We examined the effect of the wind vector analyzed into its three components (direction, speed and persistence), on the circulation of pollen from different plant taxa prominent in the Thessaloniki area for a 4-year period (1996–1999). These plant taxa were Ambrosia spp., Artemisia spp., Chenopodiaceae, Corylus spp., Cupressaceae, Olea europaea, Pinaceae, Platanus spp., Poaceae, Populus spp., Quercus spp., and Urticaceae. Airborne pollen of Cupressaceae, Urticaceae, Quercus spp. and O. europaea make up approximately 70% of the total average annual pollen counts. The set of data that we worked with represented days without precipitation and time intervals during which winds blew from the same direction for at least 4 consecutive hours. We did this in order to study the effect of the different wind components independently of precipitation, and to avoid secondary effects produced by pollen resuspension phenomena. Factorial regression analysis among the summed bi-hourly pollen counts for each taxon and the values of wind speed and persistence per wind direction gave significant results in 22 cases (combinations of plant taxa and wind directions). The pollen concentrations of all taxa correlated significantly with at least one of the three wind components. In seven out of the 22 taxon-wind direction combinations, the pollen counts correlated positively with wind persistence, whereas this was the case for only two of the taxon-wind speed combinations. In seven cases, pollen counts correlated with the interaction effect of wind speed and persistence. This shows the importance of wind persistence in pollen transport, particularly when weak winds prevail for a considerable part of the year, as is the case for Thessaloniki. Medium/long-distance pollen transport was evidenced for Olea (NW, SW directions), Corylus (NW, SW), Poaceae (SW) and Populus (NW).  相似文献

A recycle ultrafiltration membrane reactor was used to develop a continuous synthesis process for the production of isomaltooligosaccharides (IMO) from sucrose, using the enzymes dextransucrase and dextranase. A variety of membranes were tested and the parameters affecting reactor stability, productivity, and product molecular weight distribution were investigated. Enzyme inactivation in the reactor was reduced with the use of a non-ionic surfactant but its use had severe adverse effects on the membrane pore size and porosity. During continuous isomaltooligosaccharide synthesis, dextransucrase inactivation was shown to occur as a result of the dextranase activity and it was dependent mainly on the substrate availability in the reactor and the hydrolytic activity of dextranase. Substrate and dextranase concentrations (50-200 mg/mL(-1) and 10-30 U/mL(-1), respectively) affected permeate fluxes, reactor productivity, and product average molecular weight. The oligodextrans and isomaltooligosaccharides formed had molecular weights lower than in batch synthesis reactions but they largely consisted of oligosaccharides with a degree of polymerization (DP) greater than 5, depending on the synthesis conditions. No significant rejection of the sugars formed was shown by the membranes and permeate flux was dependent on tangential flow velocity.  相似文献

We tested for host-associated genetic differentiation in 22 populations of Thrips tabaci collected from tobacco and leek, respectively. Clustering analyses and haplotype networks based on sequence variation at a fragment of the mitochondrial cytochrome oxidase I gene yielded three major evolutionary lineages; two were clearly associated with leek and the third with tobacco. These genetic findings corroborated recent experimental observations on the heterogeneity of T. tabaci populations with regard to host-plant preference and their capacity to be vectors for tomato spotted wilt virus. Estimated divergence times suggested an ancient divergence of these lineages dating back to the Miocene 28-21 million years ago. F(ST) values between these lineages ranged between 0.824 and 0.954 (P<0.001 for all comparisons), and sequence divergences ranged between 4 and 11%. Given these findings and by the standards of genetic and ecological differentiation in other published species groups, T. tabaci must be considered a complex of cryptic (sub)species.  相似文献

RXPA380 (Cbz-PhePsi[PO(2)CH]Pro-Trp-OH) was reported recently as the first highly selective inhibitor of the C-domain of somatic angiotensin-converting enzyme (ACE), able to differentiate the two active sites of somatic ACE by a selectivity factor of more than 3 orders of magnitude. The contribution of each RXPA380 residue toward this remarkable selectivity was evaluated by studying several analogues of RXPA380. This analysis revealed that both pseudo-proline and tryptophan residues in the P(1)' and P(2)' positions of RXPA380 play a critical role in the selectivity of this inhibitor for the C-domain. This selectivity is not due to a preference of the C-domain for inhibitors bearing pseudo-proline and tryptophan residues, but rather reflects the poor accommodation of these inhibitor residues by the N-domain. A model of RXPA380 in complex with the ACE C-domain, based on the crystal structure of germinal ACE, highlights residues that may contribute to RXPA380 selectivity. From this model, striking differences between the N- and C-domains of ACE are observed for residues defining the S(2)' pocket. Of the twelve residues that surround the tryptophan side chain of RXPA380 in the C-domain, five are different in the N-domain. These differences in the S(2)' composition between the N- and C-domains are suggested to contribute to RXPA380 selectivity. The structural insights provided by this study should enhance understanding of the factors controlling the selectivity of the two domains of somatic ACE and allow the design of new selective ACE inhibitors.  相似文献

