The Familial Mediterranean Fever (FMF) shows an autosomal recessive pattern of inheritance and affects certain ethnic groups. Disease is caused by mutations in MEFV gene and more than 180 mutations have been defined in affected individuals. Current study aimed to determine the frequency-type of the mutations for MEFV gene in Sivas??middle Anatolian city. The cohort was composed of 3340 patients. MEFV gene mutations were studied by multiplex PCR based reverse hybridization stripAssay method. Patients?? clinical features were; family history: 68%, erysipelas-like erythema: 17.6%, fever: 89.9%, abdominal pain: 84.2%, peritonitis: 90.2%, arthritis: 33%, pleuritis: 14.2%, parental consanguinity: 21.2%. Current results revealed that M694V is the most frequent mutation (43.12%), followed by E148Q (20.18), M680I(G/C) (15.00%) and V726A (11.32%). The study population has a high rate of carriers and the E148Q mutation frequency was found to be highest when compared to the other regions of Turkey and other Mediterranean groups. 相似文献

Genetic factors that contribute to the risk of breast cancer are largely not known and association studies have revealed several
genes with low penetrance risk alleles for breast cancer. Analysis of these genes may provide important information on the
risk factors affecting carcinogenesis. Variations in the ARLTS1, RAD51 and MDM2 genes have been associated with increased
risk of different cancer types but for breast cancer the results are not consistent. In this study we investigated the role
of the allelic variants in candidate genes acting in the tumor suppressor, DNA repair and p53 pathways as risk factors for
familial breast cancer in 147 patients displaying characteristics of familial disease. Presence of the polymorphic variants
were investigated by amplification of the corresponding regions and restriction fragment length polymorphism analysis. Genotype
and allele frequencies in the patients were significantly different for all three variants. Our results indicate that the
polymorphic variants might affect individual susceptibility towards breast cancer. 相似文献

Hypermethylated genomic DNA is a common feature in tumoral tissues, although the prevalence of this modification remains poorly understood. We aimed to determine the frequency of five tumor suppressor (TS) genes in prostate cancer and the correlation between promoter hypermethylation of these genes and low and high grade of prostate carcinomas. A total of 30 prostate tumor specimens were investigated for promoter methylation status of TS hypermethylated in cancer 1 (HIC1), death-associated protein kinase 1 (DAPK1), secreted frizzled-related protein 2 (SFRP2), cyclin-dependent kinase inhibitor 2A (p16), and O-6-methylguanine-DNA methyltransferase (MGMT) genes by using bisulfite modifying method. A high frequency of promoter hypermethylation was found in HIC1 (70.9%), SFRP2 (58.3%), and DAPK1 (33.3%) genes in tumor samples that were examined. The current data show high frequency of hypermethylation changes in HIC1, SFRP2, and DAPK1 genes in prostate carcinomas of high Gleason Score (GS). 相似文献

Background
Embolization of atherosclerotic debris from the rupture of a vulnerable atherosclerotic plaque occurs iatrogenically during percutaneous coronary interventions (PCI) and can induce myocardial necrosis. These microembolizations are detected as high intensity transient signals (HITS) using intracoronary Doppler technology.Presentation of the hypothesis
In the presented study we will test if abciximab (ReoPro?) infusion reduces high intensity transient signals in patients with stable angina pectoris undergoing PCI in comparison to standard therapy alone.Testing the hypothesis
The High Intensity Transient Signals ReoPro? (HITS-RP) study will enroll 60 patients. It is a prospective, single center, randomized, double-blinded, controlled trial. The study is designed to compare the efficacy of intravenous abciximab administration for reduction of microembolization during elective PCI. Patients will be randomized in a 1:1 fashion to abciximab or placebo infusion. The primary end point of the HITS-RP-Study is the number of HITS during PCI measured by intracoronary Doppler wire. Secondary endpoints are bleeding complications, elevation of cardiac biomarkers or ECG changes after percutaneous coronary interventions, changes in coronary flow velocity reserve, hs-CRP elevation, any major adverse cardio-vascular event during one month follow-up.Implications of the hypothesis
The HITS-RP-Study addresses important questions regarding the efficacy of intravenous abciximab administration in reducing microembolization and periprocedural complications in stable angina pectoris patients undergoing PCI.Trial registration

A hierarchical in silico screening procedure using the crystal structure of an agonist bound chimeric α7/Ls-AChBP protein was successfully applied to both proprietary and commercial databases containing drug-like molecules. An overall hit rate of 26% (pKi ?5.0) was obtained, with an even better hit rate of 35% for the commercial compound collection. Structurally novel and diverse ligands were identified. Binding studies with [3H]epibatidine on chimeric α7/5-HT3 receptors yielded submicromolar inhibition constants for identified hits. Compared to a previous screening procedure that utilized the wild type Ls-AChBP crystal structure, the current study shows that the recently obtained α7/Ls-AChBP chimeric protein crystal structure is a better template for the identification of novel α7 receptor ligands. 相似文献

We previously identified claudin-2 as a functional mediator of breast cancer liver metastasis. We now confirm that claudin-2 levels are elevated in liver metastases, but not in skin metastases, compared to levels in their matched primary tumors in patients with breast cancer. Moreover, claudin-2 is specifically expressed in liver-metastatic breast cancer cells compared to populations derived from bone or lung metastases. The increased liver tropism exhibited by claudin-2-expressing breast cancer cells requires claudin-2-mediated interactions between breast cancer cells and primary hepatocytes. Furthermore, the reduction of the claudin-2 expression level, either in cancer cells or in primary hepatocytes, diminishes these heterotypic cell-cell interactions. Finally, we demonstrate that the first claudin-2 extracellular loop is essential for mediating tumor cell-hepatocyte interactions and the ability of breast cancer cells to form liver metastases in vivo. Thus, during breast cancer liver metastasis, claudin-2 shifts from acting within tight-junctional complexes to functioning as an adhesion molecule between breast cancer cells and hepatocytes. 相似文献

A morphological comparison is made with the closely related species; V. oreophilum K. Koch and V. pyramidatum M. Bieb. The present study was carried out to determine interactive and comparative effects of salinity and water stress on growth,
proline accumulation, chlorophyll, carotenoid and macro nutrient content and antioxidative enzymes such as superoxide dismutase
(SOD), guaiacol peroxidase (POX), and polyphenol oxidase (PPO) in hydroponically grown maize (Zea mays L.cv DKC647) plants. Plants were treated two salt (NaCl) concentrations and polyethylene glycol 6000 (PEG 6000) to create
water stress. The results obtained from this experiment show that high salinity reduced growth through decreasing shoot and
root dry and fresh weight, chlorophyll, and carotenoid content, but PEG treatment had no significant effect on this parameters.
Under NaCl and PEG 6000 treatment, uptake and translocation of mineral nutrients changed drastically. The high presence of
Na+ in nutrient solution affected considerably the plant nutritional requirement, especially influencing the uptake of Ca2+ and K+, which were restricted for competition. Proline accumulation, and SOD, POX and PPO activities were increased with the increasing
intensity of NaCl stress, but PEG 6000 treatment in addition to NaCl had more significant effect on this enzyme activities.
These results suggest that maize plants may be increased proline content to maintain osmotic adjustment and increased the
activity of antioxidant enzymes to have a better protection against active oxygen species (AOS) under salt and water stress. 相似文献
