The sulfated polysaccharides (SPs) from Chlamydomonas reinhardtii (Cr) were isolated by hot water method using 80% alcohol and semi purified by anion-exchange column chromatography. The chemical analysis of the extract showed 78% carbohydrates, 18% reducing sugars, 60% non-reducing sugars, 2% protein, 33% sulfate, 39% uronic acid, and 4% ash. The elemental analysis of this C. reinhardtii sulfated polysaccharide-enriched extract (Cr-SPs) showed 53% carbon, 8% hydrogen, and 6% nitrogen. FTIR analysis of Cr-SPs showed characteristic bands of sulfated polysaccharides. Further, the Cr-SPs showed significant hydroxyl radical scavenging activity of 22.29–80.9% at 0.01–1 mg mL?1, 38–77% of DPPH radical scavenging activity at 0.01–1 mg mL?1, 9.8–81% ABTS radical scavenging activity, 34.5–67.6% of ferrous chelating ability, and 11.62–75% of total antioxidant capacity. Cr-SPs also showed efficient in vitro anticancer activity. Specifically, they inhibited triple-negative breast cancer cell (MDA-MB-231) proliferation with an IC50 of 172 μg mL?1. Concentration-dependent reduction in the number of colonies formed by MDA-MB-231 cells suggested their potential to inhibit the clonal expansion of the cancer cells. Higher concentrations of crude extract were found to disrupt the microtubule networks in these cells. The cells treated with Cr-SPs eventually underwent apoptosis as evidenced by the formation of characteristic DNA ladder. These results indicate that Cr-SPs find promising opportunities for cancer treatment.  相似文献

To explore bacterial diversity for elucidating genetic variability in acylhomoserine lactone (AHL) lactonase structure, we screened 800 bacterial strains. It revealed the presence of a quorum quenching (QQ) AHL-lactonase gene (aiiA) in 42 strains. These 42 strains were identified using rrs (16S rDNA) sequencing as Bacillus strains, predominantly B. cereus. An in silico restriction endonuclease (RE) digestion of 22 AHL lactonase gene (aiiA) sequences (from NCBI database) belonging to 9 different genera, along with 42 aiiA gene sequences from different Bacillus spp. (isolated here) with 14 type II REs, revealed distinct patterns of fragments (nucleotide length and order) with four REs; AluI, DpnII, RsaI, and Tru9I. Our study reflects on the biodiversity of aiiA among Bacillus species. Bacillus sp. strain MBG11 with polymorphism (115Alanine > Valine) may confer increased stability to AHL lactonase, and can be a potential candidate for heterologous expression and mass production. Microbes with ability to produce AHL-lactonases degrade quorum sensing signals such as AHL by opening of the lactone ring. The naturally occurring diversity of QQ molecules provides opportunities to use them for preventing bacterial infections, spoilage of food, and bioremediation.  相似文献

ProjectSelenium deficiency has been associated with enhanced propensity of seizures in man and laboratory animals. Therefore, the present study has been designed to investigate the anti-convulsant effect of sodium selenite and seleno-dl-methionine on pentylenetetrazole induced seizures in mice and the role of prostaglandin receptor activation in the proposed anticonvulsant effect of sodium selenite.ProcedureSodium selenite (1, 3 and 10 mg kg?1, i.p.) and seleno-dl-methionine (0.3, 1 and 3 mg kg?1, i.p.) was used to evaluate the potential effect on pentylenetetrazole induced seizures in mice. Pentylenetetrazole induced seizures were assessed in terms of onset time of straub's tail phenomenon, jerky movements of the whole body and convulsions. Additionally, an isobolographic study design was used to examine the interaction between sodium selenite and celecoxib (a cyclooxygenase-2 inhibitor). Sodium selenite and seleno-dl-methionine significantly attenuated pentylenetetrazole induced seizures in mice.ResultsPrior administration of misoprostol (a selective agonist of prostaglandin E1 receptors) markedly attenuated the anticonvulsant effect of sodium selenite as well as seleno-dl-methionine in mice. However, the administration of misoprostol per se did not produce any behavioral changes. Further, sodium selenite was observed to exert a synergistic interaction with celecoxib.ConclusionsSelenium induced reduction in seizure like behavior might be ascribed to the activation of a prostaglandin E1 receptor activation linked mechanism. It is further proposed that sodium selenite exerts a synergistic anti-convulsant effect with celecoxib indicating the therapeutic usefulness of combining the two agents to treat epilepsy.  相似文献

