Regulated irrigation has the potential to improve crop quality in woody ornamentals by reducing excessive vigour and promoting a more compact habit. This research aimed to compare the effectiveness and the mode of action of two techniques, regulated deficit irrigation (RDI) and partial root drying (PRD), when applied to container-grown ornamentals through drip irrigation. Results showed that RDI and PRD reduced growth in Cotinus coggygria 'Royal Purple', but in Forsythia x intermedia 'Lynwood', significant reductions were recorded only with RDI. Physiological measurements in Forsythia indicated that reductions in stomatal conductance (gs) occurred in both treatments, but those in the RDI tended to be more persistent. Reduced gs in PRD was consistent with the concept that chemical signals from the root can regulate stomatal aperture alone; however, the data also suggested that optimising the growth reduction required a moderate degree of shoot water deficit (i.e. a hydraulic signal to be imposed). As RDI was associated with tissue water deficit, it was used in a second experiment to determine the potential of this technique to precondition container-grown plants against subsequent drought stress (e.g. during retail stages or after planting out). Speed of acclimation would be important in a commercial context, and the results demonstrated that both slow and rapid imposition of RDI enabled Forsythia plants to acclimate against later drought events. This article discusses the potential to both improve ornamental plant quality and enhance tolerance to subsequent adverse conditions through controlled, regulated irrigation.  相似文献

Anoplophora glabripennis (Motschulsky) (Coleoptera: Cerambycidae), an invasive pest quarantined in the United States, is difficult to detect because the larvae feed unseen inside trees. Acoustic technology has potential for reducing costs and hazards of tree inspection, but development of practical methods for acoustic detection requires the solution of technical problems involving transmission of resonant frequencies in wood and high background noise levels in the urban environments where most infestations have occurred. A study was conducted to characterize sounds from larvae of different ages in cambium, sapwood, and heartwood of bolts from three host tree species. Larval sounds in all of the tested trees and tissues consisted primarily of trains of brief, 3-10-ms impulses. There were no major differences in the spectral or temporal pattern characteristics of signals produced by larvae of different ages in each tissue, but larval sounds in sapwood often had fewer spectral peaks than sounds in cambium and heartwood. A large fraction, but not all background sounds could be discriminated from larval sounds by automated spectral analyses. In 3-min recordings from infested bolts, trains containing impulses in patterns called bursts occurred frequently, featuring 7-49 impulses separated by small intervals. Bursts were rarely detected in uninfested bolts. The occurrence of bursts was found to predict infestations more accurately than previously used automated spectral analyses alone. Bursts and other features of sounds that are identifiable by automated techniques may ultimately lead to improved pest detection applications and new insight into pest behavior.  相似文献

Complement appears to play a dual role in the progression of systemic lupus erythematosus, serving a beneficial role in enhancing immune complex clearance, while serving a pathogenic role in inducing local inflammation. To investigate these different roles of complement in a therapeutic setting, MRL/lpr mice were treated with the targeted murine C3 complement inhibitor, CR2-Crry, from 16 to 24 wk of age (after the development of proteinuria). The targeting moiety, CR2, binds to C3 breakdown products deposited at sites of complement activation and has the potential to provide complement inhibition locally without causing systemic inhibition. Administration of CR2-Crry i.v., at a dose of 0.25 mg once a week, was associated with a significant survival benefit, improved kidney function, and a significant reduction in glomerulonephritis and renal vasculitis. The presence of skin lesions and lung bronchiolar and vascular inflammation was also dramatically reduced by CR2-Crry treatment. CR2-Crry treatment also resulted in a significant reduction in autoantibody production, as measured by anti-dsDNA Ab levels, and did not cause an increase in circulating immune complex levels. These effects on autoimmunity and circulating immune complexes represent significant potential advantages over the use of Crry-Ig in MRL/lpr mice, a systemic counterpart of CR2-Crry. CR2-Crry localized preferentially to the kidneys in 16-wk MRL/lpr mice with a kidney-localized half-life of approximately 24 h. Thus, targeted complement inhibition at the C3 level is an effective treatment in murine lupus, even beginning after onset of disease.  相似文献

