Transgenics from several forest tree species, carrying a number of commercially important recombinant genes, have been produced,
and are undergoing confined field trials in a number of countries. However, there are questions and issues regarding stability
of transgene expression and transgene dispersal that need to be addressed in long-lived forest trees. Variation in transgene
expression is not uncommon in the primary transformants in plants, and is undesirable as it requires screening a large number
of transformants in order to select transgenic lines with acceptable levels of transgene expression. Therefore, the current
focus of plant transformation is toward fine tuning of transgene expression and stability in the transgenic forest trees.
Although a number of studies have reported a relatively stable transgene expression for several target traits, including herbicide
resistance, insect resistance, and lignin modification, there was also some unintended transgene instability in the genetically
modified (GM) forest trees. Transgene dispersal from GM trees to feral forest populations and their containment remain important
biological and regulatory issues facing commercial release of GM trees. Containment of transgenes must be in place to effectively
prevent escape of transgenic pollen, seed, and vegetative propagules in economically important GM forest trees before their
commercialization. Therefore, it is important to devise innovative technologies in genetic engineering that lead to genetically
stable transgenic trees not only for qualitative traits (herbicide resistance, insect resistance), but also for quantitative
traits (accelerated growth, increased height, increased wood density), and also prevent escape of transgenes in the forest

The heterogeneous Indian tea germplasm includes ‘China’, ‘Assam’, ‘Cambod’, and their hybrids which were evaluated using biochemical markers viz., total catechin and their fractions, for varietal identification and characterization. Principal component analysis (PCA) of biochemical characters showed that the total catechin and trihydroxylated catechin has higher eigenvalues. The first two principal components (PCs) could differentiate more than 90% of the clones studied. This grouping based on first two principal component matrices differentiated ‘China’, and their hybrids with ‘Assam’ and ‘Cambod’ variety. Morphologically indistinct large‐leaved ‘Cambod’ variety and ‘Assam’ varieties could not be differentiated using biochemical markers, since both varietal types taxonomically belong to a single species. Clones of ‘China’ type showed low total catechin content and catechin ratio which are distinctly grouped. The ‘China–Assam’ and ‘China–Cambod’ hybrids formed intermediate groups between ‘China’ PC group and ‘Cambod’/‘Assam’ PC groups, providing evidence for genetic control of catechin ratio variation. Tea clones which are differentially positioned in the PC group could be explained based on the genetic contribution by other varietal type as parents. Rheum grows luxuriantly in a niche of low temperature (LT) at high altitudes in Himalayan belt. The plant is expected to harbor
novel genes particularly for tolerance to LT. Using differential display, two cDNAs RaMPK1 and RaMPK2, showing homology to mitogen-activated protein kinases (MAPKs) were isolated. As compared to RaMPK1, RaMPK2 exhibited strong up-regulation in response to LT. RaMPK1 was novel in terms of possessing a small glutamine and proline rich
region at the N-terminal end. Secondly, though RaMPK1 showed homology with salicylic acid (SA) responsive MAPKs, the gene was down-regulated by SA but activated by jasmonate (JA). Abscisic acid (ABA) and polyethylene glycol (PEG) also
down-regulated RaMPK1. RaMPK2 showed down-regulation within 5 min of exposure to JA and SA treatments, followed by gradual increase in expression. Expression

Anthrax lethal toxin (LT) comprises two proteins: the protective antigen (PA) and the lethal factor (LF). The LT is cytotoxic to macrophage-like cell line J774A.1. Pre-treatment of these cells with neomycin, a phospholipase C inhibitor, protected them against anthrax LT cytotoxicity. Protection obtained with neomycin indicated that LT stimulates phospholipase C in these cells. It was found that levels of inositol 1,4,5-triphosphate (IP3) dramatically increased in toxin-treated cells. The rise in IP3 levels was proportional to the dose of LF that was allowed to bind to receptor-bound PA. By using protein kinase C (PKC) inhibitors, we found that the activation of PKC is required for mediating anthrax LT cytotoxicity. Activation of phospholipase C or PKC is not required for the binding of PA to the cell surface receptors or for the uptake or internalisation of the toxin. In this study, we demonstrate that the IP3 signalling cascade is initiated by anthrax lethal toxin in J774A.1 cells. The second messengers generated during the cascade aid LF in mediating lethality only after its translocation into the cytosol. 相似文献

