The NOD1/2‐RIPK2 is a key cytosolic signaling complex that activates NF‐κB pro‐inflammatory response against invading pathogens. However, uncontrolled NF‐κB signaling can cause tissue damage leading to chronic diseases. The mechanisms by which the NODs‐RIPK2‐NF‐κB innate immune axis is activated and resolved remain poorly understood. Here, we demonstrate that bacterial infection induces the formation of endogenous RIPK2 oligomers (RIPosomes) that are self‐assembling entities that coat the bacteria to induce NF‐κB response. Next, we show that autophagy proteins IRGM and p62/SQSTM1 physically interact with NOD1/2, RIPK2 and RIPosomes to promote their selective autophagy and limit NF‐κB activation. IRGM suppresses RIPK2‐dependent pro‐inflammatory programs induced by Shigella and Salmonella. Consistently, the therapeutic inhibition of RIPK2 ameliorates Shigella infection‐ and DSS‐induced gut inflammation in Irgm1 KO mice. This study identifies a unique mechanism where the innate immune proteins and autophagy machinery are recruited together to the bacteria for defense as well as for maintaining immune homeostasis.  相似文献

Gingivitis is highly prevalent in adults, and if left untreated, can progress to periodontitis. In this article, we present an interesting case study where the resolution of gingivitis was followed over a period of 10 days using optical coherence tomography (OCT) and light-induced autofluorescence (LIAF). We demonstrate that OCT and its functional angiography can distinctively capture the changes during the resolution of gingivitis; while LIAF can detect red-fluorescent signals associated with mature plaque present at the inflamed site. The acute inflammatory region showed evidence of angiogenesis based on the quantification of vessel density and number; while no angiogenesis was detected within the less inflamed region. Gingival thickness showed a reduction of 140 ± 26 μm on average, measured between the peak gingivitis event and the period wherein the inflammation was resolved. Vessels in the angiogenesis site was found to reduce exponentially. The mildly inflamed site showed a decreasing trend in the vessel size, which however was within the error of the measurement.  相似文献

Measuring thermal behavior in smaller insects is particularly challenging. In this study, we describe a new horizontal thermal gradient apparatus designed to study adult thermal behavior in small insects and apply it using D. melanogaster as a model and case study. Specifically, we used this apparatus and associated methodology to examine the effects of sex, geographic origin, and developmental rearing temperature on temperature preferences exhibited by adults in a controlled laboratory environment. The thermal gradient established by the apparatus was stable over diurnal and calendar time. Furthermore, the distribution of adult flies across thermal habitats within the apparatus remained stable following the period of acclimation, as evidenced by the high degree of repeatability across both biological and technical replicates. Our data demonstrate significant and predictable variation in temperature preference for all 3 assayed variables. Behaviorally, females were more sensitive than males to higher temperatures. Flies originating from high latitude, temperate populations exhibited a greater preference for cooler temperatures; conversely, flies originating from low latitude, tropical habitats demonstrated a relative preference for higher temperatures. Similarly, larval rearing temperature was positively associated with adult thermal behavior: low culture temperatures increased the relative adult preference for cooler temperatures, and this response was distinct between the sexes and for flies from the temperate and subtropical geographic regions. Together, these results demonstrate that the temperature chamber apparatus elicits robust, predictable, and quantifiable thermal preference behavior that could readily be applied to other taxa to examine the role of temperature-mediated behavior in a variety of contexts.  相似文献

Success of ex situ storage of germplasm for trade and conservation essentially depends upon the precision of the protocol employed for the assessment of germination potential. Active oxygen species and antioxidative enzymes during natural ageing (NA) and controlled deterioration (CD) was monitored during the loss of seed vigour and germination potential in neem seeds showing intermediate seed storage behaviour. Higher levels of SOD, CAT and APX were strongly and positively associated with germination and vigour. The loss of CAT and APX activity estimated quantitatively and number of isoenzymes were closely accompanied with the simultaneous increase in the amounts of H2O2 and OH-radical. The decline in germination and vigour was negatively related with the levels of H2O2 and OH-radical and enhanced electrolyte leakage. The amounts of OH-radical were positively correlated with the decline in DNA content and DNA damage. The levels of SOD isoenzymes initially increased as the germination index of seeds declined from 5250 to 762 and 882 under NA and CD conditions, respectively. Increasing activity of SOD in the ageing seeds were associated with the accumulation of H2O2. The role of antioxidative enzymes in maintaining signalling and damaging amounts of AOS as well as revelations of different pathways of ageing during NA and CD in the ageing neem seeds were discussed.  相似文献

Changes in the cellular content of three polyamines (putrescine, spermidine and spermine) were compared at different stages of development in zygotic and somatic embryos of Pinus radiata D. Don . During embryo development, both the zygotic and the somatic embryos showed a steady increase in spermidine content, with either a small decrease or no significant change in putrescine. This led to a several-fold increase in spermidine/putrescine ratios during development of both types of embryos. Cell cultures of plant-forming and non-plant-forming lines derived from the same clone and growing on proliferation (maintenance) medium differed significantly in their polyamine levels. Mature, cotyledonary stage somatic embryos capable of germination and formation of plants could be distinguished by their higher spermidine/putrescine ratios from abnormal cotyledonary stage somatic embryos which were incapable of forming plants.  相似文献

