Spectrophotometric titrations of slightly substituted carboxymethylamylose (CM-Amy) and diethylaminoethylamylose (DEAE-Amy) with iodine in the presence of iodide (I2/I?) were carried out as a function of iodide concentration, temperature, and polymeric charge. Binding isotherms for the polymer-I2/I? complex are reported in terms of an apparent binding constant (Ka) plotted versus degree of saturation of the complex (θ). The dependence of Ka upon polymeric charge is interpreted as evidence for the negatively charged character of the bound species. The cooperative nature of the binding process is evident in the positive slope of Ka vs (θ). Whereas the apparent binding constants and binding cooperativities for the derivatives are smaller than for the amylose-I2/I? complex, the binding enthalpies deduced from the temperature dependence of Ka at θ = 0.5 appear to be the same for amylose and CM-Amy. A viscometric titration of fully charged CM-Amy with I2/I?, conducted at dialysis equilibrium between the CM-Amy-I2/I? solution and the polymer-free solvent phase, disclosed a maximum in the plot of intrinsic viscosity ([η]) vs θ. The increase in [η] at small θ was interpreted as a reflection of polyelectrolyte expansion provoked by absorption of the negatively charged bound species; the subsequent decline in [η] is attributed to stabilization by I2/I? of compact helical sequences or to the formation at higher θ of intermolecular aggregates.  相似文献

This study reports on the biochemical characterization as well as the kinetic and thermodynamic study of Aspergillus tamarii URM4634 β-fructofuranosidase (FFase) with transfructosylating activity. Conditions for FFase activity were optimized by means of a central composite rotational design using pH and temperature as the independent variables, while residual activity tests carried out in the temperature range of 45–65°C enabled us to investigate FFase thermostability and estimate the kinetic and thermodynamic parameters of enzyme denaturation. Optimal conditions for sucrose hydrolysis and fructosyl transfer catalyzed by crude FFase were 50°C, and pH 6.0 and 7.4, respectively. The thermodynamic properties of irreversible enzyme inactivation were found to be activation energy of 293.1 kJ mol−1, and activation enthalpy, entropy, and Gibbs free energy in the ranges 290.3–290.4 kJ mol−1, 568.7–571.0 J mol−1 K−1, and 97.9–108.8 kJ mol−1, respectively. The results obtained in this study point out satisfactory enzyme activity and thermostability at temperatures commonly used for industrial fructo-oligosaccharide (FOS) synthesis; therefore, this novel FFase appears to be a promising biocatalyst with great potential for long-term FOS synthesis and invert sugar production. To the best of our knowledge, this is the first report on kinetic and thermodynamic parameters of an A. tamarii FFase.  相似文献

Flavescence dorée (FD) is a grapevine disease caused by associated phytoplasmas (FDp) which are epidemically spread by their main vector Scaphoideus titanus. The possible roles of alternative and secondary FDp plant hosts and vectors have gained interest in terms of better understanding of the FDp ecology and epidemiology. The findings of a survey conducted in the surroundings of three vineyards in the southern Swiss Alps aimed at studying the possible epidemiological role of the FDp secondary vector Orientus ishidae and the FDp host plant Alnus glutinosa are reported. This work demonstrates that O. ishidae is able to complete its biological cycle on A. glutinosa and to acquire FDp and 16SrV phytoplasmas very efficiently with an infection rate of 69% for the nymphal instars and 85% for the imagoes. A high prevalence of the map genotype M50 (map type FD1), which is included in the S. titanus—grapevine epidemiological cycle, was found in O. ishidae and A. glutinosa. Additionally, M12 (map type FD3), M44 and M47 were also sporadically detected. Surprisingly, the grapevines tested during this work were all infected by M54 (map type FD2) only, while the few S. titanus caught in the vineyard canopy were all FDp free. In conclusion, the occurrence of infected common alder stands and O. ishidae nearby vineyards do not seem to play a prominent role in FD epidemics in southern Switzerland. Nevertheless, wild vegetation acts as a reservoir of the FDp inoculum, which may locally trigger a FD emergence if S. titanus populations are established inside vineyards.  相似文献

AlphaB-crystallin (HSPB5) is one of the most prominent and well-studied members of the small heat shock protein (sHsp) family. To date, it is known that this protein modulates significant cellular processes and therefore, it is not surprising that its deregulation is involved in various human pathologies, including cancer diseases. Despite the pathogenic significance of HSPB5 in cancer and its regulatory mechanism related to aggressiveness is poorly understood, several reports describe the association of breast carcinoma progression with HSPB5, whose expression is also considered an independent predictor of breast cancer metastasis to the brain. Indeed, numerous authors indicate HSPB5 as a new valuable biomarker for clinicopathological parameters and poor prognosis in breast cancer. Considering the cytoprotective, anti-apoptotic, pro-angiogenic, and pro-metastatic properties of the sHsps, it is not surprising that they are considered as promising targets for anticancer treatment, even though, at present, a deeper understanding of their mode of action is needed to allow the development of precise therapeutic interventions. Data on the direct inhibition of different sHsps demonstrate promising results in cancer pathologies; however, specific strategies against HSPB5 have not been considered. This review highlights the most relevant findings on HSPB5 and its role in breast cancer, as well as the possible strategies in using HSPB5 inhibition for therapeutic purposes.  相似文献

The aim of this investigation was to study the welfare of 3 captive groups of cotton-top tamarins housed in different zoological parks. Ethological observations were conducted during 1 year. In addition, fecal samples were collected and the concentrations of glucocorticoids, androgens, and progestogens were measured. Within each group, no significant differences in fecal cortisol concentrations were found between subjects. The fecal concentrations of testosterone and progesterone significantly differed depending on the sexes and ages of the tamarins. A significant association was found among hormone concentrations, exhibit dimensions, and group composition. A highly significant correlation was found between all hormones considered and the space available for each subject. Significant differences in behavioral patterns were observed among groups, including social–individual, affiliative–aggressive, and anogenital–suprapubic scent marking. Correlations between hormone measurements and behaviors were detected. In conclusion, this study confirmed the associations between some behaviors exhibited by these nonhuman primates and both cortisol and testosterone; these data also highlight the role played by progesterone in these behaviors.  相似文献

Encapsulation in alginate hydrogels has been extensively used for several applications in food, pharmaceutical, and biomedical fields. The rational design of a functional polymer network is based on the identification of key parameters and mechanisms governing rate and extent of release of the immobilized molecular species. In the present work, a calorimetric study of the water evaporation under non-isothermal conditions is aimed at evaluating functional properties of a series of alginate-based gel beads. The experiments show how a number of variables, such as scan rate, calcium and alginate concentration, operational procedures, and addition of biopolymer co-solutes influence the temperature evolution of the water evaporation from beads. Given the simplicity and the rapidity of the calorimetric experiment, the issue is raised that a scaling approach could be reached by using water as reference material for the prediction of the diffusion kinetics of encapsulated molecules of variable size and properties.  相似文献