Triplet repeat expansion in 3 untranslated region of myotonic dystrophy protein kinase (DMPK) gene has been implicated as causative in myotonic dystrophy (DM). In cases of DM, high levels of somatic instability have been reported, in which inter-tissue repeat length differences as large as 3000 repeats have been observed. This study highlights the inter-tissue (CTG)n expansion variability at the DMPK locus. Molecular analysis of DMPK gene, encompassing the triplet repeat expansion, was carried out in 31 individuals (11 clinically identified DM patients, 20 controls). All controls showed a 2.1kb band (upto 35 CTG repeats), while four cases exhibited an expansion (>50 repeats). A novel observation was made in one case, wherein the DNA from lymphocytes showed a normal 2.1kb band while the muscle tissue DNA from the same patient was heterozygous for normal and 4.3 kb band (>700 repeats). Our results suggested that because inter-tissue variability existed in the (CTG)n repeat number at DMPK locus, an attempt should be made to evaluate affected tissue along with blood wherever possible prior to making a final diagnosis. Aquifex aeolicus, an organism that flourishes at 95 degrees C, is one of the most thermophilic eubacteria thus far described. The A. aeolicus pyrB gene encoding aspartate transcarbamoylase (ATCase) was cloned, overexpressed in Escherichia coli, and purified by affinity chromatography to a homogeneous form that could be crystallized. Chemical cross-linking and size exclusion chromatography showed that the protein was a homotrimer of 34-kDa catalytic chains. The activity of A. aeolicus ATCase increased dramatically with increasing temperature due to an increase in kcat with little change in the Km for the substrates, carbamoyl phosphate and aspartate. The Km for both substrates was 30-40-fold lower than the corresponding values for the homologous E. coli ATCase catalytic subunit. Although rapidly degraded at high temperature, the carbamoyl phosphate generated in situ by A. aeolicus carbamoyl phosphate synthetase (CPSase) was channeled to ATCase. The transient time for carbamoyl aspartate formation was 26 s, compared with the much longer transient times observed when A. aeolicus CPSase was coupled to E. coli ATCase. Several other approaches provided strong evidence for channeling and transient complex formation between A. aeolicus ATCase and CPSase. The high affinity for substrates combined with channeling ensures the efficient transfer of carbamoyl phosphate from the active site of CPSase to that of ATCase, thus preserving it from degradation and preventing the formation of toxic cyanate. 相似文献

Seedling raised elites of Dendrocalamus hamiltoniiNees et Arn. Ex Munro were chosen as the source of nodal explants from precocious branches. While axillary bud break was accomplished in hormone free 1/2MS medium containing sucrose (3%, w/v), BA supplementation was required for shoot proliferation. A variety of hormonal combinations induced rooting in clumps of shoots. Somatic embryogenesis was also obtained in callus cultures raised in 2,4-D supplemented MS medium and plantlets derived from somatic embryos were hardened for field transfer. Comparative growth performances of plants raised from nodal cuttings of field-grown plants, those from single node cuttings of precocious branches and from somatic embryos indicated that growth performance of the tissue culture raised plants was relatively better than those from nodal cuttings. Improved protocols for efficient micropropagation are visualized to provide an impetus to raising of bamboo nurseries of elite genotypes in bamboo growing areas of western Himalayas. 相似文献

A few constituents of the brain of rats subjected to long-term shock avoidance training in the jump-box were investigated. The effects of training and that of the concomitant stress on these constituents were differentially examined. Brain DNA increased in both the trained rats and those subjected to stress. The total RNA showed a specific decrease in the trained rats. The total protein content remained unaffected, while the content of soluble protein showed a greater increase in the trained rats compared to that in animals subjected to stress. The Na+, K+-ATPase activity in the brain decreased in the trained rats, whereas the glial carbonic anhydrase activity increased in the stressed as well as the trained rats. No change was found in the activities of Mg++-ATPase and pseudocholinesterase in the experimental groups. 相似文献

62 patients suffering from rheumatic heart disease were taken from Institute of Child Health, Niloufer Hospital, Hyderabad, for the present dermatoglyphic investigation. The finger, palm and toe prints were analysed to see if there was an association between rheumatic heart disease and any of the dermatoglyphic traits. The parameters which were significantly different from the controls are: (1) reduced frequency of arches on the finger tips in males and increased frequency of whorls in females; (2) increased frequency of patterns in the III interdigital area in males; (3) decreased d-t ridge count in females, and (4) higher incidence of multiple axial triradii in females. 相似文献
