Background

Glucose effects on beta cell survival and DNA-synthesis suggest a role as regulator of beta cell mass but data on beta cell numbers are lacking. We examined outcome of these influences on the number of beta cells isolated at different growth stages in their population.

Methods

Beta cells from neonatal, young-adult and old rats were cultured serum-free for 15 days. Their number was counted by automated whole-well imaging distinguishing influences on cell survival and on proliferative activity.

Results

Elevated glucose (10–20 versus 5 mmol/l) increased the number of living beta cells from 8-week rats to 30%, following a time- and concentration-dependent recruitment of quiescent cells into DNA-synthesis; a glucokinase-activator lowered the threshold but did not raise total numbers of glucose-recruitable cells. No glucose-induced increase occurred in beta cells from 40-week rats. Neonatal beta cells doubled in number at 5 mmol/l involving a larger activated fraction that did not increase at higher concentrations; however, their higher susceptibility to glucose toxicity at 20 mmol/l resulted in 20% lower living cell numbers than at start. None of the age groups exhibited a repetitively proliferating subpopulation.

Conclusions

Chronically elevated glucose levels increased the number of beta cells from young-adult but not from old rats; they interfered with expansion of neonatal beta cells and reduced their number. These effects are attributed to age-dependent differences in basal and glucose-induced proliferative activity and in cellular susceptibility to glucose toxicity. They also reflect age-dependent variations in the functional heterogeneity of the rat beta cell population.  相似文献

Yeasts are important microorganisms used for ethanol production; however, they are not equally efficient in the amount of ethanol production under different environmental conditions. It is, therefore, necessary to screen for elite strains to utilize them for commercial production of these commodities. In this study, yeasts were isolated from different Ethiopian traditional fermented alcoholic beverages (teji, tella, shamiata and areqe tinisis), milk and ergo, teff and maize dough, soil and compost, flowers, and fruits to evaluate their potential use for ethanol fermentation process. Isolates were screened for efficient ethanol production and the selected ones were identified using phenotypic and genetic characters using D1/D2 region of LSU rDNA sequence analysis. The yeast isolates were evaluated based on their growth and fermentation of different carbon sources. Response surface methodology (RSM) was applied to optimize temperature, pH and incubation time using central composite design (CCD) in Design-Expert 7.0.0. A total of 211 yeasts colonies were isolated of which 60% were ethanologenic yeasts (ethanol producers) and 40% were non-ethanol producers. The yeast population detected from various sources was in the range of 105 CFU from traditional foods and beverages to that of 103 CFU from fruits and soil samples. The data also showed that the number of colony types (diversity) did not correlate with population density. The highly fermentative isolates were taxonomically characterized into four genera, of which 65% of the isolates (ETP37, ETP50; ETP53, ETP89, ETP94) were categorized under Saccharomyces cerevisiae, and the remaining were Pichia fermentans ETP22, Kluyveromyces marxianus ETP87, and Candida humilis ETP122. The S. cerevisiae isolates produced ethanol (7.6-9.0 g/L) similar with K. marxianus ETP87 producing 7.97 g/L; comparable to the ethanol produced from commercial baker's yeast (8.43 g/L) from 20 g/L dextrose; whereas C. humilis ETP122 and P. fermentans ETP22 produced 5.37 g/L and 6.43 g/L ethanol, respectively. S. cerevisiae ETP53, K. marxianus ETP87, P. fermentans ETP22 and C. humilis ETP122 tolerated 10% extraneous ethanol but the percentage of ethanol tolerance considerably decreased upon 15%. S. cerevisiae ETP53 produced ethanol optimally at pH 5.0, 60 h, and 34oC. pH 4.8, temperature 36oC, and 65 h of time were optimal growth conditions of ethanol fermentation by K. marxianus ETP87. The ethanol fermentation conditions of P. fermentans ETP22 was similar to S. cerevisiae ETP53 though the ethanol titer of S. cerevisiae ETP53 was higher than P. fermentans ETP22. Therefore, S. cerevisiae ETP53, K. marxianus and P. fermentans ETP22 are good candidates for ethanol production.  相似文献

Main Objective

Few studies have examined the long-term, impact of large-scale interventions to strengthen primary care services for women and children in rural, low-income settings. We evaluated the impact of the Ethiopian Millennium Rural Initiative (EMRI), an 18-month systems-based intervention to improve the performance of 30 primary health care units in rural areas of Ethiopia.

Methods

We assessed the impact of EMRI on maternal and child survival using The Lives Saved Tool (LiST), Demography (DemProj) and AIDS Impact Model (AIM) tools in Spectrum software, inputting monthly data on 6 indicators 1) antenatal coverage (ANC), 2) skilled birth attendance coverage (SBA), 3) post-natal coverage (PNC), 4) HIV testing during ANC, 5) measles vaccination coverage, and 6) pentavalent 3 vaccination coverages. We calculated a cost-benefit ratio of the EMRI program including lives saved during implementation and lives saved during implementation and 5 year follow-up.