Global sensitivity analysis (GSA) can be used to quantify the importance of model parameters and their interactions with respect to model output. In this study, the Sobol' method for GSA is applied to a dynamic model of monoclonal antibody-producing mammalian cell cultures in order to identify the parameters that need to be accurately determined experimentally. Our results show that most parameters have low sensitivity indices and exhibit strong interactions with one another. These parameters can be set at their nominal values and unnecessary experimentation can therefore be avoided. In contrast, certain parameters are identified as sensitive, necessitating their estimation given sufficiently rich experimental data. Moreover, parameter sensitivity varies during culture time in a biologically meaningful manner. In conclusion, GSA can serve as an excellent precursor to optimal experiment design.  相似文献

Cuttings of Arbutus unedo (strawberry tree) and Taxus baccata (yew) were treated with 8.0 and 10.0 g l–1, respectively, of KIBA, IBA, IAA, NAA and Paclobutrazol. No rooting occurred without growth regulator treatment. The effect of growth regulators on percentage of rooting followed the order KIBA > IBA > IAA = NAA = Paclobutrazol = 0% (for A. unedo) and KIBA > IBA > IAA > NAA > Paclobutrazol = 0% (for T. baccata). Genotypes of the above plant species had significant effects on the number and length of roots, percentage of rooting and peroxidase specific activity (PA) on KIBA-treated cuttings. High PA seems to be related with low percentage of rooting in the case of A. unedo cuttings while no similar results were noticed in the case of T. baccata. Electrophoretic analysis revealed the appearance of two to three anionic and one cationic peroxidase isoforms in A. unedo cuttings, while six to nine anionic and no cationic peroxidases isoforms appeared in the case of T. baccata genotypes. During adventitious rooting, the PA showed the three interdependent phases (induction, initiation, expression) in both K-IBA treated cuttings of A. unedo and T. baccata, but in a different time course.  相似文献

We investigated the pattern of glutathione S-transferase (GST) activity in the course of the development of Apis mellifera macedonica. GST activity is present in all developmental stages of A. mellifera macedonica. The highest activity towards the substrate 1-chloro-2,4-dinitrobenzene (CDNB) is found in the adult stage and the lowest in the egg. The kinetic characteristics of the whole enzyme change as the insect develops. Significant changes are observed in substrate specificity, inhibitor sensitivity and V(max). The number of isoenzymes and their rate of expression vary as the insect develops. However, two main isoenzymes are present in all developmental stages, one in the alkaline area and the other in the acidic. While in the larval stage the acidic isoenzyme is expressed at a slightly higher rate (52.2% over 47.8% for the alkaline isoenzyme), in the adult stage, the rate is reversed dramatically (13.24% and 84.2%, respectively).  相似文献

Small-scale GC headspace analyses combined with SPME sampling of individual oil glands have been used to measure the variation in volatile content and composition in and within different oregano plants as affected by age, season and developmental state. The main monoterpenes found were p-cymene, carvacrol and their precursor gamma-terpinene. The early season preponderance of p-cymene over carvacrol was reversed as the season progressed and this pattern could also be seen at any time within the plant, from the latest leaves to be produced (low in cymene) to the earliest (high in cymene). Seedlings from the same mother plant developed this pattern at different rates. Within individual leaves the pattern was not observed, even within the youngest developing leaves. However it was found that the oil composition of individual glands within a single leaf varied considerably, most notably in respect of the production of carvacrol and its isomer thymol.  相似文献

An investigation of selected Aleppo pines in the forests of Mt Hymettus and Mt Parnis near Athens (Greece) was undertaken at three different sites in the period 1999–2003, because a considerable proportion of pine trees showed visible signs of chlorotic mottle. This condition is characteristic of high and prolonged levels of ozone exposure. Needles from Aleppo pine trees (Pinus halepensis Mill.) were analyzed for their manganese content in combination with Electron Paramagnetic Resonance (EPR) spectra of Mn2+, involved in photosystem II. Manganese is considered as an important bioindicator for the vitality of trees. Also, we investigated the EPR spectrum of the needles in the region of g=2.0045 for healthy and diseased trees. The antioxidant capacity of the needles extract was measured from trees by the DPPH method. Finally, seasonal changes in chlorophyll concentration in the needles were measured to evaluate the effects of ozone. Measurements of ozone concentrations at the three sites showed that there were elevated levels during the summer months. Our experimental results suggest that the concentration of manganese in the needles was lower in the area with higher ozone concentrations, supported by EPR measurements. Higher ozone concentrations also affected the antioxidant potential of the needles and their chlorophyll content during summer months. Our findings also confirmed the resilience of Aleppo pines under stressful conditions and recovery in winter months. Despite the experimental problems, EPR spectra of Mn2+ in combination with other methods can be used as a sensitive bioindicator for ozone pollution, and is the result of oxidative stress affecting the growth cycle of the pine trees and their photosynthetic mechanisms.  相似文献