Aim We conducted a meta‐analysis of species–area relationships (SARs) by combining several data sets and important covariates such as types of islands, taxonomic groups, latitude and spatial extent, in a hierarchical model framework to study global pattern and local variation in SARs and its consequences for prediction. Location One thousand nine hundred and eighteen islands from 94 SAR studies from around the world. Methods We developed a generalization of the power‐law SAR model, the HSARX model, which allows: (1) the inclusion of multiple focal parameters (intercept, slope, within‐study variance), (2) use of multiple effect modifiers based on a collection of SAR studies, and (3) modelling of the between‐ and within‐study variability. Results The global pattern in the SAR was the average of local SARs and had wide confidence intervals. The global SAR slope was 0.228 with 90% confidence limits of 0.059 and 0.412. The intercept, slope and within‐study variability of local SARs showed great heterogeneity as a result of the interaction of modifying covariates. Confidence intervals for these SAR parameters were narrower when other covariates in addition to area were accounted for, thus increasing the accuracy of the predictions for species richness. The significant effect of latitude and the interaction of latitude, taxa and island type on the SAR slope indicated that the ‘typical’ latitudinal diversity gradient can be reversed in isolated systems. Main conclusions The power‐law relationship underlying the HSARX model provides a good fit for non‐nested SARs across vastly different spatial scales by taking into account other covariates. The HSARX framework allows researchers to explore the complex interactions among SAR parameters and modifying variables, to explicitly study the scale dependence, and to make robust predictions on multiple levels (island, study, global) with associated prediction intervals. From a prediction perspective, it is not the global pattern but the local variation that matters.  相似文献

Our goal was to design nanocarriers that specifically target and deliver therapeutics to polarized macrophages. Mannose receptors are highly overexpressed on polarized macrophages. In this study, we constructed Pluronic® -F127 polymer and tannic acid (TA) based nanoparticles (F127-TA core nanoparticles) with varying mannose densities. The particle size of the optimized mannose-decorated F127-TA hybrid nanoparticles (MDNPs) was found to be ~ 265 nm with a negative zeta potential of ~ ? 4.5 mV. No significant changes in the size and zeta potentials of nanoparticles were observed, which demonstrated structural integrity and stability of the nanoformulation. Physicochemical characteristics of MDNPs were evaluated by FTIR and TGA and demonstrated the presence of mannose units on surface nanoparticles. A mannose-dependent cellular targeting and uptake of MDNPs was found in U937 macrophages. The uptake process was found to vary directly with time and volume of MDNPs nanoparticles. The uptake pattern is higher in M2 than M1. This behavior was also evident from the instantaneous and superior binding profile of M2 macrophage lysate protein with MDNPs over that of M1 macrophage lysate protein. These results demonstrated that an appropriate mannose ligand density was confirmed, suggesting efficient targeting of M2. Altogether, these data support that the MDNPs formulation could serve as a targeted therapeutic guide in the generation of nanomedicine to treat various conditions as an anti-inflammation therapy.  相似文献

We report the development of an integrated multifunctional imaging system capable of providing anatomical (optical coherence tomography, OCT), functional (OCT angiography, OCTA) and molecular imaging (light‐induced autofluorescence, LIAF) for in vivo dental applications. Blue excitation light (405 nm) was used for LIAF imaging, while the OCT was powered by a 1310 nm swept laser source. A red‐green‐blue digital camera, with a 450 nm cut‐on broadband optical filter, was used for LIAF detection. The exciting light source and camera were integrated directly with the OCT scanning probe. The integrated system used two noninvasive imaging modalities to improve the speed of in vivo OCT data collection and to better target the regions of interest. The newly designed system maintained the ability to detect differences between healthy and hypomineralized teeth, identify dental biofilm and visualize the microvasculature of gingival tissue. The development of the integrated OCT‐LIAF system provides an opportunity to conduct clinical studies more efficiently, examining changes in oral conditions over time.  相似文献

Zoonoses disproportionately affect tropical communities and are associated with human modification and use of ecosystems. Effective management is hampered by poor ecological understanding of disease transmission and often focuses on human vaccination or treatment. Better ecological understanding of multi-vector and multi-host transmission, social and environmental factors altering human exposure, might enable a broader suite of management options. Options may include “ecological interventions” that target vectors or hosts and require good knowledge of underlying transmission processes, which may be more effective, economical, and long lasting than conventional approaches. New frameworks identify the hierarchical series of barriers that a pathogen needs to overcome before human spillover occurs and demonstrate how ecological interventions may strengthen these barriers and complement human-focused disease control. We extend these frameworks for vector-borne zoonoses, focusing on Kyasanur Forest Disease Virus (KFDV), a tick-borne, neglected zoonosis affecting poor forest communities in India, involving complex communities of tick and host species. We identify the hierarchical barriers to pathogen transmission targeted by existing management. We show that existing interventions mainly focus on human barriers (via personal protection and vaccination) or at barriers relating to Kyasanur Forest Disease (KFD) vectors (tick control on cattle and at the sites of host (monkey) deaths). We review the validity of existing management guidance for KFD through literature review and interviews with disease managers. Efficacy of interventions was difficult to quantify due to poor empirical understanding of KFDV–vector–host ecology, particularly the role of cattle and monkeys in the disease transmission cycle. Cattle are hypothesised to amplify tick populations. Monkeys may act as sentinels of human infection or are hypothesised to act as amplifying hosts for KFDV, but the spatial scale of risk arising from ticks infected via monkeys versus small mammal reservoirs is unclear. We identified 19 urgent research priorities for refinement of current management strategies or development of ecological interventions targeting vectors and host barriers to prevent disease spillover in the future.  相似文献