Results

A total of 134 lives (all children) were estimated to have been saved due to the EMRI interventions during the 18-month intervention in 30 health centers and their catchment areas, with an estimated additional 852 lives (820 children and 2 adults) saved during the 5-year post-EMRI period. For the 18-month intervention period, EMRI cost $37,313 per life saved ($42,366 per life if evaluation costs are included). Calculated over the 18-month intervention plus 5 years post-intervention, EMRI cost $5,875 per life saved ($6,671 per life if evaluation costs are included). The cost effectiveness of EMRI improves substantially if the performance achieved during the 18 months of the EMRI intervention is sustained for 5 years. Scaling up EMRI to operate for 5 years across the 4 major regions of Ethiopia could save as many as 34,908 lives.

Significance

A systems-based approach to improving primary care in low-income settings can have transformational impact on lives saved and be cost-effective.  相似文献

Ecological survey was executed to assess woody species encroachment into the grassland plain of Nechisar National Park (NNP). Forty‐one woody species were recorded. Dichrostachys cinerea Wight & Arn., Acacia mellifera (Vahl) Benth., Acacia nilotica (L) Willd., Acacia senegal (L.) Willd., Acacia seyal Del. and Acacia tortilis (Forssk.) Hayne were among the major encroaching woody species. The majority of the woody species were found to be highly aggregated in their pattern of distribution, while only few species showed some degree of randomness. The mean woody species density was ca. 1995 woody plants ha?1. Mean cover of woody, grass, unpalatable forbs and total herbaceous species were 31%, 58%, 68% and 121%, respectively. The woody species density and cover, unpalatable forbs and bare land cover were significantly higher in the highly grazed and fire‐suppressed part of the grassland plain. Pearson correlation coefficient matrix indicated that woody species cover and density were negatively correlated with total herbaceous and grass cover. The high woody, unpalatable forbs and bare land cover indicated the progressively increasing perennial grass species diversity deterioration in the grass plain of the Park. Decline in the grassland condition, unless reversed, will jeopardize the biological diversity as well as the aesthetic value of the NNP.  相似文献

The guild of stem-infesting insect pests of cultivated sunflower, Helianthus annuus L., within the central Plains is a concern to producers, chiefly due to losses caused by plant lodging from the sunflower stem weevil, Cylindrocopturus adspersus (LeConte) (Coleoptera: Curculionidae), and Dectes texanus texanus LeConte (Coleoptera: Cerambycidae). The incidence of a root boring moth, Pelochrista womonana (Kearfott) (Lepidoptera: Tortricidae), also has increased. Experiments were conducted in Kansas during 2000-2001 to investigate the effect of irrigation timing and intensity on densities of C. adspersus, D. texanus, and P. womonana larvae within cultivated sunflower stalks. Supplemental soil moisture provided by irrigation during the growing season increased both seed yield and oil content, and it reduced insect densities of the sunflower stem weevil and P. womonana in the sunflower stalk. Results showed that ensuring adequate moisture during the growing season can assist in reducing stem-infesting insect densities, revealing an additional advantage of crop irrigation beyond improved sunflower productivity.  相似文献

Background

Sheep pox and goat pox are contagious viral diseases of sheep and goats, respectively. The diseases result in substantial economic losses due to decreased milk and meat production, damage to hides and wool, and possible trade restriction. A study was undertaken in Amhara region of Ethiopia. A cross-sectional study design was used to estimate the sero-prevalence and identify associated risk factors, while retrospective study design was used to assess the temporal and spatial distribution of the disease. A total of 672 serum samples were collected from 30 Kebeles and tested using virus neutralization test.

Results

From a total of 672 sera tested, 104 (15.5%) were positive for sheep and goat pox virus antibody; from which 56 (17%) were sheep and 48 (14%) were goats. The diseases were prevalent in all study zones, the highest sero-prevalence was observed in South Gondar (20.9%) and the lowest in North Gondar and West Gojjam zones (11.9% each). From the potential risk factors considered (species, sex, age, agro-ecology and location); only sex and age were significantly associated (p?<?0.05) with the diseases in multivariable logistic regression. Female and young animals were at higher risk than their counterparts. From January 2010 to December 2014, a total of 366 outbreaks, 12,822 cases and 1480 deaths due to SP and 182 outbreaks, 10,066 cases and 997 deaths due to GP were recorded in Amhara National Regional State.

Conclusion

Both the serological and the outbreak data revealed that sheep and goat pox is one of the most prevalent and widespread diseases of sheep and goats in the study area. Hence, annual mass vaccination program must be implemented for economic and viable control of sheep and goat pox diseases in the Amhara region in particular and at a national level in general.

