional vibration energy and serving as a self‐powered vibration sensor.  相似文献

The goal of this research was to elucidate the relationship between WHO 2016 molecular classifications of newly diagnosed, nonenhancing lower grade gliomas (LrGG), tissue sample histopathology, and magnetic resonance (MR) parameters derived from diffusion, perfusion, and 1H spectroscopic imaging from the tissue sample locations and the entire tumor. A total of 135 patients were scanned prior to initial surgery, with tumor cellularity scores obtained from 88 image-guided tissue samples. MR parameters were obtained from corresponding sample locations, and histograms of normalized MR parameters within the T2 fluid-attenuated inversion recovery lesion were analyzed in order to evaluate differences between subgroups. For tissue samples, higher tumor scores were related to increased normalized apparent diffusion coefficient (nADC), lower fractional anisotropy (nFA), lower cerebral blood volume (nCBV), higher choline (nCho), and lower N-acetylaspartate (nNAA). Within the T2 lesion, higher tumor grade was associated with higher nADC, lower nFA, and higher Cho to NAA index. Pathological analysis confirmed that diffusion and metabolic parameters increased and perfusion decreased with tumor cellularity. This information can be used to select targets for tissue sampling and to aid in making decisions about treating residual disease.  相似文献

Spatial variation in the strength of trophic cascades in arctic tundra has been related to flows of subsidies across ecosystem boundaries. Here, we ask whether the input of marine subsidies in tundra systems would cause spatial variation in the strength of rodent–plant interactions between coastal areas, where predators have access to marine‐derived resources, and non‐subsidized inland areas of northern Fennoscandia. We present a detailed evaluation of predator–rodent–vegetation interactions along a coast‐inland gradient, during the 2011 rodent outbreak and the two following decline years, by using direct assessments of rodent impacts and tracing of marine‐derived nutrients in the food web. Our results revealed that the main rodent predator during summer, the long‐tailed jaeger Stercorarius longicaudus, did not benefit from marine resources while breeding (relative dietary proportion in chicks’ diet = 0–3%). Contrary to this pattern, parasitic jaegers S. parasiticus, bred exclusively near the coast and preyed effectively on both marine resources (41% of chicks’ diet) and rodents (12%). Mammalian predators also showed a higher activity during winter near the coast. Despite overall higher predator numbers, no evidence was found for lower rodent population growth rates during the three monitoring summers and for weaker rodent grazing impacts in the coastal area. Instead, we documented pronounced damages caused by lemmings and voles on bryophytes and vascular plants, especially dwarf shrubs (e.g. Vaccinum myrtillus) all along the coast–inland gradient. Taken together, our results did not support the hypothesis that marine subsidies would trigger a trophic cascade in coastal tundra areas of northern Fennoscandia during a major rodent outbreak, probably due to a relatively low diversity of marine‐subsidized predators in the region. Comparative observational and experimental studies at large spatial scales in various arctic regions are absolutely necessary for a better understanding of factors causing regional variations in the functioning of arctic food webs.  相似文献

Objectives

While early diagnosis of dementia is important, the question arises whether general practitioners (GPs) should engage in direct referrals. The current study investigated current referral practices for neuroimaging in dementia, access to imaging modalities and investigated related GP training in Ireland and North Wales.

Methods

A questionnaire was distributed to GPs in the programme regions which included approximately two thirds of all GPs in the Republic of Ireland and all general practitioners in North Wales. A total of 2,093 questionnaires were issued.

Results

48.6% of Irish respondents and 24.3% of Welsh respondents directly referred patients with suspected dementia for neuroimaging. Irish GPs reported greater direct access to neuroimaging than their Welsh counterparts. A very small percentage of Irish and Welsh GPs (4.7% and 10% respectively) had received training in neuroimaging and the majority who referred patients for neuroimaging were not aware of any dementia-specific protocols for referrals (93.1% and 95% respectively).

Conclusions

The benefits of direct GP access to neuroimaging investigations for dementia have yet to be established. Our findings suggest that current GP speciality training in Ireland and Wales is deficient in dementia-specific and neuroimaging training with the concern being that inadequate training will lead to inadequate referrals. Further training would complement guidelines and provide a greater understanding of the role and appropriateness of neuroimaging techniques in the diagnosis of dementia.  相似文献

Two different phytoplankton sampling methods (bottle and net sampling) were used to evaluate the concentration of toxicDinophysis species in seawater and their correlation to mussel toxicity, assessed by mouse bioassay.Dinophysis concentration in net samples revealed the higher correlation to mussel toxicity (r=0.75,p<0.01). Net sampling therefore seems more suitable for the detection of low abundance species likeDinophysis characterised by vertical aggregations at different depths in the water column.  相似文献
