Fourteen seasonally anoestrous, non-lactating fallow does were each treated with an intravaginal progesterone device for 14 days followed by a subcutaneously implanted osmotic minipump delivering synthetic GnRH at doses of 125 ng/h (7 does) or 250 ng/h (7 does) for up to 7 days, about 6 weeks before the natural breeding season. One doe (low delivery rate) lost its intravaginal device and 6 of the remaining does (46.2%) exhibited oestrus between 71 and 120 h after progesterone withdrawal/minipump insertion. Only one of these does received the low GnRH delivery rate and 5 received the high rate. Serum progesterone profiles indicated that an induced oestrus was followed by apparently normal luteal development. Does which did not exhibit oestrus failed to show a luteal response. Only 1 doe conceived at induced oestrus, producing a viable female fawn 6 weeks before the start of the natural fawning season. Cellulase production was examined in two strains of Neocallimastix frontalis, namely, PN-1 isolated from the ovine rumen, and PN-2 from the bovine rumen. For both strains, carboxymethylcellulase (CMCase) had a pH optimum of 6.0 and a temperature optimum of 50 degrees C. CMCase resided mainly in the culture fluid, and activities up to 170 U ml-1 (1 U represents 1 microgram of glucose equivalents released per min) were obtained for cultures grown on 2.5 mg of cellulose ml-1. For resting cultures of strain PN-1, the yield of CMCase increased from 9.9 X 10(3) to 10.4 X 10(4) U per g of cellulose degraded, as the initial cellulose concentration decreased from 10 to 0.58 mg ml-1. The range for PN-2 was 8.1 X 10(3) to 11 X 10(4) U g-1. Shaking cultures improved yields for strain PN-1 but not for PN-2. Decreased CMCase production at high initial cellulose concentrations concurred with accumulation of glucose, and addition of glucose (4 mg ml-1) to cultures grown on low cellulose in which none of the sugar accumulated repressed CMCase. Adsorption of CMCase was excluded as a likely explanation for decreased yields at high initial cellulose as only a low proportion (less than 20%) of the enzyme was adsorbed onto the growth substrate. Exoglucanase, measured with alkali-treated Sigmacell or Avicel, gave low levels of activity in the culture fluid (less than 2 U ml-1) and did not appear to be associated with the fungal rhizoid, as treatment with various solubilizing agents failed to give increased activity.(ABSTRACT TRUNCATED AT 250 WORDS) 相似文献

Mechanical perturbation by rubbing of the first internode of 11–12 day old plants of Phaseolus vulgaris L. cv. Cherokee wax induces the rapid deposition of callose in the cells of phloem and other tissues. Callose deposition begins immediately after mechanical perturbation, and shows a minor transient peak 1.5 h, and a major peak 6 h later. The callose gradually disappears and is gone after 3 days. If the stems are perturbed every day, the amount of callose decreases by day 2 but then gradually increases again through day 12. Both the top and bottom of the internode produce callose in response to mechanical perturbation. The evolution of ethylene in response to mechanical perturbation begins after 1 h, peaks at 2–3 h and is gone by 5–6 h. A spray of 10−2 M 2-deoxy-D-glucose (DDG) completely blocks stem thickening, callose deposition and ethylene evolution due to mechanical perturbation. DDG at 10−5 to 10−4 M blocks callose production in mechanically perturbed stem segments and increases ethylene evolution from unperturbed stem segments to greater levels than those obtained by mechanically perturbed segments. It is concluded that mechanical perturbation of bean stems tissue induces deposition of callose more rapidly than it induces evolution of ethylene and that DDG can block both processes. 相似文献

Summary An interdisciplinary approach to breeding for stress tolerance in plants has gained considerable recognition in the past few years. Accordingly, this article presents a synthesis of the genetic, physiological, and ecological aspects of salt tolerance in plants. An understanding of these aspects and the interrelationships between them is essential for an efficient breeding program.A significant part of the presentation concentrates on the basic problems associated with the genetics of tolerance to stresses and of quantitative characters in general, since many of the unsolved problems relevant to the genetics of salt tolerance are still general. Significant progress in the breeding of quantitative as well as qualitative traits in multicellular organisms depends on an understanding of the genetic and epigenetic dimensions of gene action. The discussion therefore includes an overview of (1) the limited existing knowledge on the genetic control of salt tolerance and (2) the physiological mechanisms and molecular targets central to the control of salt resistance as expressed by the amount and stability of yield.An additional subject emphasized here concerns the main strategies of adaptation of wild species to their natural habitats. An understanding of them is essential to (1) enable distinction between traits that can increase agricultural yield and traits that are favorable only for survival under natural conditions (such a distinction is essential, especially when wild species are used as a gene source), and (2) predict the best combinations of characters for efficient agricultural production in stressful environments. 相似文献