Purpose

The aim of this study is to develop and test the applicability of a new subcategory assessment method (SAM) for social life cycle assessment using a case study on high-density polyethylene (HDPE) production by Dow Chemical Canada facility in Alberta, Canada.

Methods

The methodology is characterized by six steps: (1) definition of the goal and scope of the assessment; (2) life cycle inventory data collection including context data at country level and company-specific data for foreground processes; (3) impact assessment where the subcategories’ results for foreground processes at company level are evaluated using a new SAM developed and the country social performance in the different subcategories is evaluated using some assessment intervals; (4) comparing the social performance of foreground processes to the social background context in sector or country; (5) evaluating the social performance of background processes using sector performance evaluation from Product Social Impact Life Cycle Assessment (PSILCA) database or country performance evaluation conducted in the study; (6) and discussion and conclusion.

Results and discussion

The method developed was able to identify the subcategories that need high level or some level of improvements along the cradle-to-gate life cycle of HDPE. In addition, the stakeholders with high negative effect were highlighted in every life cycle stage. Through this approach, Dow Chemical Canada is able to prioritize its actions and focus on the areas where its performance is still low compared to its peers in the sector or compared to the situation in the country. Moreover, through using PSILCA database or country performance evaluation to investigate the social performance of background processes, Dow Chemical Canada can determine the social hotspots areas that need more focus from its suppliers.

Conclusions

Applying the new subcategory assessment approach proposed in this study provides an objective way to assess the subcategories while differentiating between two levels of assessment: (1) the commitment to the social subcategory in the company’s policy (defined as cutoff requirement) and (2) the evidence of good/bad practices of the subcategory. In addition, the comparison of performance of the companies in foreground processes to the social background in sector or country has improved the objectivity further.

The first part of this study evaluated contents of hydrolysable tannins (HT) and condensed tannins (CT) of 65 accessions of tagasaste. In the second part, the effects of harvesting stage and growing season on the contents of HT, CT and alkaloids in different plant parts of the widely grown tagasaste variety ‘MOA’ was studied in a randomized complete block design. Regrowths of tagasaste was harvested in one experiment at 4, 6, 8 and 10 months, separated into growing buds, leaves, barks, branches and stems and in another trial at 6 months regrowth during the main rainy, dry and short rainy seasons. The leaves of different tagasaste accessions contained on average 115 g/kg dry matter (DM) HT in a range of 16–197 and 12.5 abs/g NDF CT in a range of 6.9–35.0 abs/g NDF. The edible fractions (growing buds and leaves) were higher in HT and CT contents compared to the structural parts (stems and barks). The leaf fraction contained higher HT and CT (177.2 g/kg DM and 20.1 abs/g NDF) and the HT tended to decline with progressing harvesting stages. High concentration of alkaloids was found in the non-edible fractions, with the highest content in the bark (235 mg/kg) and the lowest in the leaf (40 mg/kg DM) fractions. These levels increased in the fractions from the 4th to the 8th month of harvesting stage and then declined at the 10th month. About 90% of the alkaloids found in tagasaste were sparteine. Higher concentration of HT was found in tagasaste leaves, harvested during the rainy season compared to the dry season. The concentration of CT was low during the main rainy season, while the highest was during the short rainy and dry seasons. Contents of HT and CT were positively correlated (r = 0.25, P<0.01). However, both the HT and CT contents had negative correlation coefficient of r = −0.43 (P<0.001) and r = −0.73 (P<0.001) with alkaloids, respectively. The results of this study showed that varietal selection, and harvesting management could be used as a means to modulate the level of tannins and alkaloids in tagasaste.  相似文献

Shrubland vegetation and environmental data in western Shewa, Ethiopia have been analysed. Vegetation data include cover-abundance values of vascular plant species; environmental data comprise physical and chemical properties of the soil, altitude, slope, grazing and browsing pressure. The vegetation data were subj ected to hierarchical and non-hierarchical classification and ordination with correspondence analysis. The classification resulted in seven different vegetation types, ranging from grassland with scattered shrubs to degenerated forest. Ordination of the data and biplot analysis showed that the vegetation is influenced by anthropogenic factors and altitudinal variation. Sand content is related to a low level of anthropogenic influence whereas silt content is related to a high level. This is explained by historical events rather than by the present situation. Total nitrogen, organic carbon, altitude and slope are positively correlated and these variables are negatively related to anthropogenic influences. The shrubland vegetation may have expanded from lower altitudes and drier sites as forests gradually disappeared. The recovery of an economically more rewarding vegetation type may be achieved through pro viding alternative sources of fuel and construction and through prohibiting cultivation and grazing in the shrublands on the hillsides. Regeneration can be accelerated by actively introducing seedlings of tree species that do not need a heavy canopy cover for establishment and growth.  相似文献
