In order to determine the chromosomal locations of an autoimmune antigen, the coilin gene and its pseudogene, we amplified the segments of the two genes by the polymerase chain reaction (PCR) and screened a panel of somatic cell hybrids for the presence of the gene products. The results indicate that the human coilin gene and its pseudogene can be assigned to chromosome 17 and chromosome 14, respectively. Further analysis of cell hybrids bearing chromosome 17 with various deletions localized the coilin gene to the region q21–q23.  相似文献

Animals make decisions based on subjective assessments of their environment. To determine their future foraging activities, animals probably assess food availability from past foraging experiences. Thus, foraging also functions as a way for animals to collect information, with the uncertainty of an assessment decreasing as foraging activity increases. This suggests that different needs for a correct assessment may affect the investment made in foraging activities. Orb‐web spiders sometimes relocate their webs and relocation rate differs among species. After web relocation, several spider species have been reported to construct the first webs at newly occupied web sites using less silk than usual, possibly to avoid the risk of an overinvestment at sites where food availability has not been determined. Nevertheless, they may pay a cost, because of inadequate decision‐making, if webs constructed with less silk convey less information and increase the uncertainty of an assessment. We expect that stronger site tenacity necessitates a greater requirement for correct assessment of web site and the degree to which spiders reduce the amount of web silk in the first web after web relocation is smaller in species that use the same site longer. To test this hypothesis, we examined web construction in two orb‐web spiders, Cyclosa octotuberculata and C. argenteoalba. At the same time we found that these two species exhibit different web‐site tenacity, as C. octotuberculata does not relocate its webs as frequently as does C. argenteoalba. After artificially induced web relocation, C. argenteoalba constructed webs that were initially smaller and contained only about 2/3 of the silk in control webs that were constructed at the original site. In contrast, C. octotuberculata did not exhibit such decreases in web size or in the amount of web silk used. This result is consistent with our hypothesis.  相似文献

The nuclear 18S‐rRNA gene has been used as a metabarcoding marker in massively parallel sequencing (MPS)‐based environmental surveys for plankton biodiversity research. However, different hypervariable regions have been used in different studies, and their utility has been debated among researchers. In this study, detailed investigations into 18S‐rRNA were carried out; we investigated the effective number of sequences deposited in international nucleotide sequence databases (INSDs), the amplification bias, and the amplicon sequence variability among the three variable regions, V1–3, V4–5 and V7–9, using in silico polymerase chain reaction (PCR) amplification based on INSDs. We also examined the primer universality and the taxonomic identification power, using MPS‐based environmental surveys in the Sea of Okhotsk, to determine which region is more useful for MPS‐based monitoring. The primer universality was not significantly different among the three regions, but the number of sequences deposited in INSDs was markedly larger for the V4–5 region than for the other two regions. The sequence variability was significantly different, with the highest variability in the V1–3 region, followed by the V7–9 region, and the lowest variability in the V4–5 region. The results of the MPS‐based environmental surveys showed significantly higher identification power in the V1–3 and V7–9 regions than in the V4–5 region, but no significant difference was detected between the V1–3 and V7–9 regions. We therefore conclude that the V1–3 region will be the most suitable for future MPS‐based monitoring of natural eukaryote communities, as the number of sequences deposited in INSDs increases.  相似文献

Summary Preinduction of potent haptenic muramyl dipeptide (MDP)-reactive helper T cell activity and subsequent immunization with MDP hapten-coupled syngeneic tumor cells resulted in enhanced induction of tumor-specific immunity through T-T cell collaboration between anti-MDP hapten helper T cells and tumor-specific effector T cells. The present study establishes two types of tumor-specific immunotherapy protocols utilizing helper T cells against MDP hapten cross-reactive with Bacillus Calmette Guérin (BCG). In the first model, naive normal C3H/He mice or mice in which MDP hapten-reactive helper T cells had been generated by BCG-sensitization were inoculated i.d. with syngeneic X5563 tumor cells. When both groups of mice were allowed to generate MDP hapten-modified tumor cells in the tumor mass in situ by intratumoral injection of MDP hapten, an appreciable number of growing tumors in the BCG-presensitized but not in the unsensitized group were observed to regress. In the second model, a growing X5563 tumor mass was removed by the surgical resection 9 days after the tumor implantation. Approximately 90% of C3H/He mice receiving such treatment died from tumor metastasis by about 30 days after the tumor resection. However, immunization of mice with MDP hapten-coupled X5563 tumor cells subsequent to the tumor resection resulted in an increased survival rate. Such protection from the tumor metastasis was appreciably stronger when compared to the protection obtained by immunization with MDP hapten-uncoupled tumor cells. The mice surviving in both models were also demonstrated to retain X5563 tumor-specific immunity. These results indicate that the presentation of MDP hapten-modified tumor cells to BCG-sensitized recipients results in potent tumor-specific immunity which contributes to the regression of the primary tumor or inhibition of metastatic tumor growth.This work was supported by a Grant-in-Aid for the Special Project Cancer Bioscience from the Ministry of Education, Science and Culture, Japan  相似文献

Purification and Characterization of Dehydroascorbate Reductase from Rice   总被引:2,自引:0,他引:2  
Dehydroascorbate reductase (DHAR; EC 1.8.5.1 [EC] ) is an enzyme thatis critical for maintenance of an appropriate level of ascorbatein plant cells. This report describes the purification and characterizationof a GSH-dependent DHAR from rice (Oryza saliva) bran and isthe first, to our knowledge, of such an analysis of DHAR froma monocot. The enzyme was a monomeric thiol enzyme, resemblingDHARs purified from dicots, but it was different from them interms of heat stability and antigenicity. The amino-terminalamino acid sequence of the DHAR from rice did not show any obvioussimilarity to those of known proteins with DHAR activity, suchas, glutaredoxin (thioltrans-ferase), protein disulfide isomerase,and trypsin inhibitor. Immunoprecipitation analysis showed thatthis enzyme was a major DHAR in etiolated seedlings. Westernblot analysis indicated that this enzyme was distributed ubiquitouslyin rice tissues. ? Despite the wide inflorescence diversity among angiosperms, the effects of inflorescence architecture (three-dimensional flower arrangement) on pollinator behaviour and mating success have not been sufficiently studied in natural plant populations. ? Here, we investigated how inflorescence architecture affected inter- and intra-plant pollinator movements and consequent mating success in a field population of Spiranthes sinensis var. amoena (S. sinensis). In this species, the flowers are helically arranged around the stem, and the degree of twisting varies greatly among individuals. The large variation in inflorescence architecture in S. sinensis results from variation in a single structural parameter, the helical angle (the angular distance between neighbour-flower directions). ? The numbers of visits per inflorescence and successive probes per visit by leaf-cutting bees decreased with helical angle, indicating that individual flowers of tightly twisted inflorescences received less visitations. As expected from pollinator behaviour, pollinia removal and fruit set of individual flowers decreased with helical angle. Meanwhile, geitonogamy decreased in tightly twisted inflorescences. ? Our novel findings demonstrate that natural variation in inflorescence architecture significantly affects pollinator behaviour and reproductive success, suggesting that inflorescence architecture can evolve under pollinator-mediated natural selection in plant populations. We also discuss how diverse inflorescence architectures may have been maintained in S. sinensis populations.  相似文献