Food size selection of four Daphnia, species (D. magna, D. hyalina, D. galeata, D. pulicaria) was investigated using spherical plastic beads as artificial food and with small bacteria. The size of the particles ranged from 0.1 to 35 m with special emphasis to the particle diameters between 0.1 and 1 m. In one set of experiments a mixture of differently sized particles was offered as food suspension and the selectivity of filtering was determined by comparing the size spectrum of the particles found in the gut contents with the spectrum in the food suspension. In a second series of experiments suspensions of uniformly sized particles were offered to single animals and their feeding activity was observed directly. In both types of experiments the mesh sizes of the filtering apparatus of the respective animals studied were measured after the experiments by, scanning electron microscopy. The mean sizes of the filter meshes were about 0.4–0.7 m. In all experiments the size of the particles found in the gut or those which caused high feeding activities were larger than the smallest mesh sizes of the filters. As a consequence simple mechanical sieving provides a sufficient explanation for the mechanism of particle retention of the filtering process in Daphnia. D. magna was found to feed with high efficiency on suspended freshwater bacteria, the residual species investigated showed low filtering efficiencies when bacteria were offered as food.The present study was supported by Deutscher Akademischer Austauschdients 相似文献

Abstract Three proteins from Halobacterium marismortui , malate dehydrogenase (hMDH), glutamate dehydrogenase (hGDH) and ferredoxin (hFD) were purified and characterized with respect to their molecular masses, amino acid composition and, for hFD only, primary structure. Striking features of halophilic proteins are: the high excess of acidic over basic residues; acidic clusters in the sequence. Low-salt concentration causes inactivation and changes in structural parameters of hMDH and hGDH. Reactivation of hMDH involves long-lived stable intermediates. The salt concentration optimum of enzymic activity is independent of salt nature. The high capacity of halophilic proteins to retain water and salt is due to unique molecular properties, studied by physico-chemical techniques. 相似文献

Although mature citrus fruits [ Citrus sinensis (L.) Osbeck cv. Shamouti] did not abscise at the peduncle-shoot abscission zone (AZ–A) when incubated in ethylene environment, abscission processes did occur in a limited number of cell layers situated in the inner bark, the starch sheath region, and in the pith of AZ–A. These processes were regulated by 2,4-D and ethylene treatments. Cells responding to the "separation processes", particularly in the ethylene treatment, underwent either (a) cell wall swelling, dissolving and breakdown, or (b) growth and expansion in a radial plane. Further away from the dissolving area, the response of some cells of the mid and outer bark took the form of divisions or growth in a circumferential plane, while other cells remained unchanged. Non-responding tissues of the outer bark formed a "sleeve" of undissolved cells, and the vascular cylinder produced no abscission in AZ–A. It is concluded that the partial cell wall dissolution in AZ–A explains the increased activity of cellulase and polygalacturonase in the non-abscising AZ–A of the mature fruit (Greenberg et al. 1975. Summary The yeast Cryptococcus neoformans may develop under certain conditions a large polysaccharide capsule 50–100 M in diameter and therefore cannot be phagocytosed by either polymorphonuclear cells (PMN's) or mononuclear phagocytes (MN's). The cellular defense mechanism — in various animals — against the yeast is composed by formation of ringlike structure of PMN's or MN's cells which surround the C. neoformans. Ring structures develop either in vivo or in vitro in tissue culture; destruction of the yeast occurs within 36–72 hours.Several hydrolases, such as acid phosphatase, -glucuronidase and non-specific esterase were found to be released from the phagocytic cells into the enclosed yeast. Considerable reduction of NBT used as a marker for oxidative activity was observed in MN rings at contact regions of the MN cells and the yeast. Electron microscopic studies indicate that the phagocytic cells in the ring structure have many pseudopodes penetrating into the polysaccharide capsule of the yeast. Disintegration of the capsule was observed as well as phagocytosis of its material. A possible analogy between normal phagocytosis of small-sized bodies and the ring structure obtained when large bodies are involved is discussed. 相似文献