Abstract. 1. The gall wasp Biorhiza pallida (Hymenoptera: Cynipidae) reproduces by cyclical parthenogenesis. The adults of the sexual generation develop within galls (oak apples) that contain many larval cells.
2. Folliot [(1964) Annales Des Sciences Naturelles: Zoologie, 12, 407–564] found asexual generation females to be of three reproductive types. Androphores produce only sons, gynophores produce only daughters, and gynandrophores produce both sons and daughters. In nature, most oak apples give rise to either only males or only females but a proportion produces both sexes. These mixed‐sex galls could result either from eggs laid by one or more gynandrophores or from eggs laid by androphores and gynophores developing within a single gall (multiple founding).
3. Here the frequency of mixed‐ and single‐sex galls was quantified, and morphological and genetic analyses were carried out on the adults emerging from 10 galls to determine the frequency of multiple founding in B. pallida .
4. Seventy-five per cent of 627 galls yielded only one sex. The majority of the remaining 25% had a highly skewed sex ratio. Low genetic variation in B. pallida limited the application of allozyme-based genetic techniques, however seven of the 10 galls analysed in detail, including mixed-sex galls, appeared to have been multiply founded. Contributions by the different foundresses in multiply founded galls were highly skewed.
5. The significance of multiple founding is discussed in the light of possible adaptive scenarios (reduction of parasitoid-induced mortality, avoidance of local stochastic extinction and inbreeding) and possible competition for oviposition sites.  相似文献

Over the last 30 yr, the loss of traditional crop rotations and the polarisation of pastoral and arable farming have led to a marked reduction in mixed agriculture in Britain at both the individual farm and the landscape scale. We assess the potential impact of this change on lowland farmland birds by examining the extent to which distributions (as shown by national atlases of summer and winter birds) of different species are associated with arable, pastoral or mixed‐farming landscape types. Relatively few species appeared as either widespread generalists, equally associated across the three broad farmland types, or specialists associated only with one landscape type. Most were associated with two farmland types but distributions showed seasonal differences. During the breeding season there was an approximate ratio of 2:2:1 of species associated with arable, mixed and pastoral landscapes. However, in winter, most species were at highest abundance in mixed farming landscapes. This coincided with a reduction in the number of species associated with pastoral landscapes relative to the breeding season, whereas the number of species associated with arable landscapes remained relatively stable. These patterns are likely to be related to foraging requirements; granivorous birds tended to be associated with arable habitats, which tend to be more seed‐rich, and invertebrate‐feeding species tended to be associated with mixed or pastoral ones, which may be more invertebrate‐rich. The relative importance of mixed landscapes in winter was attributed to seasonal changes in the distribution of some invertebrate feeders, particularly small insectivores (pipits, chats and wagtails), and some of those species considered to be widespread in summer. The results have two important implications for future research. First, most research on farmland birds has, to date, focused on species associated with arable farming during the breeding season. More research is required on species associated with agricultural grasslands, reflecting its importance for farmland birds in Britain, and on the winter ecology of farmland birds in general. These results also emphasise the importance of mixed habitats for farmland birds, particularly in winter, and suggest that further changes in agricultural practices causing reduction in habitat diversity will be detrimental for the farmland bird community. However, widespread benefits may be derived from small scale measures that increase habitat diversity within farmland, for example field margin management options within agri‐environment schemes and support for traditional farming practices in some Environmentally Sensitive Areas.  相似文献

Background

CD25, a component of the IL-2 receptor, is important in T cell proliferation, activation induced cell death, as well as the actions of both regulatory (Treg) and effector (Teff) T cells. Recent genome wide association studies have implicated the CD25 locus as an important region for genetic susceptibility to a number of autoimmune disorders, with serum levels of soluble CD25 receptor (sCD25) serving as a potential phenotypic marker for this association. However, the functional impact of CD25 cleavage, as well as the influence of sCD25 on immunoregulatory activities, remain largely unknown and form the basis of this effort.