Variegin is a 32‐amino acid long thrombin inhibitory peptide isolated from the salivary gland extract of tropical bont tick Amblyomma variegatum. It was identified to be O‐glycosylated on its Thr‐14 side chain, and this glycosylated form was 14‐fold more potent than that of its non‐glycosylated form. However, as the identity of this glycosylation remained elusive, the mechanistic details underlying its functional impact are not yet known. In this report, we synthesized four different O‐glycosylated analogs of variegin bearing physiologically relevant sugars on its Thr‐14. Functional characterization of these analogs by enzyme inhibitory kinetics and surface plasmon resonance methods showed that all the synthesized glycopeptides are strong thrombin inhibitors. Structural studies by macromolecular docking identified that the sugar moiety of these peptides can potentially mediate favorable interactions with amino acids at the base of thrombin's autolysis loop. This report, for the first time, describes the impact of differential glycosylation on the function of a thrombin inhibitory peptide and tries to provide structural insights into the relevance of peptide glycosylation in thrombin inhibition. Four drill cores and a clay pit section have been examined in the southern part of the Körös plain to understand the history and controls on alluvial sedimentation for the last ~ 40 ka. Four facies groups were identified, such as channel, channel margin, floodplain and floodbasin with seven distinctive facies. Magnetic susceptibility and mineralogy have further characterized the sedimentary facies indicating shifts in humidity conditions, variations in sediment flux and pedogenesis. Detailed pollen analysis of a 7.5 m thick clayey succession indicated climatic variability within the MIS 3 period. The spatial distribution of the different facies allowed outlining alluvial architecture of the study area. Three depositional units composed of various facies were identified based on OSL and radiocarbon data. These packages correspond to three major phases of channel activity: (F-I) pre-LGM period (> 30 ka to 24 ka), (F-II) post-LGM interstadial (18–16 ka), and (F-III) Late Glacial < 15 ka to ~ 10 ka). The pre-LGM and post-LGM “interstadial” phases are characterized by meandering river patterns, while the Late Glacial fluvial activity is characterized by a braided system in the area. Higher sediment supply feeding this braided river was probably caused by neotectonic uplift of the southern margin of the basin, documented by a significant stratigraphic gap between 25 and 14 ka.  相似文献

Sodium dodecyl sulfate (SDS)-capped 1-pyrenecarboxaldehyde nanoparticles (PyalNPs) were prepared using a reprecipitation method in an aqueous medium and exhibited red-shifted aggregation-induced enhanced emission (AIEE). The dynamic light scattering (DLS) examination showed narrower particle size distribution with an average particle size of 41 nm, whereas −34.5 mV zeta potential value indicate the negative surface charge and good stability of nanoparticles (NPs) in an aqueous medium. The AIEE was seen at λmax = 473 nm in a fluorescence spectrum of a PyalNP suspension. In the presence of Cu2+ ions, the fluorescence of PyalNPs quenches very significantly, even in the presence of other metal ions like Ba2+, Ca2+, Cd2+, Co2+, Al3+, Fe2+, Hg2+, Ni2+ and Mg2+. The changes in the fluorescence lifetime of PyalNPs in the presence of Cu2+ ions suggested that the type of quenching was dynamic. The fluorescence quenching data for the NPs suspension fitted well into a typical Stern–Volmer relationship in the concentration range 1.0–25 μg/ml of Cu2+ ions. The estimated value of the correlation coefficient R2 = 0.9877 was close to 1 and showed the linear relationship between quenching data and Cu2+ ion concentration. The limit of detection (LOD) was found to be 0.94 ng/ml and is far below the tolerable intake limit value of 1.3 μg/ml accepted by the World Health Organization for Cu2+ ions in drinking water. The fluorescence quenching approach for a SDS-capped Pyal nanosuspension for copper ion quantification is of high specificity and coexisting ions were found to interfere very negligibly. The developed method was successfully applied for the estimation of copper ions in river water samples.  相似文献