Methodology/Principal Findings

The generation of sCD25 by Treg (CD4+CD25+) and Teff (CD4+CD25) cells was examined during in vitro suppression assays, efforts that demonstrated constitutive and stable surface CD25 expression on Treg throughout the period of in vitro assessment. In contrast, Teff cells increased CD25 expression during the process of in vitro suppression, with supernatant sCD25 levels correlating to the amount of cellular proliferation. Interestingly, under serum-free conditions, Tregs partially lost their characteristic anergic and suppressive properties. sCD25 supplementation at physiological concentrations to serum free in vitro suppression assays reduced Teff proliferation without specifically influencing suppression. Indeed, sCD25 production within these cultures correlated with cell death.

Conclusions/Significance

These results support the notion that sCD25 functions as both a surrogate marker of T cell activation as well as an indicator of subsequent cellular death. In addition, the role of CD25 in immunomodulation is likely dependent on the local inflammatory milieu, with molecules capable of modulating surface CD25 expression playing a key role in defining immune responsiveness.  相似文献

Two dimensional polyacrylamide gel electrophoresis (2D PAGE) is used to identify differentially expressed proteins and may be applied to biomarker discovery. A limitation of this approach is the inability to detect a protein when its concentration falls below the limit of detection. Consequently, differential expression of proteins may be missed when the level of a protein in the cases or controls is below the limit of detection for 2D PAGE. Standard statistical techniques have difficulty dealing with undetected proteins. To address this issue, we propose a mixture model that takes into account both detected and non-detected proteins. Non-detected proteins are classified either as (a) proteins that are not expressed in at least one replicate, or (b) proteins that are expressed but are below the limit of detection. We obtain maximum likelihood estimates of the parameters of the mixture model, including the group-specific probability of expression and mean expression intensities. Differentially expressed proteins can be detected by using a Likelihood Ratio Test (LRT). Our simulation results, using data generated from biological experiments, show that the likelihood model has higher statistical power than standard statistical approaches to detect differentially expressed proteins. The copepod Boeckella poppei is a major species in high latitude lakes of the Southern Hemisphere. In such lakes the reduced diversity of metazoans contrasts with a rich microbial assemblage, making these systems amenable to the study of predation controls on the microbial food web. However, the diet of B. poppei is subject to conflicting reports, with little information on feeding rates. We incubated this species in water from Sombre Lake, a much-studied maritime Antarctic Lake on the South Orkney Islands, in order to quantify its feeding rates and potential impact on the microbial assemblage. Overall, clearance rates were similar across 4 experiments spanning November 1999–March 2000, but increased with prey size over the range of 2.7–18 μm equivalent spherical diameter (esd). B. poppei fed omnivorously, although small phototrophic flagellates comprised the bulk of the diet because of their overwhelming dominance in the incubation water. Larger motile preys—heterotrophic ciliates of ~18 μm esd—were cleared fastest (mean 555 ml mg−1 dry mass day−1) and at equivalent rates to those found for freshwater and marine copepods of similar size and at similar temperatures. Estimated predation impact on the microbial food web varied with the abundance of copepods; these were ~30-fold greater in March than in December. In March even the relatively abundant B. poppei (1.7 adults l−1) had a negligible impact on nanoflagellates, due to the low clearance rate on these small cells. However, in March, B. poppei adults were estimated to clear 24% of the lake water of ciliates daily. Given the generation time of ciliates (1.6 days measured in a previous summer study), and the fact that other larval stages of B. poppei were not assessed, this species has the potential to control this part of the microbial assemblage in Sombre Lake.  相似文献
